The kamado differs from a regular barbecue in its versatility. Thanks to the good insulation, very accurate temperature control is possible and you can also smoke, slow cook, stew and bake pizzas on the ceramic BBQ. The kamado is fired on charcoal, which gives a real wood and fire flavor to the product.During the first 3 uses of the kamado you are obliged to follow the followingsteps! Incorrect use will void the warranty.Preparation:•Make sure that the Grill Guru kamado is always well and firmly in its stand. Check that all bolts and nuts of the undercarriage are tightened and that the wheels are on their brakes.•Test the opening and closing of the kamado and check whether the bolts to the steelwork are properly tightened and there is no large play on the hinge.•Open the Top Cap and the air slide both slightly for the first 3 uses of the kamado . If these are opened too far, there is a risk ofcracking the ceramic.•Fill the firebox halfway with charcoal only. The use of briquettes is strongly discouraged. Briquettes are made from pressed residual materials and cannegatively affect the taste of your food•During the first 3 uses of the kamado, the charcoal should be lit in a maximum of 1 place. When the charcoal is lit in several places at the same time, thetemperature rises too quickly and there is a risk of cracking the ceramic. Thekamado should not get hotter than 200 degrees.•Several tools and articles can be used to light the kamado. One of those tools is the Grill Guru One Minute Lighter. With this tool the charcoal can be lit safely andquickly. Other options are firelighters or woks, but make sure that these aremade of a natural material!•Never use chemical firelighters, spirits or other flammable substances with a kamado. These are dangerous and can also penetrate into the ceramics, whichcan negatively affect the taste of your dishes.•When you have ignited the charcoal in 1 place, place the Plate Setter back on the fire box, also place the ceramic Plate Setter stone in the Plate Setter. Finally, thegrill can be placed on the Plate Setter. Now the lid of the kamado can be closed. Before you can start, the temperature has to increase a little bit, but don't do this too quickly. Temperatures rising too quickly can be bad for the ceramic. With the first 3 uses, slowly bring the kamado to 200 degrees in at least 60 minutes.•In the preparation steps, it has been checked whether the Top Cap and air slide are slightly ajar, check again whether this is the case.•The temperature of the kamado can be adjusted by opening and/or closing the air slide and top ventilation further. We will look at this in detail later. For now it is important to have both open slightly ajar.•With the first 3 uses, let the temperature of the kamado slowly climb to 200 degrees Celsius within at least 60 minutes so that moisture that has beenabsorbed into the ceramic can slowly evaporate and the ceramic is BBQ ready fora long time.•If the temperature rises too quickly, close the air slide and top ventilation further.•If the kamado does not reach the desired temperature and sticks, gradually open the Top Cap and the air slide and wait for the temperature to react.•Never leave the kamado unattended when it is on.•Never open a warm kamado in 1 go! Always open the lid slightly, wait a few seconds and then open fully. When the lid is opened in one movement, there is a risk of a flash fire due to a sudden excess of oxygen.•Stay close to the Kamado. The first few times it always takes some getting used to how it works with adjusting the temperature of the BBQ, for this werecommend that you always stay nearby and keep a close eye on thethermometer.•Use appropriate tools and gloves when moving food, grill and plate setter stone. A kamado gets very hot and can cause serious burns if used incorrectly.•The height of the temperature is very much dependent on the amount of oxygen and other conditions such as wind, humidity, type of coal and the outsidetemperature. Controlling the temperature is therefore almost never 100% thesame. It is not that difficult to achieve a constant high temperature of, for example, 250 degrees Celsius or a low temperature of 80-90 degrees Celsius. You can alsoachieve a constant low temperature for up to 12 hours without adding newcharcoal. You control the temperature with the top ventilation disc and the bottomventilation. To slowly increase the temperature, start with the top and bottom vents only slightly open. You can slowly open it a little further to increase thetemperature. The more oxygen, the higher the temperature.•Do you want to put out the fire? Then you close the top and bottom ventilation. It couldn't be easier! Note: do not use water to extinguish the coals, as this candamage the ceramic.•TIP: Always use a plate setter for slow cooking at a low temperature. Do not let the grill get too hot. It takes hours to bring the temperature down. If you want to lower the temperature, slide the top and bottom ventilation closer.•Remove all food residues. With burnt food remains , it helps to fire up your BBQ first. After cooling, you can easily remove the food residues with a BBQ brush or scraper. You can grease the stainless steel grill grate with vegetable oil to prevent rust.