以奥沙西泮为原料,制备得到纯度99%以上的杂质Ⅱ,方法条件温和,操作简单,收率高,合成的杂质Ⅱ可用于杂质的定性和定量分析,从而提高奥沙西泮的用药安全性。
关键词:奥沙西泮;6-氯-4-苯基喹啉-2-甲醛;制备中图分类号:R 971+.3 文献标识码:A 文章编号:1671-9905(2020)08-0020-03作者简介:付林(1972-),男,工程师,从事药物研究开发收稿日期:2020-05-15奥沙西泮(Oxazepam)属于苯二氮类镇静催眠药,具有显著的镇静、抗焦虑作用,同时具有较好的松驰肌肉和抗抽搐作用,能缓解神经官能症患者的焦虑不安、精神紧张及烦恼失眠等症状,用于神经官能症、失眠及癫痫的辅助治疗,适用于老年人或肾功能不全者。
在同类苯二氮卓类镇静催眠药中,奥沙西泮的代谢过程简单,受年龄及肝功能影响较小;半衰期较短,不易在体内蓄积;成瘾性较小,可广泛应用于成瘾行为的基础研究[1]。
进行奥沙西泮杂质的质量研究,对于原料药研究和制剂研究有着重要而深远的意义。
有关物质是药品的关键质量属性之一,也是仿制药一致性评价的重要内容,涉及到药品的安全性及其质量的可控性[2]。
随着仿制药市场的逐步扩张及国内仿制药一致性评价的进行,制药企业对杂质对照品的需求迅猛增长。
中国药典2015版中,对奥沙西泮原料及其片剂中有关物质的检测项规定了2种已知杂质,分别为(3RS)-7-氯-2-氧代-5-苯基-2,3-二氢-1H -1,4-苯并二氮杂-3-乙酸酯(杂质Ⅰ)和6-氯-4-苯基喹啉-2-甲醛(杂质Ⅱ)[3]。
3.2.P.5.5杂质谱分析模板的整理格式模板首先列出产品的杂志谱列表,比如:****产品杂质情况分析表反应过程的描述:1、详细的反应方程式,包括结构式,反应温度,所有试剂,助剂,溶剂,催化剂等。
2、结合CTD资料的其他部分,对物料控制进行说明,包括起始物料、其他原料、溶剂、辅料(活性炭,硅藻土,硅胶等。
)。
3、起始物料说明。
起始物料符合广泛、易得、质量稳定,适合保存运输等的原则;还应对多个供应商提供的多批次物料进行质量研究,同供应商签署的质量协议以及供应商工艺变更告知义务等协议。
比如头孢克洛,要对起始物料7-ACCA的工艺、杂质控制和质量情况进行详细的说明。
4、说明制定起始物料的质量控制策略的依据,比如头孢克洛的起始物料7-ACCA的关键杂质△异构体,结合工艺和实验数据,说明杂质产生来源,分布,控制策略等。
第2、3、4内容可以在CTD的其他部分,比如物料说明部分进行。
但是本部分内容讨论的展开需要物料控制说明作为基本的理论依据。
5、关于ICH的杂质鉴定、报告和质控限度:主要参考ICHQ3A(R2)到ICHQ3D的相关规定2 克/天结合上述反应过程对杂质谱进行分析,主要分起始物料引入杂质,反应杂质,降解杂质等。

奥美拉唑杂质奥美拉唑( omeprazole),化学名为5-甲氧基-2 - [ [ ( 4-甲氧基-3,5-二甲基-2-吡啶基)甲基]亚硫酰基]-1-苯并咪唑,是英国AstraZeneca公司研发的质子泵抑制剂类抗溃疡药,临床主要用于治疗消化道溃疡和反食性胃炎。
欧洲药典(European Pharmacopeia ,缩写为EP)共列出其杂质9个。
奥美拉唑及其九由于合成方法和步骤的不同,所产生的杂质也有所不同。
根据本实验所采用的方法及所使用的原料情况,现分析其可能产生杂质如下:1. Omeprazole Imp. A (EP):化学名5-Methoxy-1H-benzimidazole-2-thiol ,是合成奥美拉唑中间体奥美拉唑硫醚的一个原料。
该方法的具体步骤为:500ml 三颈圆底烧瓶1只,安装搅拌轮,100。
C 温度计和加液管置于恒温水槽中。
烧瓶内加入苯并咪唑物(5-Methoxy-1H-benzimidazole-2-thiol )26.8g (0.15mol )、去离子水30ml 、NaOH6.0g(0.15mol)和乙醇200ml ,搅拌溶解后加入氯甲基物(2-Chloromethyl-4-methoxy-3,5-dimethylpyridine hydrochloride )33.3g (0.15mol ),加热搅拌回流3h 。
反应完毕,冷却,过滤,滤液用乙酸调至pH=7,加入去离子水适量,用乙酸乙酯萃取4次,合并有机层,水洗,无水Na 2SO 4干燥。
过滤,滤液减压蒸馏,剩余物用乙酸乙酯重结晶得米黄色固体,即奥美拉唑硫醚。
(李鸿运.奥美拉唑合成工艺研究.哈尔滨商业大学学报(自然科学版),2006).本实验所用方法与其所用方法与其相似,但有所改进,将氢氧化钠得使用量加大3倍,用乙酸调PH=8~9。
所以反应中有该杂质。
由于其为原料,所以较易获得。
2. Omeprazole Imp. B (EP):化学名2-[(R ,S)-[(3,5-Dimethylpyridin-2-yl)methyl]sulphinyl]-5-methoxy-1H-benzimi dazole ,是另一种方法所产生的副产物,该方法使用2-氯甲基-3,5-二甲基-4-硝基吡啶-N-氧化物与苯并咪唑物进行反应,在甲醇钠进行取代时未反应完全会产生该杂质,(颜国和,王飞武.奥美拉唑合成路线图解[J].中国医药工业杂志,1991,2 2( 6 ) :2 83-28)本反应与其使用方法不同,反应历程也不相同,最终结果未检测到该杂质,所以不对该杂质进行研究。

超级微波消解—ICP-AES法测定铱粉中16种杂质元素谢 阳1,赵志虎1,白伟道2,宋 翔1,孙海荣1(1.金川集团股份有限公司检测中心,甘肃 金昌 737102;2.金川集团铜业有限公司,甘肃 金昌 737100)摘 要:本方法充分发挥超级微波仪消解过程简单、产率高、消耗成本低及酸用量少、引入的污染少、安全、环保的优点,将超级微波技术与ICP-AES相结合,建立了 ICP-AES法测定铱粉中16种杂质元素的分析方法[1],选用盐酸和双氧水于聚四氟消解管(TFM管)溶解铱粉,避免了氯酸钾和石英管带来的盐类污染和硅污染,方法简便、快捷,填补了铱粉中硅元素分析的空白,能满足铱粉产品测定要求。
