BBM培养基——Bold’s Basal Medium(Deason &bold 1960;Bischoff &Bold 1963)NaNO3 250mg KH2PO4 175mg
K2HPO4 75mg MgSO4·7H2O 75mg
CaCl2·2H2O 25mg NaCl 25 m
EDTA 50mg KOH 31mg
FeSO4·7H2O 4.98mg H3BO3 11.42mg
ZnSO4·7H2O 8.82mg MnCl2 1.44mg
MoO3 0.71mg CuSO4·5H2O 1.57 mg
Co(NO3)2 ·6H2O 0.49mg 加蒸馏水补足1000ml
可以在以上组份中加入15~20g琼脂,并蒸馏水补足1L制成固体培养基。
SE(Selenite Enrichment)培养基(配方1)
NaNO3 0.25 (g/l)
K2HPO4.3H2O 0.075 (g/l)
MgSO4.7H2O 0.075 (g/l)
CaCl2.2H2O 0.025 (g/l)
KH2PO4 0.175 (g/l)
NaCl 0.025 (g/l)
Soilextract(土壤提取液) 40 (ml/l)
FeCl3•6H2O 0.005 (g/l)
Fe-EDTA 1(ml/l)
A5 solution 1(ml/l)
distilled water 958(ml/l)
A5溶液配制方法:
H3BO3 286 mg
ZnSO4 . 7H2O 22 mg
MnCl2. 4H2O 181 mg
CuSO4. 5 H2O 7.9 mg
(NH4) 6Mo7O24 . 4H2O 3.9 mg
按按上述称量药品,溶于100ml水即可。
EDTA-Fe的配置方法:
A液: Na2EDTA 1g
distilled water 50ml
B液FeCl3•6H2O 81 mg
HCl(0.1N)(0.1 mol/L) 50ml
分别配置A,B液,充分搅拌溶解后,将A,B液混合搅拌均匀即可。
SE培养基配方2 (Brostol’s solution) Component Amount Stock Solution
(1) NaNO3 1 mL/L 25 g/100mldH2O
(2) K2HPO4 1 mL/L 7.5 g/100 mL dH2O
(3) MgSO4·7H2O 1 mL/L 7.5 g/100 mL dH2O
(4 ) CaCl2·2H2O 1 mL/L 2.5 g/100 mL dH2O
(5 ) KH2PO4 1 mL/L 17.5 g/100 mL dH2O
(6 ) NaCl 1mL/L 2.5g/100ml dH2O
(7) FeCl3. 6H2O 1mL/L 0.05g/100ml dH2O
(8) EDTA-Fe 1mL/L
1N HCl: 取4.1ml浓盐酸用蒸馏水稀释至50ml1N EDTA-Na 2 称取0.9306g 溶解至50ml蒸馏水中称取FeCl3. 6H2O 0.901g 溶于10ml以上步骤已经配制完成的1N HCl 中,然后与10ml已经配制完成的0.1N EDTA-Na 2 混合,加入蒸馏水稀释至1000ml。

(10)土壤提取液40 mL/L
土壤提取液配制方法:
取花园土未施过肥200g置于烧杯或三角瓶中,加入蒸馏水1000毫升,瓶口用透气塞封口,在水浴中沸水加热3小时,冷却,沉淀24小时,此过程连续进行3次,然后过滤,取上清液,于高压灭菌锅中灭菌后于4℃冰箱中保存备用。
