1、在使用ASF-800 自动进样、收集装置时,请您在使用过程中不要随便移动、变更样品盘上的样品架,因为该位置为我们的工程师再给您安装时以调整好了的,否则将会使进样针折断等事故发生。
在实验结束以后,请尽可能的对其做一些防尘措施,以便延长其使用的寿命。
2、对于仪器的凝胶净化(GPC)模块而言,务必要对所使用的流动相(乙酸乙酯:环己烷=1:1)溶液进行超声、过滤处理以后方可在仪器中运行,否则将会使仪器的放空滤片座堵塞,从而导致仪器的高压泵压力超限而无法进行后续的操作。
若出现此类情况,请将仪器的放空滤片座拆除下来,超声清洗后,在安装上使用。
3、在使用GPC时,请注意流动相对凝胶色谱柱的影响, 即,在选择溶剂时,应该选择与仪器、净化柱兼容的溶剂及样品溶解性相符的溶剂,对于柱体上标有流动相为乙酸乙酯:环己烷=1:1的凝胶色谱柱,流动相只能使用乙酸乙酯:环己烷=1:1,尤其是不能使用二氯甲烷作为它的流动相,否则将会对凝胶色谱柱的填料的膨胀系数造成影响,从而使得柱子不能够有效的使用。
另外,在较长时间内不使用GPC凝胶系统时,请您将凝胶色谱柱取下,用特有的聚四氟堵头将其两端封好,在下次使用时在安装上,然后以其所使用的流动相冲洗十分钟以上(使其基线达到平稳)后,在进行使用。
4、GPC操作的注意事项a、分离原理的局限需注意的是,任何方法都不是万能的。
凝胶净化的原理是依据分子尺寸的排阻,若杂质与目标物的分子量接近,GPC就无法分离。
b、流动相纯度标准方法中使用的GPC流动相为乙酸乙酯:环己烷 1:1的流动相,考虑到降低本底效应,建议客户使用农残级的试剂,若没有也需将分析级或色谱级的试剂进行重蒸净化后方能使用。
c、流动相的用量由于方法中的流速为5mL/min,如果实验样品的数量很多,应该一次性配好流动相充足的量,避免中途在往试剂瓶里添加。

3、对称放置离心管,14520rpm(即14000g)离心。
(离心5min约浓缩2倍,10min约4倍,15min约6倍,20min约8倍,30min约10倍)。
4、离心结束后,将内管倒置套在另一个干净的离心管呢,3880rpm离心min收集浓缩后的样品。
或者用移液器直接吸出内管中的样品。
2.2 浓缩管使用后的处理和保存
1、使用完的浓缩管,立即加入500µl含1MNaCl的缓冲液(此缓冲液与浓缩样品的缓冲液一致),8000rpm离心20min。
2、甩净内管中的盐溶液,将内管放入超纯水中浸泡1h。
3、内管加入超纯水500µl,8000rpm离心5min。
4、甩净内管中的水,加入500µl0.2MNaOH,8000rpm离心20min。
5、甩净内管中的NaOH溶液,用超纯水再8000rpm离心5min。
6、甩净内管中的水,放入含0.2-0.5‰NaN3的生理盐水中。
7、外管用自来水洗净后,用超纯水冲洗干净,晾干。

4.2验证目的采用物理、化学分析方法和微生物检测方法来确认按《双效浓缩罐清洁操作规程》对双效浓缩罐进行清洁后,该设备内表面最难清洁部位的药品最难溶活性成分残留量及微生物数不超过规定的限量,证明《双效浓缩清洁操作规程》的可行性和可靠性,以表明本清洁方法能够达到防止污染与交叉污染的目的,特制定本验证方案进行验证。

2.2 浓缩管使用后的处理和保存1、使用完的浓缩管,立即加入500µl含1MNaCl的缓冲液(此缓冲液与浓缩样品的缓冲液一致),8000rpm离心20min。
2、甩净内管中的盐溶液,将内管放入超纯水中浸泡1h。
3、内管加入超纯水500µl,8000rpm离心5min。
4、甩净内管中的水,加入500µl0.2MNaOH,8000rpm离心20min。
5、甩净内管中的NaOH溶液,用超纯水再8000rpm离心5min。
6、甩净内管中的水,放入含0.2-0.5‰NaN3的生理盐水中。
7、外管用自来水洗净后,用超纯水冲洗干净,晾干。

也有其它型号的、不同体积大小和MWCO超滤管可选,视目的蛋白的分子量与浓缩前体积、浓缩目标体积而定。
可重复使用,使用一次就扔掉太浪费。
以下是个人总结的使用方法和注意事项。
1、选择合适的超滤管,主要考虑MWCO和浓缩体积,最常见的是Ultra-15(10kD)。
到底选择多大截留分子量比较合适呢?10kDa、5kDa、还是30kDa?通常应截留分子量不应大于目的蛋白分子量的1/3,比如目的蛋白分子量为35kDa,就可以选择10kDa截留分子量的超滤管。
