Tecnai 1 & 2

Specifications and Capabilities

Point Resolution (TEM): 0.5 nm (0.35 nm)

Point Resolution (STEM): 0.35 nm (N/A)

Line Resolution (TEM): 0.35 nm (0.2 nm)

Elemental Analysis: EDS (N/A))

Energy-Filtered TEM: N/A (N/A)

Image Recording: 1 CCD Camera (1 CCD Camera)

Emergencies

Power Failure

In the case of a power failure, the microscope will shut down safely. When power is restored, it is necessary to restart the microscope manually (see the CoreLab Staff to perform this task). Cooling Water Disruption

As we know that the power to high voltage supply as well as electron lenses will stop as soon as chilled water is stopped. Upon resumption of the chilled water, the microscope must be brought back to the working condition (see the CoreLab Staff to perform this task). Emergency Stop

Use the big red button on a power supply (back of the curtains) in case of fire or severe flood. Avoid this method of shutdown unless absolutely necessary.

0.Planning for having a TEM session on Tecnai

∙Make sure that you have all the work-related training certificates from respective departments of KAUST

∙Make sure, you have taken the Advanced TEM training course by the CoreLab Instructor ∙Make sure, you make the booking on Badger according to directions given to you

∙Make sure that the TEM Instructor has authorized to use the Titan-CT microscope

∙Enable your session on the Badger according to the direction given to you

1.Starting the TEM session on Tecnai

∙Login to computer by using the “supervisor” for User and “tecnaid1545 ” for password on Tecnai1 and “tecnaid3375” on Tecnai2

∙Wait for few moments till all processes are loaded

∙Launch Tecnai-User-Interface (TUI) by its Icon on the Start-bar and wait till its launched completely

∙Launch Digital Micrograph (DM) next (only on Tecani2)

∙Launch TEM Imaging and Analysis (TIA) application

∙Check the status of vacuum on the Setup-page of TUI and notice if the vacuum are in the following range (log-scale);

o Gun/column----6-12

o Projection-chamber------19-45

o Backing-line----should be green

o Buffer-tank-----should be green

∙Cool down the “cold-finger” with liquid-nitrogen (LN2) and wait for ~20-30 minutes

2.Loading the sample into Tecnai Microscope

∙Load the sample on Single or Double Tilt holder as per the directions given to you during the training session.

∙Make sure, the pumping time, given on the Setup page of TUI, for the load-lock is at least 100 seconds

∙Load the holder into the CompuStage of the scope and wait till the pumping cycle is over

∙Load the holder as per the directions given to you during the training sessions

∙Notice the vacuum particularly for Column and Gun

∙Wait for another 5-10 minutes till the vacuum in the octagon region comes in to the range of 15-20 log scale

3.Making the Tecnai ready for Imaging the samples

∙Apply the HT in steps till 120 kV

∙Saturate the filament and observe the stability of emission current

∙Open the column valve of the scope given on the setup page of TUI
