丹参素对糖皮质激素诱导骨丢失大鼠胫骨近端骨密度和骨微结构的影响陈景锋;罗世英;崔燎【摘要】Aim To investigate the effect of tanshinol on bone mineral density and microstructure of proximal tibias in rats with bone loss induced by glucocorticoid. Methods Sixty 7-month-old female SPF SD rats were randomly divided into 6 groups with 1 0 rats per group:controlgroup(saline:5 ml·kg -1 ·d -1 ),glucocorti-coid group (prednisone acetate:6 mg·kg -1 ·d -1 ), glucocorticoid +low dose of tanshinol group(1 2.5 mg ·kg -1 ·d -1 ),glucocorticoid +medium dose of tan-shinol group (25 mg·kg -1 ·d -1 ),glucocorticoid +high dose of tanshinol group (50 mg·kg -1 ·d -1 ), glucocorticoid +(positive control drug)calcitriol group (0.045 μg · kg -1 · d -1 ).Rats were gavaged with prednisone acetate continuously for 1 4 weeks to estab-lish the bone loss model.Meanwhile,tanshinol and calcitriol were orally administered to the rats which were treated with prednisone acetate for intervention. At the end of the experiment,the left proximal tibias were collected for Micro-CT scanning and three-dimen-sional reconstruction of cortical and trabecular bone re- <br> spectively to observe the changes of bone microstruc-ture and test related parameters.Results Bone min-eral density was decreased and bone microstructure was destroyed in proximal tibias of rats after treatment with glucocorticoid.Both tanshinol (25 mg·kg -1 ·d -1 ) and calcitriol(0.045 μg·kg -1 ·d -1 )could increase bone mineral density and improve bone microstructure in proximal tibias withoutsignificant differences be-twee n each other.Tanshinol (50 mg · kg -1 · d -1 ) could improve bone microstructure to some extent,but it had no significant effect on bone mineral density. Tanshinol(1 2.5 mg·kg -1 ·d -1 )had no significant effect on bone mineral density or microstructure.Con-clusion Oral administration of tanshinol (25 mg · kg -1 ·d -1 )to the rats treated with glucocorticoid can increase bone mineral density and improve bone micro-structure in proximal tibias.%目的:探讨丹参素对糖皮质激素诱导骨丢失大鼠胫骨近端骨密度和骨微结构的影响。

Methods: Thirty female SD rats were randomly divided into three groups with 10 rats in each: blank control group, model control group and Danshensu group. The osteoporosis model was induced by bilateral ovariectomy and rats in Danshensu group were fed with Danshensu 12 . 5 mg · kg-1 · d-1 by gavageafter ostroporosis model induced. All animals were sacrificed after 90 days. The bone mineral density ( BMD) of the whole body, femur and lumbar vertebra was measured by dual energy X-ray absorptiometry. The biomechanical properties of femur were measured by AG-IS mechanical universal testing machine. Serum osteocalcin and bone alkaline phosphates ( BALP ) levels were measured by ELISA. The number of osteoblasts of proximal femoral metaphysis was counted with light microscopy after HE staining. Results: Compared with blank control group, BMD, biomechanical properties of femur, serum osteocalcin and BALP levels and the number of osteoblasts were decreased in model control group ( P<0 . 