Abstract

Recentgenome-wideassociation(GWA)studieshaveidentifieddozensofcommonvariantsassociatedwithadultheight.

However,itisunknownhowthesevariantsinfluenceheightgrowthduringchildhood.Wederivedpeakheightvelocityin

infancy(PHV1)andpuberty(PHV2)andtimingofpubertalheightgrowthspurtfromparametricgrowthcurvesfittedto

longitudinalheightgrowthdatatotesttheirassociationwithknownheightvariants.ThestudyconsistedofN=3,538

singletonsfromtheprospectiveNorthernFinlandBirthCohort1966withgenotypedataandfrequentheightmeasurements

(onaverage20measurementsperperson)from0–20years.Twenty-sixofthe48variantstestedassociatedwithadult

height(p,0.05,adjustedforsexandprincipalcomponents)inthissample,allinthesamedirectionasinpreviousGWA

scans.SevenSNPsinornearthegenesHHIP,DLEU7,UQCC,SF3B4/SV2A,LCORL,andHIST1H1DassociatedwithPHV1andfive

SNPsinornearSOCS2,SF3B4/SV2A,C17orf67,CABLES1,andDOT1LwithPHV2(p,0.05).Weformallytestedvariantsfor

interactionwithage(infancyversuspuberty)andfoundbiologicallymeaningfulevidenceforanage-dependenteffectfor

theSNPinSOCS2(p=0.0030)andfortheSNPinHHIP(p=0.045).Wedidnothavesimilarpriorevidencefortheassociation

betweenheightvariantsandtimingofpubertalheightgrowthspurtaswehadforPHVs,andnoneoftheassociationswere

statisticallysignificantaftercorrectionformultipletesting.Thefactthatinthissample,lessthanhalfofthevariants

associatedwithadultheighthadameasurableeffectonPHV1orPHV2islikelytoreflectlimitedpowertodetectthese

associationsinthisdataset.Ourstudyisthefirstgeneticassociationanalysisonlongitudinalheightgrowthinaprospective

cohortfrombirthtoadulthoodandgivesgroundingforfutureresearchonthegeneticregulationofhumanheightduring

differentperiodsofgrowth.