•Close the Top Cap and air slide. Due to a lack of oxygen, the coals in the firebox will slowly extinguish and the BBQ will slowly cool down again. This leaves coal that can be used again the next time. It may take a while to cool down as theceramic can retain its heat for a very long time. Depending on the amount of coal left and of course the temperature at which the barbecue was used, cooling can take several hours or even an entire night. We recommend that you only cover the kamado with a rain cover when it has completely cooled down. Neverextinguish the kamado with water.•Leave the kamado closed until it has cooled completely. The coals in the kamado can continue to smolder for a long time and can catch fire again if the kamado is opened too early .•When the kamado has completely cooled down, the grids can be removed and cleaned.•Charcoal left over from a BBQ session can remain in the firebox and be used the next time.。

GoStix Plus cytokine assays are designed to accurately quantify human cytokines extremely rapidly, using only 20 µl of cell culture supernatant or diluted serum. Ten minutes after applying your sample, a band will appear in the window of the GoStix cassette at an intensity that correlates with the amount of analyte applied. The cassette is then scanned using a smartphone camera or equivalent mobile device running the GoStix Plus app that will calculate pg/ml of a given cytokine based on the intensity of the band. Control protein included with each kit allows for confirmation of GoStix function. See Section III, Table 1 for applicable assay information. To learn more about GoStix assays and the GoStix Plus app, visit /gostixhelp.Figure 1. Cytokine GoStix Plus workflow. When you first use the app, you will need to connect your device to the internet so that the standard curves for available lots of cytokine GoStix Plus products can be downloaded into the app. The app uses the standard curve to calculate pg/ml of cytokine. Contact your network administrator if you experience issues downloading standard curve data or if the app cannot find your lot number upon initial entry. If the problem persists, please contact Technical Support.I. Testing Your Sample1.Download the GoStix Plus app from the App Store® or on Google Play™.NOTE: The GoStix Plus app has not been validated for use with tablets.2.Open the app on your mobile device and enter a valid email address to begin. Click [Start].NOTE: This email can be used for sending results to the user from the mobile device (see Step 15).3.Enter the lot number by scanning the QR code on the foil pouch containing the GoStix cassette. (Press the QRcode icon to activate the scanner.) The lot number can also be entered manually.NOTE: Do not use the QR code on the outside of the box; it is not recognized by the app software.4.Enter the number of tests to be scanned (1–8 tests). The appropriate number of sample name windows willappear below.5.Enter in the sample names and the appropriate dilutions. Click [Start].NOTES:Assay range: Each cytokine GoStix Plus assay is designed to accurately quantify a given cytokine within adesignated range (specified in the corresponding Certificate of Analysis for the assay). Similar to ELISAmethods, first-time users are advised to consider running the assay on several sample dilutions in addition totesting an undiluted sample to ensure that they obtain a valid reading within the designated range of the assay-specific standard curve.Serum samples: We recommend diluting serum samples by at least 1:10 to minimize assay variability. TheGoStix Plus app includes a dropdown menu for entering dilutions as high as 1:100,000.6.Following the prompts within the app, take 20 µl of your sample and apply it to the sample well (S) of theGoStix cassette. If you expect the sample to have high levels of analyte, you can perform dilutions prior toloading the sample.7.Add 80 µl of chase buffer to the sample well and allow the chase buffer front to appear in the cassettewindow.NOTE: For better reproducibility, dilute 20 µl of your sample with 80 µl of chase buffer and then load theGoStix cassette with the resulting 100 µl.8.Press [Start] to activate the timer on the app.9.Allow the lateral flow test to run for the full 10 minutes. A test band (T) will start to appear within 5 minutesand reach maximum intensity at 10 minutes if your sample contains sufficient levels of analyte. The controlband (C) will always appear when the test is functioning properly.NOTE: The test will not give consistent results if the full development time is not observed. A warningwithin the app will appear if the [Skip] button is pressed before the 10 minutes has expired. For accurateresults, it is important that the intensity of the test band is less than that of the control band. The closer thisratio is to 1, the closer the sample is to exceeding the standard curve. If a reading exceeds the standardcurve, the app will return a Gostix Value (GV) of “Off Scale” and an error message will appearrecommending that the sample be diluted. If no analyte is detected in the sample, the app will return aGV of 0 pg/ml.10.After 10 minutes, the app will alert you to take a picture of the