05 or P<0 . 01 ) . While compared with model control group, BMDs of the whole body, femur and lumbar vertebra, the elastic modulus,maximum load, yield strength, breaking point load of femur, the serum levels of osteocalcin and BALP, and the number of osteoblasts were significantly improved in Danshensu group ( P<0 . 05 or P<0 . 01 ) . Conclusion: Danshensu can improve bone quality by increasing bone density, improving biomechanical properties, promoting the expression of osteogenesis-related factors, and increasing the number of osteoblasts.%研究口服丹参素能否促进去势大鼠骨形成或减少骨丢失,从而提高骨质量。
丹参多酚酸盐对大鼠骨髓间充质干细胞增殖及血管内皮生长因子表达的影响王晓娟【摘要】目的观察中药丹参多酚酸盐对SD大鼠骨髓间充质干细胞的增殖及血管内皮生长因子(VEGF)表达的影响,以探讨其对促骨折愈合的可能机制.方法将0.5 mg/ml浓度丹参多酚酸盐与大鼠骨髓间充质干细胞共培养,对照组为无药物单位培养的大鼠骨髓间充质干细胞.培养48小时镜下观察细胞情况,1、3、5、7天行MTT检测细胞增殖情况.行实时荧光定量PCR检测方法,对培养7天的两组细胞的VEGF mRNA水平进行检测分析.结果共培养48小时后骨髓间充质干细胞生长密集程度明显优于对照组;MTT检测也显示实验组细胞增殖情况优于对照组;实验组VEGF mRNA表达高于对照组.结论丹参多酚酸盐能促大鼠骨髓间充质干细胞增殖,并提高其VEGF的表达.可能是其促骨折愈合的作用机制之一.【期刊名称】《环球中医药》【年(卷),期】2013(006)007【总页数】4页(P492-495)【关键词】丹参多酚酸盐;骨髓间充质干细胞;增殖;血管内皮生长因子【作者】王晓娟【作者单位】830002,乌鲁木齐,解放军第23医院药械科【正文语种】中文【中图分类】R285.6丹参多酚酸盐(salvianolate)为丹参的水提物,其主要成分为丹参乙酸镁(magnesium lithospermate B)。
文献报道[1-2]丹参多酚酸盐对缺血性疾病有明显的疗效,对人血管内皮细胞有促迁移及增殖的作用。
骨折的愈合一直是学者们长期研究的难题[3]。
骨折愈合过程中,骨折端新生血管的长入是其重要的步骤之一[4-5],干细胞的归巢是骨折修复的重要来源[6-7]。
如果提高干细胞的促新生血管形成能力,将大大提高骨折的愈合能力。
本研究采用细胞共培养的方法,观察丹参多酚酸盐对骨髓间充质干细胞的增殖及与血管形成密切相关的血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达情况,初步探讨中药对促骨折愈合的作用机制。
丹参素的药理研究进展杨春欣(上海医科大学附属中山医院药剂科,上海 200032)中国图书分类号 R282.71;R284.1;R972 丹参为唇形科鼠尾草属植物(Salvia miltior-rhiza Bunge)的干燥根部,是常用的活血化瘀中药,其水溶性提取物丹参注射液已广泛应用于临床治疗心脑血管疾病达20余年。
从丹参水溶性部位分离到的有效成分——丹参素(danshensu),即D(+)B-(3,4-二羟基苯基)乳酸〔1〕,我国学者已进行了许多药理研究,证明其有多种药理活性。
1 对心肌的作用1.1 具有缩小心肌梗塞范围和减轻病程的作用 观察结扎兔心左前降支1/2处所造成的小范围心肌梗塞,丹参素治疗组梗塞范围为4.31%±1.04%,而生理盐水对照组为11.95%±1.92%,两组间有明显差异;对于结扎兔心左前支上1/3处所造成的较大范围的心肌梗塞,对照组梗塞范围为17.62%±1.45%,而丹参素治疗组11.55%±1.65%,明显小于对照组,提示丹参素具有明显缩小心肌梗塞范围和减轻病变程度的作用〔2〕。

相反原儿茶醛则更明显地降低左心室收缩锋压和升高左心室舒张末压。
24h后心梗范围缩小,以丹参素最为明显,各组狗心肌缺血重量,对照组为13.46±1.4g,丹参素组为2.67±0.36g, P<0.01〔3〕。
1.2 具有对大鼠心肌缺血/再灌注损伤的保护作用 采用Langendorff灌注模型,对心肌缺血/再灌注损伤中氧自由基清除酶系统超氧化歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-P X)和超微结构的变化及丹参素的抗氧化效应进行了观察,证实心肌组织缺血时,SOD和GSH-16的活性逐渐降低,预先给丹参素再进行缺血/再灌注,心肌SOD、GSH-P X的活性明显高于单纯缺血/再灌注时,超微结构损伤也较轻,丹参素的保护作用效果明显优于公认的具有抗氧化作用的亚硒酸钠,但对GSH-P X的活性影响与亚硒酸钠比较无明显差异〔4〕。
