BIOLOGICAL ACTIVITY:
Astragaloside II is a natural isolated from Astragalus.

In vitro:
In vivo: The developed and validated method has been successfully applied to the quantification and pharmacokinetic study of AST II in rats after intravenous and oral administration of AST II. The oral absolute bioavailability (F) of AST II was calculated to be 0.79 ±0.16% with an elimination half–life (t1/2) value of 1.92 ± 0.30 h, suggesting its poor absorption and/or strong metabolism in vivo [1].PROTOCOL (Extracted from published papers and Only for reference)
Cell assay [3] The in–vitro cytotoxicity of the drugs was determined using the MTT assay. Bel–7402 cells and Bel–7402/FU cells were harvested and seeded at 1 × 104 cells per well in 96–well plates and cultured for 12 h at 37°C in an atmosphere containing 5% CO2.The cells were treated with 0.04, 0.08, 0.16, 0.32, or 0.64 mg/ml Astragaloside II in the presence of 10% serum medium. Following 48 h of treatment, using the same MTT assay as above, values were calculated by SPSS.

ST2 (growth STimulation expressed gene 2; also known as IL1RL1, or Interleukin-1 ReceptorLike 1) is a member of the interleukin-1 receptor family. The ST2 protein has two isoforms directly implicated in the progression of cardiac disease: soluble ST2 and a cell membranebound isoform, ST2L. When soluble ST2 levels are low, ST2’s ligand, IL-33, is available to bind to ST2L and has a cardioprotective effect resulting in preserved cardiac function. However, when soluble ST2 levels are high, soluble ST2 competitively binds to IL-33, making IL-33 less likely to bind to ST2L and thereby making IL-33 unavailable for cardioprotective signaling. As a result, the heart is subjected to greater stress in the presence of high levels of soluble ST2, leading to cellular death and tissue fibrosis, reduced cardiac function, and an increase in the rate of disease progression (Kakkar & Lee 2008; Seki et al. 2009). The Presage® ST2 Assay quantitatively measures the concentration of soluble ST2, providing a physician with an accurate tool to assess prognosis in patients with chronic heart failure.

细胞周期索A2检测试剂盒说明书实验原理：本试剂盒应用双抗体夹心法测定标本中血清素/血清胺(ST)水平。

用纯化的血清素/血清胺(ST)抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入血清素/血清胺(ST),再与HRP标记的血消素/血清胺(ST)抗体结合，形成抗体-抗原-的标抗体梵合物，经过彻底洗涤后加底物TMB显色。

TMB在HRP酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。

颜色的深浅和样品中的血清索/血清胺(ST)呈正相关。

用假标仪在4SO AM波长下测定吸光度(OD值)，通过标准曲线计算样品中血清素/血清胺(SD浓度。

试剂盒性能：1.灵敏度：Z山小的检测浓度小于1号标准品。

稀释度的线性。

样品线性回归与预期浓度相关系数尺值为Oqqo2∙特异性：不与其它细胞因子反应。

3.重且性：板内、板间变异系数均小于工O%∙标本要求：1 .标本采集后尽早进行提取，提取按相关文献进行，提取后应尽快进行实验。

若不能马上进行试验，可将标本放于-2。

C 保存，但应避免反完冻融2 .不能检测含NaN3的样品，因NaN3抑制辣根过氧化物陶的(HRP)活性。

样本处理及要求，1■.血清：全血标本请于室温放置2小时或4C过夜后于工Og离心2。

分钟，取上清即可检测，或将标本放于-WC或-8OC保存，但应避免反复冻融。

2∙血浆：可用EDTA或肝素作为抗凝剂，标本采集后30分钟内于2-8。

ClRog离心20分钟，或将标本放于-3七或-8O c C 保存，但应避免反复冻融，3 .组织匀浆：用预冷的PBS9Ql∙M,pH=7.4)冲洗组织，去除残留血液(匀浆中裂解的红细胞会影响测量结果)，称重后将组织剪碎。

将剪碎的组织与对应体积的PBS(一般按1:9的重量体积比，比如Ig的组织样品对应9M L的PBS,具体体积可根据实验需要适当调整，并做好记录。

推荐在PBS中加入蛋白酶抑制剂)加入玻璃匀浆器中，于冰上充分研磨。

为了进一步裂解组织细胞，可以对匀浆液进行超声破碎，或反熨冻融。

采用网络药理学的方法筛选出黄芪关键活性成分8个，预测获得与2型糖尿病相关靶点204个，筛选出关键靶点35个，包括AKT1、IL6、VEGFA、TP53和EGF等。

黄芪治疗2型糖尿病主要与细胞增殖、分化、凋亡、炎症等生物学过程有关。

黄芪通过与糖尿病并发症有关的AGE-RAGE信号通路、IL-17信号通路和TNF 信号通路等协调发挥直接或间接治疗作用。

Using network pharmacology, 8 effective ingredients of Radix Astragali were obtained, and 204 intersection targets of Radix Astragali and T2D were identified. PPI network mainly involved 35 core target proteins, including AKT1, IL6, VEGFA, TP53 and EGF, respectively. GO functional enrichment analysis showed that the cell proliferation, differentiation, apoptosis and inflammation were mainly involved in T2D therapy.
