仪器介绍⼁赛默飞Q-ExactiveOrbitrap质谱仪经常会遇到这样的问题,'做代谢组学⽤什么仪器⽐较好?','这两台⾼分辨仪器怎么选择?','这台仪器都能做什么?',这些可能是困扰研究者开展代谢组学研究的主要问题,特别是对于⼀些想去从事该⽅⾯研究,且有⼤笔仪器采购经费的实验室。
因为进⾏代谢组学研究有⼀段时间,且⽤过不同品牌的质谱仪,所以近期就根据⾃⼰的使⽤体会做⼀下仪器评价,其实也说不上什么评价,只是⾃⼰的使⽤感受⽽已,希望对⼤家有⽤。
想要开展代谢组学研究,特别是⾮靶向代谢组学研究,需要选择⼀台⾼分辨质谱仪(关于分辨率的解释,可以参看公众号之前⽂章:代谢组学研究中需要了解的质谱知识⼁分辨率),因此这⾥所评价的仪器也均是⾼分辨质谱仪。
今天要说的这台仪器是赛默飞公司的Q-Exactive Orbitrap质谱仪,因为前两天刚有合作实验室进⾏了该仪器的装机和培训,所以就先介绍⼀下这台仪器吧。

另外,在这⾥也仅是仪器在代谢组学应⽤⽅⾯的使⽤体会。
仪器介绍该仪器提供了四种分辨率,以m/z 200为例分别为17500,35000,70000,140000,⽤户可以根据⾃⼰检测的⽬的⾃⾏选择。
扫描⽅法主要包括:1. Full Mass:全扫,扫描范围m/z A~m/z A×152. SIM:选择离⼦监测3. AIF:All Ion Fragmentation,DIA的⼀种形式,将扫描范围内的所有离⼦进⾏碎裂4. dd-MS2:DDA扫描5. PRM:平⾏反应监测,⽤来做定量编辑⽅法时,可以根据⾃⼰的需要对这⼏种扫描⽅法进⾏组合,当然,仪器的⽅法编辑器中也内嵌了很多组合形式,研究者可以根据实验⽬的去选择。
