开展异种器官移植临床试验的几个关键伦理问题王 华,吴卫星,栾 尧 (军事医学科学院输血医学研究所,北京 100850)〔摘 要〕目前,异种器官移植由于动物道德权、医疗资源分配及危害大众健康等问题而受到广泛地关注与争论,本文提出了四个关键伦理问题,即利弊界限、供研究患者的知情同意、第三方组织的知情同意及临床试验的监督和批准,作为开展异种器官移植临床试验的先决条件。

【摘要】利用猪的器官来解决当前人源器官严重短缺,为解决移植器官短缺的可行的途径.用定向基因转移(gene targeting)手段,直接并准确地对α-1,3半乳糖苷转移酶(α-1,3GT)基因进行同源重组,使α-1,3GT失活,再结合猪体细胞克隆技术,对其进行人源化改造,减弱或消除排异反应.除对2-1.3GT进行基因定向修饰外,阻断由异种器官移植而激活的人类补体的串联反应是猪异种器官人源化修饰的另一途径.然而,猪内源性逆转录病毒(porcine endogenous retrovirus,PERV)造成的公共卫生问题,给异种器官移植的前景投下了阴影.因此,即要剔除导致人类排异反应的猪细胞表面的α-1,3GT及其相关的分子, 又要确保猪器官异种移植的安全性, 是尚待研究的重大课题.

国际首例6-基因编辑猪-猴肝肾联合移植围术期治疗研究韩士超;马帅军;戚若晨;王国辉;张小燕;刘克普;范效铮;魏迪;张玄;杨晓剑;陶开山;秦卫军;窦科峰【期刊名称】《空军军医大学学报》【年(卷),期】2024(45)1【摘要】目的探讨国际首例6基因编辑猪猴肝肾联合移植模型的构建,探究异种肝脏移植对异种肾脏移植的影响。
方法将人源化6基因编辑猪的肝脏、肾脏移植给恒河猴受体,肝脏移植采用腹腔异位移植,肾脏移植采用原位移植,保留对侧肾脏,观察血流灌注恢复后,移植肝、肾的情况及受体存活情况;定期进行移植物超声及功能检查;监测受体猴生化指标及生理指标变化;受体猴生命终点取移植肾、移植肝进行病理检查。
结果手术历时14 h结束,术后受体猴各项生理指标基本正常,移植物开放血流后可见移植肝灌注良好,即刻可见胆汁流出;移植肾灌注良好,即刻可见尿液流出,未见明显超急性排斥反应发生;术后移植肾功能良好,术后5 d移植肾尿量减少,考虑排斥反应发生,术后受体猴最终存活14 d。
结论多基因编辑猪猴异种肝肾联合移植模型构建成功,术后移植肾未见明显超急性排斥反应发生,异种肝移植可减轻异种肾脏移植急性排斥反应发生。

猪到人肝异种移植的研究英文回答：Pig-to-human liver xenotransplantation is a field of research that explores the possibility of transplanting pig livers into humans. By doing so, the pig organs can be "humanized" to a certain extent, reducing the likelihood of rejection.Another challenge in pig-to-human liver xenotransplantation is the risk of transmission of porcine endogenous retroviruses (PERVs) from the pig organ to the human recipient. Although studies have shown that PERVs are not capable of infecting human cells efficiently, there is still a concern about the possibility of PERV transmission and its long-term effects. 2动物遗传修饰的概念与理解动物遗传修饰（Genetic Modification）是指人为改变动物的基因组（遗传物质），主要包括转基因与基因敲除。
转基因动物（Transgenic Animal ）是指利用实验方法将外源基因导入动物染色体基因组内，外源基因能在动物体内表达并稳定遗传给后代的一类动物[8]。
