中药莲子心中总生物碱的提取及特征图谱研究

摘要

目的:本实验是对莲子心药材中的化学成分进行初步分离,并对所得到的总生物碱成分进行HPLC分析,根据特征图谱来确定莲子心总生物碱的种类以及成分。相信随着科学技术的进一步发展,也会有越来越多的相关研究,能够早日实现其最大的药用价值。

方法:经过前期查阅文献从而确定本次实验方案,本实验将莲子心药材(10.5 kg)粉碎,加12倍80%乙醇,回流提取3次,每次2 h。合并提取液,浓缩至无醇味,浓缩液用1%盐酸调至pH = 3,纱布过滤;滤液加氨水调PH至9,二氯甲烷萃取,浓缩萃取液,得到暗黄色沉淀200 g,即为莲子心总生物碱部分。对莲子心总生物碱部分进行HPLC分析,探索总生物碱的HPLC分析条件,确定了最优的分析条件。在最优的分析条件下,建立了HPLC特征图谱。

结果:结合所得的特征图谱发现莲子心中所含生物碱成分远远多于文献报道的十几个化合物。这表明其中包含有大量结构未知的生物碱类化合物,需要进一步系统研究。

结论:通过此次实验所得到的实验数据,可以为莲子心总生物碱成分的探索和技术开发提供理论基础,为日后研究莲子心总生物碱的药理作用奠定基础。

Abstract

Objective:This experiment is to preliminary separate the chemical composition of Plumula,and HPLC analysis the total alkaloids,which make sure the kind and composition of total alkaloids separate from separate. I believe that with the further development of science and technology , there will be more and more relevant research , as early as possible to achieve its maximum medicinal value.

Method: After a preliminary literature review to determine this experimental program , This experiment crushed the Plumula(10.5 kg),added 12 times 80% ethanol to the powder,refluxed and extracted them three times,each time two hours.And combined the extract,concentrated it until we could’t smell the taste of alcohol, the concentrate was adjusted to pH=3 with 1% hydrochloric acid, then filtered with gauze. The filtrate was adjusted to pH=9 with ammonia, the solvent extraction with dichloromethane,concentrated the extracts, we got 200g dark yellow precipitate., which was the total alkaloids of plumula. After a HPLC analysis to the total alkaloids, explored the analytical conditions of HPLC, made sure the best of the analytical conditions of HPLC.The characteristic spectrum was bulit in the best of the analytical conditions of HPLC.

Results: It was found that the kind of alkaloids in Plumula contained far more than reported in the literature through the result of characteristic spectrum.It indicated that the Plumula contains a large number of unknown structure of alkaloids.

Conclusion:The experimental data obtained in this experiment may provided a theoretical basis for the exploration and technology odevelopment of Plumula total alkaloids, and lay the foundation for future study of the pharmacological effects .
