抗人转铁蛋白受体单克隆抗体增强姜黄素对雄激素非依赖性前列腺癌的抑制效应刘双林;王志华;胡志全;曾星;李有元;李恒;章传华;叶章群【摘要】Objective To study the anti-tumor activity of anti-human transferrin receptor monoclonal antibodyCTfR mAb) in androgen-independent prostate cancer in vitro and investigate the effect of TfR mAb on the anti-tumor activity of curcu-min. Methods After curcumin treatment, transferrin receptor expression was detected by using flowcytometry(FCM)in androgen-independent prostate cancer cells. The viability of prostate cancer cells after TfR mAb treatment was determined by MTT assay. After treatment with TfR mAb and curcumin, the proliferation of prostate cancer cells was assessed by CFSK dilution. FCM was used to analyze the cell cycle and the induction of apoptosis in tumor cells. Results Curcumin could increase the transferrin receptor expression of androgen-independent prostate cancer cells significantly(P<CO. 05). Anti-human TfR mAb a-lone had no significant effect on the proliferation, apoptosis and cell cycle of androgen-independent prostate cancer cells(P> 0. 05). But anti-human TfR mAb could enhance the inhibitory effect of curcumin on the apoptosis of PC-3 cells significantlyCP <C0. 05). Conclusion Curcumin treatment could increase the transferrin receptor expression of androgen-independent prostate cancer cells. Anti-human TfR mAb alone had no obvious anti-tumor activity in androgen-independent prostate cancer cells. Study of the molecular mechanisms of Taiwanese propolis derivatives on the induction of apoptosis in human hepatoma cells




Activating transcription factor 3 (ATF3) is a member of the ATF/CREB subfamily of the basic-region leucine zipper (bZIP) family, which includes members of CCAAT/enhancerbinding protein (C/EBP) and AP-1. ATF3 is rapidly up-regulated under various stress conditions and plays an important role in induction of cancer cell apoptosis. NBM-TP-007GS-002 (GS-002) and PPG are Taiwanese propolis derivatives. Recent reporters indicated that propolis had various biological activities, including anti-microbial, antioxidative, anti-inflammatory and anti-tumoral activity. Our previous examination have found that both GS-002 and PPG significantly inhibited tumor growth in nude mice (Balb/cAnN-Foxn1nu/CrlNarl) which bearing xenografts of C6 glioma cells. In this study, we have examined the anti-tumoral effect of GS-002 and PPG in human hepatoma cells Hep3B and HepG2 in vitro. First we found that GS-002 and PPG significantly inhibited cell proliferation and induced cell apoptosis by a dose-dependent manner. Several main apoptotic indicators were found in GS-002- and PPG-treated cells, such as cleavage form of caspase-3、caspase-9 and PARP. Both GS-002 and PPG also dose-dependently induced the mRNA and protein expression of ATF3. The induction of ATF3 expression is mediated through mitogenactivated protein kinases (MAPKs) signalling pathways in GS-002-treated cells. Furthermore we found that GS-002 induced more cell apoptosis in ATF3 overexpression cells than control cells. These results suggest that the induction of apoptosis by propolis derivatives GS-002 is partially mediated through ATF3-dependent pathway, and the GS-002 has a potential to develop as a anti-tumoral drug.

