comatus)中一种抗病毒蛋白y3特性和氨基酸序列分析-中国生物化学与分子生物学报2008,24(7)食用茵中含有多种抗病毒蛋白,可用于植物保护,利用离子交换层析技术和凝胶层析技术,从食用茵毛头鬼伞中提取到抗植物病毒蛋白y3,实验结果表明,y3是一种糖蛋白,利用Western杂交方法可以在发酵茵丝体和子实体中同时检测到,说明可能是组成型表达,根据其N端氨基酸序列,使用RACE-PCR克隆技术,获得了蛋白的氨基酸序列和部分cDNA序列,浓度为2.0 μg/ml时,蛋白y3对烟草花叶病毒(TMV,20 μg/m1)侵染心叶烟的抑制率为50%,实验同时表明,y3还可抑制病毒在寄主普通烟Nicotirma tabacum Var.k326中的复制.2.学位论文付鸣佳食用菌抗病毒蛋白的特性、基因克隆与表达2002采用离子交换柱DEAE-SepharoseFF和凝胶层析柱Superdex75 Prep Grade和SephacrylS-200High Resolution,可从杏鲍菇(Pleurotus eryngii)干样中分离得到多个蛋白组分,结枯斑寄主检测,发现多个蛋白组分都有抗烟草花叶病毒(Tobacco mosaic virus,TMV)的活性,对TMV的抑制率均在70﹪以上,高者可达99﹪.对Zb蛋白质生化特性进行了研究,研究表明该蛋白是分子量为30kDa的单亚基蛋白,没有结合糖,其等电点为4.7.测定了它的N-端20个氨基酸序列为:PQVKTSWEDLANLGWPIQQV.将Zb基因导入烟草中,检测表明Zb基因已经转入烟草植株中.目前发现有一株转基因烟有异常现象,其它均生长正常.经供毒试验表明,发现一株生长矮小的植株有一定抗性.3.期刊论文付鸣佳.吴祖建.林奇英.谢联辉榆黄蘑中一种抗病毒蛋白的纯化及其抗TMV和HBV的活性-中国病毒学2002,17(4)采用阴离子交换层析和凝胶层析方法从新鲜食用菌榆黄蘑(Plearotus citrinopileatus)中进行了抗病毒蛋白的纯化,结果获得了一个纯化蛋白YP46-46,经SDS-PAGE可确定其分子量为27.4kD.以半叶法在枯斑寄主心叶烟上检测该蛋白对烟草花叶病毒(TMV)的抑制率,发现有较好的抗TMV活性,其抑制TMV的中浓度为0.24μg/mL.同时以HepG2.2.2.15细胞株为模型,对所获得的蛋白进行体外抗乙型肝炎病毒效果的评价.结果表明该蛋白对HBsAg的50%抑制时的浓度为0.08μg/mL,但对HBeAg效果不大.4.期刊论文吴丽萍.曹郁生.吴祖建.林奇英.谢联辉.WU YP3:食用菌榆黄蘑中新的植物病毒抑制物蛋白-天然产物研究与开发2009,21(3)食用菌中含有多种抗病毒蛋白,可用于植物保护.采用硫酸铵分级沉淀、DEAE-纤维素离子交换层析和SephacrylTMS-200凝胶层析方法,从食用菌榆黄蘑新鲜子实体中提取到一单亚基蛋白,命名为YP3.利用SDS-聚丙烯酰胺不连续凝胶电泳初步估计其分子量大约为27.6 kDa.氨基酸组成分析表明该蛋白非常类似其它的抗植物病毒蛋白,并且几乎不含糖.其N-末端序列为NRDVAACARFIDDFCDTLTP,在GenBank中没有找到同源序列.浓度为0.24 mg/L时蛋白YF3对烟草花叶病毒(TMV 20 mg/L)侵染心叶烟的抑制率为50%.同时还发现YP3对供试的细菌和真菌没有抑制活性,对胃癌细胞株MGC-803、肝癌细胞株SMMC-7721、肺癌细胞株SPC-A1的细胞增殖具有一定的抑制作用,其IC50大约为20 mg/L.5.学位论文吴丽萍两种食用菌活性蛋白的分离纯化及其抗病特性2004植物病毒病害造成重大的经济损失.解决作物上植物病毒的危害,一直是许多研究者的目标.从生物体中提取抗植物病毒物质是防治植物病毒病害的方法之一.该实验从食用菌中获得了两种抗植物病毒蛋白,其结果如下:用离子交换层析(CM-Sepharose FF)和凝胶层析(Superdex<'TM> 75)方法,从新鲜食用菌毛头鬼伞(Coprinus comatus)子实体中分离纯化出一碱性蛋白y3,经SDS-PAGE及PAGE确定为单亚基组成,MALDI-TOF质谱确定其分子量为12.3kDa.以碘酸-Schiff染色法和酚-硫酸法测定蛋白含糖量高达30%.对其氨基酸组成进行分析后表明含有大量的精氨酸和天冬酰胺.y3蛋白活性检测结果显示:当其浓度为12.5μg.mL<'-1>时,在心叶烟枯斑寄主上对烟草花叶病毒(Tobacco Mosaic Virus,TMV)的侵染抑制率达83.0%;y3蛋白对兔血凝集活性滴度为2<'-5>,对人血凝集活性滴度为2<'-6>,其浓度分别为1.562μg.mL<'-1>和0.781μg.mL<'-1>,凝集活性能被半乳糖、γ-球蛋白、麦芽糖抑制;对三种细胞即胃癌细胞株MGC80-3、肝癌细胞SMMC-7721,及肺癌细胞SPC-A<,1>体外抗性测定结果表明,y3蛋白对MGC80-3和SPC-A1的抑制中浓度约为12.5μg.mL<'-1>,对SMMC-7721的抑制中浓度则较高,约30μg.mL<'-1>,形态观察表明蛋白对SPC-A<,1>具有类似诱导凋亡作用;对供试的植物病原真菌和细菌均无抑制作用.采用硫酸铵分级沉淀、阴离子交换层析(DEAE-SepharoseFF)和凝胶层析(Sephacry1<'TM>S-200High Resolution)方法,从新鲜食用菌榆黄蘑(Plearotus citrinopileatus)中进行了抗病毒蛋白的纯化.以半叶法在枯斑寄主心叶烟上检测YP46-46蛋白对TMV的抑制率,发现有较好的抗TMV活性,其抑制TMV的中浓度C<,50>为0.240μg.mL<'-1>.YP46-46蛋白对兔和人血凝集活性滴度为2<'-7>,其浓度分别为0.219μg.mL<'-1>.对y3蛋白的抗TMV机制作了初步分析.提取毛头鬼伞中的总RNA,采用RACE方法和巢式-PCR技术,根据y3蛋白N-端20个氨基酸序列设计引物进行了基因克隆.获得了y3的部分cDNA序列以及3'端非编码区.其序列长度为414bp,其中ORF长度包括终止密码共291bp,编码蛋白序列共96个氨基酸,3'非编码区含有123个碱基,其中polyA为22个.经NCBI中Blast比较没找到同源序列,表明该基因为一全新序列(GenBank接收号:602564).加上N-端未用来设计引物的5个碱基,全长共101个氨基酸.等电点为8.17.该蛋白序列中有7个半胱氨酸,可形成分子内二硫键.蛋白在Swiss-Prot登录号P83811(登录时采用名称为CAP).本文链接:/Periodical_syjxb200902016.aspx授权使用:上海海洋大学图书馆(shhydxtsg),授权号:4e23fd8b-e104-44ee-a321-9e1a014af829下载时间:2010年10月25日。