cassette. Proper alignment and focal length forimaging is achieved by using the outline of the cassette in the scanning window. Your sample name willappear below the outline of the cassette. Once proper alignment is achieved, the outline will turn green andthe cassette will automatically be scanned.NOTE: Depending on the device used,it may be necessary to tap the device screen in order to focus prior toscanning. Also, if possible, avoid creating shadows when imaging the GoStix cassette.11.Once all samples are read, the results will be displayed in the Result detail window.12.If desired, add notes for each sample in the “N otes” section of the Result detail window, at this time.13.Press [Done] when finished to save each data entry.14.If you wish to rescan a sample, repeat steps 3, 4, and 5, skip the protocol page(s) and timer (press [Start],[Skip], and then [Yes]) to proceed. Do not exceed 20 minutes of total development time (i.e., rescanning must take place within 10 minutes of initial timer expiration).NOTE: Lateral flow tests can continue to develop after the initial 10-min development time, such thatvariation in development time can contribute to read variability. If replicate reads from the same test aredesired, we recommend timing the acquisition of the images as close to each other as possible. Typicalcoefficients of variation (%CV) for replicate reads of the same test vs. replicate tests of the same sample are<10% and <15%, respectively.15.If desired, individual results from the Result history page (accessed from the main menu) can be emailed orsent via SMS by pressing the [Share] button at the bottom of each overview page. Accumulated results canalso be downloaded as a single batch to your device using the [Download all] button on the Result historypage. Please visit /gostixhelp for further instructions on how to access results downloaded toyour device.II. Calibrating GoStix results with other assays using a reference standard GoStix assay results can be calibrated to international standards using conversion factors specific to each assay. These conversion factors are provided in the corresponding assay’s Certificate of Analysis.III. Testing the cytokine control protein1.Control protein for each assay is supplied in a dried format. To resuspend, add 500µl of chase buffer andvortex.2.Dilute the control protein for your specific assay as indicated in the table at the bottom of this section.NOTE: The control protein is provided to confirm the proper function of the lateral flow test. Followinginitial resuspension in 500 µl of chase buffer, the control protein is too highly concentrated to yield a readingwithin the range of the stan dard curve referenced in the app, and will cause the app to return a GV of “OffScale”. Further dilution is required to ensure that the control reading falls within the standard curve.3.Apply 100 µl of the diluted control to the sample well (S) of the GoStix cassette.4. A control band (C) and test band (T) will appear in 10 minutes.5.For future control tests, the reconstituted control protein can be stored at 4o C for up to 1 week. For extendedstorage, prepare aliquots to store at –20o C and avoid repeated freeze-thaw cycles.Table 1. Recommended control protein dilutions.IFN-γ GoStix Plus 631285, 631286 1:8 (25 μl + 175 μl)Notice to PurchaserOur products are to be used for research purposes only. The “balanced force concept”, i.e. small clockwise and counter-clockwise movements, was developed over a period of twelve years, and proposed in 1985 by Roane as a means of overcoming the curvature influence. Using the balanced force technique, it is possible to shape curved canals with larger diameter hand instruments. The use of stainless steel hand instruments, however, is time-consuming and strenuous, and there is a high frequency of preparation errors. The development of continuous rotary preparation with nickel-titanium (NiTi) instruments solved some of these issues, although it is still necessary to use several hand and rotary files in different steps, and there may be a lengthy learning curve before proficiency can be achieved.Inspired to find a simpler and more convenient safe way to successfully prepare a root canal, Prof. Ghassan Yared, who was at that time Professor of the Endodontic Undergraduate and Graduate Programmes at the University of Toronto, started researching and testing mecha-nical reciprocation with NiTi instruments. In 2008, he published a clinical article on canal preparation using only one engine-driven NiTi instrument and joined together with VDW to develop RECIPROC®, a system specifically for use in reciprocation.4Reciprocation IReciprocation by VDWIn reciprocation, the instrument is driven first incutting direction and then reverses to releasethe instrument. One complete rotation of 360°is completed in several reciprocating movements. The angle in the cutting direction is greater thanthe angle in the reverse direction, so that theinstrument continuously progresses towards theapex. The angles of reciprocation are precise andspecific to the design of the RECIPROC® instru-ment and to VDW endo motors. They are designedto be smaller than the angle settings where theelastic limit of the instrument