Tet-On 3G results from the fusion of amino acids 1–207 of a mutant Tet repressor (TetR) to 39 amino acids that form three minimal "F"-type transcriptional activation domains from the herpes simplex virus VP16 protein. Tet-On 3G was derived from Tet-On Advanced (Zhou et al. 2006; Urlinger et al. 2000; Gossen and Bujard 1992; Gossen et al. 1995); as a result, it’s fully synthetic, lacks cryptic splice sites, and is codon-optimized for stable expression in mammalian cells. This promoter consists of a highly optimized Tet-responsive element (TRE) just upstream of a minimal CMV promoter. P TRE3G exhibits exceptionally low basal activity; it’s induced by the binding of Tet-On 3G but is virtually silent in its absence. The vector is designed to be used as part of our Tet-On 3G Inducible Expression System (Cat. No. 631164).ZsGreen1 is a human codon-optimized variant of the reef coral Zoanthus sp. green fluorescent protein (ZsGreen) that has been engineered for brighter fluorescence (excitation and emission maxima: 493 and 505 nm, respectively; Matz et al. 1999; Haas, Park, and Seed 1996). p TRE3G-ZsGreen allows Dox-inducible coexpression of ZsGreen1 and a gene of interest from a bicistronic mRNA transcript. An encephalomyocarditis virus (EMCV) internal ribosome entry site (IRES2), positioned between ZsGreen1 and the gene of interest, facilitates cap-independent translation of the gene of interest from an internal start site at the IRES2/MCS junction (Jang et al. 1988). This ensures that a high percentage of ZsGreen1-expressing clones also express the gene of interest, allowing ZsGreen1 to be used as an indicator of inducibility and transfection efficiency, as well as a marker for selection by flow cytometry. The vector also contains a pUC origin of replication and an ampicillin resistance gene (Amp r) to allow for propagation and selection in E. coli.The pTRE3G-Luc is a Tet-inducible control vector that expresses firefly luciferase under the control of P TRE3G. 注意:当轴承温度在60℃以内时,雾化器运行良好;当轴承温度在80℃以内时,雾化器可以正常运行; 当轴承温度在80-100℃时,应准备停止雾化器; 当轴承温度超过100℃时,必须立即停止雾化器运行。
2) 振动传感器雾化器安装有振动传感器,用于测量由于轴承磨损、雾化器主轴松动及料液旋转喷头不平衡等引起的振动。

[贮藏]1、未开封时,冷藏于≤8℃(≤46℉),并在保存期内用完,高温度时,凝固水可以排除,包装物最好于室温启开。
2、已开封的,将封口以胶带封紧。
3、保存再封的袋于≤25℃(≤77℉)和温度<50%,不要冷藏已开启的包装袋,并于一个月内使用完。
[样品制备]4、制备1:10和更大稀释的食物样品稀释液,0称取或吸取食物样品,置入适宜的无菌容器内,如均质袋、稀释瓶、WhirlPak bag 或者其他灭菌容器内.5、加入适量的无菌稀释液,包括Buffered peptonephopsphate buffer ,用0.0425g/L的KH2PO4调PH7.2) 、0.1%的蛋白胶水(ISO方法6887) 、缓冲蛋白胶水(ISO方法6579) 、盐溶液(0.85-0.90%)、bisulfite-free letheenbroth或蒸馏水.6、搅拌或均质样品。
[接种]7、将测试片置于平坦表面处,揭开上层膜。
8、使用吸管将1mL样液垂直滴加在测试片的中央处。
9、允许使用上层膜直接落下,切勿向下滚动上层膜。
10、使用压板隆起面底朝下,放置在上层膜中央处。
11、轻轻的压下,使样液均匀覆盖于圆形的培养面积上,切勿扭转压板。
12、拿起压板,静置至少1分钟以使培养基凝固。
[培养][解释]13、测试片的透明面朝上,可堆叠至20片,对有一定湿度养箱能保持最少份损失是需要的。
14、可目视及用标准菌落计数器或其它的照明放大镜计数,并可参考判读卡计算菌落数。
15、可以分离菌落作进一步鉴定,即掀起上层膜,由培养胶上判读卡Aerobic bacteria count=152测试片中含有一种红色指示染剂可使菌落着色,计算所有红色菌落(不论其大小和颜色深浅均计算之).Count=0在petrifilm AC测试片上,很容易解释,图2测试片上没有任何菌落生长.Count=16图3示有不多的菌落.Count=143Petrifilm AC测试片菌落数适宜计数范围是25-250,见图4Count=560测试片面积约为20cm2,当菌落数超过250个,如图5所示,为了估计菌落数,可选择其中一个或数个有代表性菌落的小方格(1cm2),计算平均菌落数,再乘以20可得到整个测试片上的菌落数.Count=tntc(estimated count=103)图6所示petrifilm AC测试片上菌落太多而无法计数(tntc)count=tntc(Estimated count=105)有很多高数量菌落,使整个生长区变粉色,如图7所示,你仅可在生长区边缘现观察到单个菌落,应计录为菌落太多无法计数(TNTC)count=tntc(Estimated count=103)有时菌落分布出现不均衡,如图8所示,这也记录为TNTC.Count=tntc(Estimated count=107)在图9中petrifilm AC测试片上的菌落,最先粗略一看是可计数的,然而,你密切注意到生长区边缘,能看到高密度的菌落,应记录为TNTC Estimated count=160很少数菌种会液化petrifilm AC测试片上的培养基,如图10所示.若有这种现象发生,可选择没有液化区的几个有代表性菌落的小方格(1cm2),计算平均菌落数.不要计数液化区内的红点.Count=83因为在petrifilm AC测试片上菌落是红色的,你会和不透明的不规则形状的事物颗粒区分开,见圆圈1和2.霉菌和酵母菌检验测试片Petrifilm and Mold count Mold TM测试片为预先制备好的培养基系统,它含有一种冷水可溶性的凝胶剂,营养基和一种指示染剂,可提供对比,利于菌落计数。

