Screening of the Gene for Chlorella Identification and Identification of oil--producing Microalgae

Abstract 『Objective]The aim was to select suitable gene for Chlorella identification and to identify the o.I.producing microalgae.[Method] Four candidate geno sequences,the nuclear genomic rDNA of the 18S r开』 gene,internal transcribed spacer(ITS),internal transcribed spa,c- er II(ITS II)and the chloroplast rbcL gene。were selected for Chlorella molecular identification.Through lhese four candidate genes,the genetic variability and distinguish ability between intra—species and inter・species was analyzed to choose the right genes for identification of the high oil- content Ch/ore//a.On this basis.application of these gene segments were classified and identified for five fresh—water isolated Ch/ore//a,which oil.content is more than 3o%.f Result],丁s gene was a suitable gene because of its high variation and short fragment length,meanwhile its ge- netic distance intra—species(0.439 6±O.135 9)WEtS largerthan inter-species(O.O45 7±O.0843).Its sequence length varied between diferent species whereas highly conserved in the same specias.By the application of ITS sequences,respectively,秆ve high oiI-content stains were{den- tifed as one vulgaris。two strains ofG sorokinianaandtwo strains of algaeChlorella sp.[Condmien】Thisstudy had provided referencefor the establishment of identification gene poel of Chlorella. Chlorella is an excellent bio—energY raw materiaI.which is widely distributed with fast growth rate,strong environmen- taI adaptability and high fat content….Chlore/la has many va- rieties。and diferent varieties show diferent adaption ability to environment.For example,the survive temIperatures of difer- ant varieties of Chlorella range fr0m 26 tO42℃ J.The classi- fication and identffication of the selected Chlore#a.as well as the selection of suitable cultivation conditions for diferent vari・ eties of Chlore/la based on their characteristics show important significance on the cultivation of Ch/ore//a and production of oiI. The traditional identification of Algae sl0ecies mainly based on morphological means by using optical and electron microscope for the observation and description of the fine structure of algae cells.However,the Chlorella cells are smal1.and the cells show certain changes in menstruaI cycle, thus making the morphological identification more dificult .