would be met, thusminimizing the risk of instrument fractures.5I ReciprocationThe standard with rotary systems up until now:initial hand filing to create a glide path. With continuous rotary NiTi systems it is necessary to create a glide path in order to mini-mize the risk of fracture due to instrument binding. During the use of a rotary instrument, the tip of the instrument may bind in the canal. For this reason, it is necessary to create an initial glide path, or a minimal canal enlargement, before using continuous rotary instruments. Just as with any continuous rotary NiTi system it is possible to use the RECIPROC® range instruments after creating an initial glide path with hand instruments (e.g. C-PILOT® File) to an ISO size 10 or 15. However, RECIPROC® and the reciprocating movement has opened a new option: using RECIPROC® without initial hand filing in the majority of cases.A paradigm shift in endodontics:using RECIPROC® without initial hand filing to create a glide path in the majority of cases.67System overview+o rRECIPROC ® blue is the new generation of a proven system. Back in 2011, VDW released RECIPROC ®, its biggest success in its history with over 7.5 million files sold and over 60 publications, becoming the market leader for reciprocating instruments worldwide. The RECIPROC ® blue system II9The RECIPROC ® blue system IIRECIPROC ® blue instrumentsRECIPROC ® blue is designed to be used as a single instrument. That means that one instrument only is required to prepare a root canal. One RECIPROC ® blue instrument does the job of several instruments which would need to be used for preparation with regular hand or continuous rotary instruments. The shape obtained by the RECIPROC ® blueinstrument enables effective irrigation and obturation with both cold and warm techniques.Narrow canalsR25 prepares the root canalto a diameter of 0.25 mmwith a taper of .08 over thefirst apical millimeters.Medium canals R40 prepares the root canal to a diameter of 0.40 mm with a taper of .06 over the first apical millimeters.Wide canals R50 prepares the root canal to a diameter of 0.50 mm with a taper of .05 over the first apical millimeters.ø16 mm 1.05 mm 3 mm 0.49 mm 2 mm 0.41 mm 1 mm 0.33 mm 0 mm0.25 mmø16 mm 1.17 mm 3 mm 0.65 mm 2 mm 0.60 mm 1 mm 0.55 mm 0 mm0.50 mmø16 mm 1.10 mm 3 mm 0.58 mm 2 mm 0.52 mm 1 mm 0.46 mm 0 mm 0.40 mmThe stopper, in the ISO color of the specificinstrument tip size, supportsinstrument when it is inserted in the contra-angle.The three points represent three movementsinstruments haveradiograph visible depth markingsWorking length 21 mm 25 mm 31 mm ShaftRECIPROC of 11 mm enabling better access to molarscompared to many other instruments which havea shaft of 13 mm or longer.w o r k i n g l e n g t h non-cutting tipFrequency of useA RECIPROC®blue instrument is designed for single use in maximum one molar. As with all NiTi instruments, it should be examined during the treatment and discarded if signs of wear, such as untwisting, can be seen. If an instrument appears to be bent after being used in a strongly curved canal, it should be discarded.Single use convenienceThe RECIPROC®blue system stands for convenience and safety. The instruments are delivered ready to use, pre-sterilizedin blister packaging and should simply be discarded after use, making work flow more efficient; eliminating the need to clean and sterilize instruments, considerably reduces the risk of contamination to office personnel and eliminating the risk of cross contamina-tion between patients. The RECIPROC®blue instrument cannot be autoclaved due to its non-autoclavable handle. This safety feature protects against metal fatigue caused by over-use.The RECIPROC®blue system IIRECIPROC®blue Paper PointsThese are highly absorbent, pre-sterilized RECIPROC®Paper Points corresponding to instrument sizes R25, R40 length control.RECIPROC®blue Gutta-PerchaThe RECIPROC®blue system includes RECIPROC®blue Gutta-Percha Master Cones. These master cones mirror the variable taper and shape of the corresponding RECIPROC®blue instruments and are available in the sizes R25, R40 and R50. Our injection molding process improves dimensional stability up to 40 % compared to traditional hand rolled gutta-percha cones. This allows a more consistent shape and fit.In addition, the unique gutta-percha formulation heats more consistently, providing better material flow and adaptation to the canal walls promoting a 3D-fill when used with warm vertical condensation for example with the BeeFill®2in1 system. Heat transfers through the gutta-percha cone upto 4 mm beyond the heat source ensuring warm, flowable gutta-percha that reaches all the way to the apex while en- abling the clinician to utilize a lower heat plugger temperature, thereby improving patient safety.II The RECIPROC®blue systemGUTTAFUSION ® for RECIPROC ® blueGUTTAFUSION ® for RECIPROC ® blue obturators havebeen developed for an easy, warm and homogeneous3D-obturation of the whole root canal system. Madeentirely of gutta-percha, the obturators can be heated inthe GUTTAFUSION ® oven and can be placed preciselyinto the root canal either by hand or with tweezers, thanksto the specially designed handle. The obturators correspond