高效液相色谱法测定甜叶菊中甜菊糖总苷的含量关键词:利用高效液相色谱法对甜叶菊干叶中甜菊糖总苷含量开发出一种新的检测方法,通过单因素试验,确立了检测方法,即样品经30%乙腈超声提取、孔径为0.45μm的有机滤膜过滤后,选择C18(250mm×4.6mm,5μm)色谱柱;以78%的0.1%磷酸水溶液+22%乙腈(体积比)为流动相,1.0mL/min的流速进行洗脱。
在210nm 波长及40℃柱温下利用DAD检测器检测,用外标法定量。
得出的图谱峰形尖锐,结果回收率高、检测结果准确、精密度高、重复性良好。
说明此方法开发具有较强的可行性。

Elution was carried out at a flow rate of 1.0mL/min with 78% 0.1% phosphoric acid aqueous solution +22% acetonitrile (volume ratio) as mobile phase. At 210nm wavelength and 40℃ column temperature, DAD detector was used for detection and external standard method was used for quantification. The obtained chromatogram has sharp peak shape, high recovery rate, accurate test results, pinpoint accuracy and beautiful repeatability. This shows that the development of this method is feasible.Key words:high performance liquid chromatography; Stevia rebaudiana; Stevioside total glycoside1 前言食物是人类得以生存的必备保障,食物的主要成分与构成人体的基本物质相同,人类可从食物中获取人体生长的营养物质及能量。
目的:通过比较不同断面颜色白术中白术内酯的含量,从而确定不同颜色的白术的质量,建立白术的质量标准。
方法:采用外标法,HPLC法测量其中一个含量。
Dikma Kromasil C。
(250 mm×4.6 miD.,5 pro)色谱柱,以乙腈(A)一水(B)为流动相梯度洗脱;体积流量为1.0 mL/min,柱温为35℃,检测波长为220 nm(白术内酯I、III)、276 nm(白术内酯I),进样量10uL。
结果:白术内酯I.II.III分别在1.14~114、0.776"-'77.6、3.21~321 ug/mL与峰面积呈良好的线性关系。
结论:白术颜色白者与颜色棕者,其中白术内酯I.II.III含量差异较大,尤其白术内酯I.III含量白者多于棕者,故而白术白者质量优于棕者。

高效液相色谱法同时测定野菊花中的4种药用成分李婷;区硕俊;王强;黄曼谷【摘要】建立高效液相色谱法(HPLC)测定野菊花中绿原酸、黄芩苷、槲皮素和蒙花苷的含量.样品经过优化条件提取后,梯度洗脱条件下分离,二极管阵列检测器进行检测.4种药用成分在相应的范围内浓度呈良好的线性关系,相关系数(r)在0.9985~0.9998之间.平均回收率为84.4%~97.2,检出限为0.1~0.5 mg/L,相对标准偏差为4.88%~8.21%.该方法回收率高、精密度高、准确度高、重现性好,是测定野菊花中4种药用成分的有效方法.%A high performance liquid chromatography (HPLC) method was established for simultaneous determination of chlorogenic acid, baicalin, quercetin and linarin in Chrysanthemum indicum. Samples were extracted with optimum conditions, then they were separated under the gradient elution conditions and finally detected by photodiode array detector. The results show that, four pharmaceutical components exhibits good liner relationship within the corresponding concentrations, correlation coefficient(r) is from 0.9985 to 0.9998. The average recoveries of four pharmaceutical components are 84.4%~97.2%, the limits of detection(LOD) of the method are 0.1~0.5mg/L and the relative standard deviations (RSDs) are 4.88%~8.21%. 高效液相色谱法同时测定干菊花中五种防腐剂作者:周斌杨怀燕张秀梅等来源:《湖北农业科学》2016年第04期摘要:建立了同时测定干菊花中5种防腐剂的高效液相色谱分析方法,优化后的色谱条件为采用Waters XTerra C18色谱柱(3.9 mm×150 mm,5 μm)分离,流动相为乙酸水溶液(0.1%)/甲醇梯度洗脱,用二极管阵列检测器在波长254 nm处进行检测。
结果表明,5种防腐剂在18 min内可获得较好的分离效果,在0.1~20.0 mg/L范围内线性关系良好(r≥0.999 8),检出限为0.001 5~0.059 0 mg/L,定量限为0.005 0~0.196 8 mg/L,加标回收率为85.9%~97.8%,RSD为0.5%~12.5%。
该方法操作简单、灵敏度高、重复性好,可用于检测干菊花中防腐剂。