丹参注射液对大鼠下颌骨骨折愈合过程中BMP2 和TGFBi的影响向乐余伟[Summary]目的探讨丹参注射液对大鼠下颌骨骨折愈合的影响,并通过测量骨形成蛋白-2 (BMP-2)、转化生长因子-Bl (TGF-Bl)水平变化,探讨丹参促进下颌骨骨折愈合的械制。
方法选取SD大鼠48只,通过双侧下颌骨体部造成颊舌侧贯穿不完全性骨折模型,按随机数字表法分为实验组和对照组,每组24只。
实验组每天腹腔注射丹参注射液(1 mL/kg),对照组每天腹腔注射生理盐水。
术后第1、2、3、4周,每组分别随机处死6只大鼠,采用酶联免疫吸附试验(ELISA)检测血清BMP-2、TGF-Bl的含量。
取出双侧下颌骨,右侧下颌骨行X线片检测骨折愈合情况并测量骨痂区的平均骨密度,标本经组织脱钙、石蜡包埋行HE染色后,采用骨组织形态计量学方法分析骨痂区骨小梁面积比、骨小梁宽度;左侧下颌骨骨痂,行组织粉碎,采用RT-qPCR技术检测BMP- 2、TGF-Bl的表达情况。
结果X线结果显示对照组骨折愈合明显延迟于实验组;从第2周开始,与对照组相比,实验组骨痂骨密度明显增加(P < 0.05或P < 0.01),骨痂区骨小梁面积比、骨小梁宽度显著性增大(P < 0.05或P < 0.01);差异随着愈合时间的延长而显著。
ELISA结果显示,术后实验组血清BMP-2、TGF-B 1的浓度均高于对照组(P < 0. 05)。
RT-qPCR结果显示,术后第2周开始,实验组骨痂中BMP-2及TGF-BlmRNA的表达均高于对照组(P < 0.01) o结论丹参注射液促进下颌骨骨折愈合,可能与促进骨折愈合过程中BMP-2、TGF-B1基因的转录及蛋白表达有关。

Methods Forty-eight SD rats were operated forincomplete fracture of hibateral mandible penetrated from buccal sideto lingual side. The rats were divided into experimental group and control group according to random number table, with 24 rats in each group. The experiment group was administered with Danshen Injectionby intraperitoneal injection, and the control group was administeredwith physiologic saline by intraperitoneal injection. The right healing of fracture were used to detect fracture healing by X-ray and to measure the average bone density of the callus area, the percent trabecular and trabecular width were analyzed by bonehi stomorphometry and the serum levels of BMP-2, TGF-P 1 was determined by ELISA at 1, 2, 3, 4 weeks after the operation. Theright healing of fracture were comminuted, and the expression of BMP-2 and TGF-P 1 were analyzed by RT-qPCR. Results The X-ray results showed that the fracture healing in the control group was obviously delayed in the experimental group. After 2 weeks, compared with the control group, the callus bone mineral density (BMD) in the experimental group was increased (P < 0. 05 or P < 0, 01) . The trabecular area ratio and trabecular width in the callus area were markedly increased (P < 0. 05 or P < 0. 01) , and the difference was even more increased along with the increasing time. ELISA results showed the serum levels of BMP-2 and TGF-P 1 in the experimental group were higher than those in the control group (P < 0, 05) . RT- qPCR results showed the expression of BMP-2 and TGF-0 1 in the experimental group were higher than those in the control group from the 2nd week after operation (P < 0. 01) . Conclusion DanshenInjection can promote the healing of mandibular fracture, which may be related to the transcription and protein expression of BMP-2 and TGF-P 1 gene during the process of fracture healing.[Key words] Danshen Injection; Mandibular fracture; Bone morphogenetic protein-2; Transforming growth factor-3 1下颌骨位于颜面下部,是颌面部体积最大且位置较为突出的骨骼。