4. 试验方法4.1 谐振频率及谐振阻抗的测试:测试夹具短路时调节信号发生器输出电压,使毫伏表指示值为50mV,接上雾化片,调节信号发生器的频率,使毫伏表指示最大,这时的频率为谐振频率;谐振阻抗按R=19.3(50/E0-1)(Ω)计算。
4.2 静电容量的测试:在频率为1KHz,电压小于1V的信号下测试。
4.3 寿命试验:将产品装上雾化器,将激励电压调至24V,使雾化器正常工作并计时或采用美国军方测试压电陶瓷寿命的计算公式来测试。
5.注意事项a. 雾化片的浸水面为零电位,电极与液体不能有电位差。
b. A型不能在无水状态下使用。

进行线粒体标记时,只需将细胞与MitoBeacon™Red简单孵育,探针就可以通过被动扩散的形式透过细胞膜,并在有活性的线粒体内富集。
一旦线粒体被染料标记,再用醛基类固定剂固定细胞,样品就可以用于下一步处理。
因为线粒体可以在细胞被固定之前进行染色,所以可以借助MitoBeacon™Red解决一些致病性细胞研究中面临的困难。
虽然四甲基罗丹明和罗丹明123等传统的荧光染料可以很容易识别出功能性的线粒体,但是一旦发生膜电位损失,这些染料很容易被冲洗出线粒体。
对于需要使用醛类固定剂或其它对线粒体活性有影响的试剂的实验操作,四甲基罗丹明和罗丹明123等传统的荧光染料的使用明显受到限制,MitoBeacon™Red可以帮助用户轻易解决这个问题。

Please note that the rear unit and each filter stage have four mushroom shaped pins.2. Also please note that the front unit and each filter stage has four key hole shaped slots.3. Make sure to feel a clear “click”.Make sure all the engravings are at the right top side.Engravings4. 八、注意事项8.1、必须避免的情况8.1.1 暴露于可挥发性硅化合物蒸气中:传感器要避免暴露于硅粘接剂、发胶、硅橡胶、腻子或其它存在可挥发性硅化合物的场所。
如果传感器的表面吸附了硅化合物蒸气,传感器的敏感材料会被硅化合物分解形成的二氧化硅包裹,抑制传感器的敏感性,并且不可恢复。
8.1.2 高腐蚀性的环境:传感器暴露在高浓度的腐蚀性气体(如H2S,SOX,Cl2,HCl 等)中,不仅会引起加热材料及传感器引线的腐蚀或破坏,并会引起敏感材料性能发生不可逆的劣变。

These serve as a qualitative control to determine the "noise" level of individual flow cytometers. Certification verifies the detection of the blank beads in fluorescence channels below that of unstained human leukocytes. The product is offered in buffered saline with stabilizer and 0.1% sodium azide (see MSDS)* as preservative. DescriptionStandardization and reproducibility of instrument performance is critical in flow cytometry.1-8 There are a variety of means for monitoring instrument parameters. It is necessary to define fluorescein threshold in order to ensure that a given flow cytometer has the necessary sensitivity for immunophenotyping of cell surface antigens expressed at low density. Instruments should be maintained at a sensitivity level greater than that detected for autofluorescent cells. Fluorescence threshold is defined as the level above which a fluorescence signal can be measured by an instrument. Mean channel may be alternatively used, however, note that mean and median channels are not inter-changeable.Run the lysed, unstained whole blood sample on the flow cytometer, set a gate for lymphocytes and collect 10,000 events.1.Record the mean or median channel at eachPMT.pare the mean or median channels of theunstained human lymphocytes to that of the Certified Blank. Each lot is accompanied by a lot-specific certificate verifying the expected mean channel values for a properly calibrated instrument. The Certified Blank microspheres are maintained at physiological pH (7.2). Vigorously mixing microspheres prior to use is essential in obtaining a uniform suspension.