It has made good progress in the research and application of anti-fibrosis of internal organs, but the effect and mechanism for hypertrophic scars and skin fibroblasts are unclear. OBJECTIVE:To investigate the effect of pirfenidone on human hypertrophic scar fibroblasts. METHODS:Human hypertrophic scar fibroblasts were cultured using tissue culture method. Passages 3-6 cel s grew wel in the logarithmic growth phase were col ected. Cel s were divided into the control group (0 g/L pirfenidone), 0.15, 0.3 and 1 g/L pirfenidone groups according to different mass concentrations. Cel s were intervened for 12, 36 and 48 hours. RESULTS AND CONCLUSION:MTT, reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay results demonstrated that compared with the control group, cel proliferation, transforming growth factorβ1 mRNA expression, types I and III col agen secretion were decreased in the 0.15, 0.3 and 1 g/L pirfenidone groups (P<0.05), and the decrease was most significant in the 1 g/Lpirfenidone group (P<0.05). At 24, 48 and 72 hours after intervention, significant differences in inhibitory rate of cel proliferation and the secretion of types I and III col agen were detected among 0.15, 0.3 and 1g/L pirfenidone groups (P<0.05).&nbsp;Results confirmed that pirfenidone apparently inhibited the secretion of col agen of hypertrophic scar fibroblasts cultured in vitro, transforming growth factorβ1 expression and cel proliferation and viability.%背景：有研究表明细胞因子抑制剂吡非尼酮通过调控多种细胞因子，抑制成纤维细胞的生物学活性，其在内脏器官的抗纤维化作用的研究和应用取得了良好的进展，但对于皮肤增生性瘢痕及成纤维细胞是否有影响及其机制尚不清楚。

姜黄素对肝癌细胞凋亡及TNF-ɑ、IL-1β、IL-6炎性因子影响的机制研究周宁;范萍;戴旖旎;白旭光;王瑜;贺勇毅;田蓉;王锻;黄晶晶;张雪竹【期刊名称】《癌症进展》【年(卷),期】2017(015)011【摘要】目的探讨姜黄素对肝癌细胞凋亡及肿瘤坏死因子α(TNF-ɑ)、白介素1β(IL-1β)、白介素-6(IL-6)炎性因子的影响及机制.方法采用5、10、20、40、80、160μmol/L的姜黄素处理人肝癌HepG2细胞24、48 h后,CCK8实验检测细胞的增殖情况,计算IC50值;50μmol/L的姜黄素处理HepG2细胞48 h后,流式细胞仪检测细胞凋亡情况;Western blot检测TNF-ɑ、IL-1β、IL-6、活化的含半胱氨酸的天冬氨酸蛋白水解酶3(Cleaved Caspase-3)、Notch1、Hes1蛋白表达情况.结果细胞抑制率随着姜黄素作用时间及浓度的增加显著升高.根据IC50值选择50μmol/L的姜黄素作为研究对象;以50μmol/L的姜黄素处理肝癌细胞的凋亡率及Cleaved Caspase-3蛋白表达明显高于对照组,TNF-ɑ、IL-1β、IL-6、Notch1、Hes1蛋白表达均明显低于对照组(P﹤0.01).结论姜黄素可呈时间及剂量依赖式抑制人肝癌HepG2细胞增殖,可诱导细胞的凋亡,可通过降低TNF-ɑ、IL-1β、IL-6的表达达到抗炎作用,其机制与抑制Notch1信号通路有关.%Objective To investigate the effect and its mechanism of curcumin on the apoptosis of hepatocellular car-cinoma (HCC) cells and TNF-ɑ, IL-1βand IL-6 inflammatory cytokines. Method 5, 10, 20, 40, 80, and 160μmol/L of curcumin were applied to treat human HCC cell line HepG2 for 24 h or 48 h, the cell proliferation was detected by CCK8 assay, and IC50 wascalculated;cell apoptosis in HepG2 cells was detected after treatment by 50μmol/L of curcumin for 48 h by flow cytometry;the expression of TNF-ɑ, IL-1β, IL-6, Cleaved Caspase-3, Notch1, Hes1 protein were detected by Western blot. Result The cells inhibition rate increased as the time elapsed and concentration elevated. 50 μmol/L curcumin was selected as the research target according to the IC50 value;the apoptosis rate of cells and Cleaved Caspase-3 protein expression in 50μmol/L curcumin treatment group was significantly higher than that in control group, and the pro-tein expression of TNF-ɑ, IL-1β, IL-6, Notch1, Hes1 were significantly lower compared with those in control group (P<0.01).