to the RECIPROC ® bluepreparation system and are available in the sizes R25,R40 and R50.RECIPROC ® endo motors from VDWVDW.CONNECT Drive ®, VDW.GOLD ®RECIPROC ® and VDW.SILVER ®RECIPROC ® endo motors are designed to enable the use of both reciprocating and continuous rotary NiTi systems. Thanks to the intuitive menu navigation and clearly structured display, the motors ensure easy and convenient use. This function is a two-stage indicator, which signals when to switch to a brushing file motion in order to reduce stress on the instrument during preparation and to enable an easy advancement towards the apex. RECIPROC REVERSE supports the user in working with the RECIPROC® system by giving a first acoustic signal to indicate an increase in friction experienced by the instrument in the root canal. In order to reduce friction, the instrument should temporarily be used in a lateral brushing motion towards the coronal section. This creates space in the root canal and the instrument can further advance towards the apex by using a pecking motion. If the instrument is subjected to further stress, a second acoustic signal is given and the motor will automatically start rotating in a clockwise direction. In this way, stress on the instrument will immediately be reduced. By releasing and pressing the foot pedal again, the motor will resume the reciprocating motion. However, the instrument must be cleaned, the canal irri- gated and an ISO size 10 C-PILOT® File should be used in order to check that the canal is not blocked before resume its reciprocating motion. The preparation can then be continued by temporarily using lateral brushing motions until the instrument can easily advance towards the apex (see “Canal preparation step by step and recommendations” on page 18).Advantages of reciprocation and the RECIPROC ® blue system One file endo. Less work steps than continuous rotary files.Chair-side preparation isreduced to a minimum, asthe RECIPROC ® blue instru-ment comes pre-sterilized.A root canal can be com-pletely prepared to a greatertaper with only one recipro-cating instrument. There isno need to change instru-ments in the contra-angleduring preparation.RetreatmentGutta-percha filling material and carrier based obturators can be removed with R25. Glide path management RECIPROC ® blue can beused without initial handfiling in the majority of cases. When a reciprocating in-strument binds in the canal,it will not fracture becauseit will never rotate past its specific angle of fracture. Therefore, the creation of a glide path to minimizebinding is not required forthe RECIPROC ® blueinstruments.Time-saving and easy to learn Designed for convenience, RECIPROC ® blue instruments are used on only one patient and then simply discarded, eliminating two work steps – cleaning and sterilizing.Centering ability In reciprocation, the instru-ment stays better centered in the root canal. Large in-struments with a large core can safely and efficiently negotiate even narrow and strongly curved canals. The risk of instrument fracture is minimized The innovative heat treat-ment gives increased resis-tance to cyclic fatigue and additional flexibility to the file. In addition, the angles of reciprocation are specific to the design of the RECIPROC ® blue instrument. They are smaller than the angle settings where theelastic limit of the instrumentwould be met, thus minimi-zing the risk of instrumentfractures.IIISelecting the correct RECIPROC ® blue instrumentApplication examplesPre-Operative Radiograph DECISIONcanal is partially or completelyinvisible canal is completely visible wide or medium canalhand instrument ISO 30goes passively*to working length hand instrument ISO 20 goes passively* to working length(small right left rotations) but without filing action.NoYes Yes No Canal is considered narrow: R25Preparation with RECIPROC ® blue IV Canal clearly visible from access cavity to apex: consideredmedium or wide (R50 was used for the canal preparation:an increased apical enlargement was obtained with a size 70hand file)Clinical orthography, Prof. Ghassan Yared, CanadaPreparation with RECIPROC® blue IVElectronic length determinationNarrow canalsBefore commencing preparation, the length of the root canal is estimated with the help of an adequately exposed and angulated pre-operative radiograph. The silicone stopper is set on the RECIPROC® blue instrument at 2/3 of its length.During preparation with R25, after approximately 2/3 of the root canal has been prepared, use a C-PILOT® File or a K-File ISO size 10 and an apex locator such as RAYPEX®6to determine the length of the root canal. The silicone stopper then can be set on the RECIPROC®blue instrument at this determined length.Medium or wide canalsBefore starting preparation with R40 and R50, determine the working length with an apex locator such as RAYPEX®6 by using a C-PILOT® File or a K-File. Set the silicone stopper of the RECIPROC® blue file at that length. After the coronal and middle thirds of the canal have been prepared, the working length should be re-checked.IV Preparation with RECIPROC ® blueCanal preparation step by step and recommendations*Ensure you have achieved a straight line access to the root canal orifice.
