方法 将C57BL/6J小鼠随机分成正常对照组、肥胖模型对照组和SEM干预组。
干预结束后取小鼠血清和脂肪组织,ELISA法测定小鼠血清和脂肪组织中炎症因子、抗炎因子及趋化因子的分泌水平,Westernblot法测定脂肪组织巨噬细胞M1型极化标志物的蛋白表达水平。
以LPS刺激RAW264 7巨噬细胞诱导炎症模型并用SEM进行干预,使用AMPK抑制剂compoundC进行机制探讨和验证,Westernblot法检测巨噬细胞极化标志物及AMPK信号通路相关蛋白表达水平,ELISA法检测巨噬细胞炎症相关因子。

方法 硼替佐米中的异戊醛用 2,4-二硝基苯肼衍生化,采用 Waters symmetry C 18 色谱柱(250 mm × 4.6 mm ,5 µm ),以乙腈(体积分数 0.1% 甲酸)-水(体积分数 0.1% 甲酸)为流动相进行线性梯度洗脱,流速为 1.0 mL•min -1,检测波长为 364 nm ,柱温为 30 ℃。
结果 异戊醛质量浓度在 0.015~2.998 mg•L -1 内与峰面积呈良好线性关系(r = 1.000 0),平均回收率为 101.1%,RSD 为 1.4%。
结论 该方法准确灵敏、线性范围宽,可以用于稳定性过程中潜在基因毒性杂质异戊醛的评估。

方法 以Agilent Eclipse Plus C 18为色谱柱,以乙腈-0.1%磷酸溶液为流动相(梯度洗脱),流速为1 mL/min ,柱温为30 ℃,检测波长为270 nm 。
分别以荭草苷、牡荆苷、荭草素-2″-O-β-L-半乳糖苷为内参物,采用一测多评法建立另外5个待测成分与内参物的相对校正因子,然后计算21批金莲清热颗粒样品中6个成分的含量,并与外标法测定结果进行比较。
结果 一测多评法测得21批金莲清热颗粒样品中芒果苷、荭草素-2″-O-β-L-半乳糖苷、荭草苷、藜芦酸、牡荆苷和哈巴俄苷的含量范围分别为0.234~0.516、1.804~2.270、2.143~2.606、0.190~0.223、0.594~0.782、0.080~0.152 mg/g ,外标法测定结果分别为0.235~0.523、1.798~2.265、2.137~2.599、0.190~0.224、0.597~0.786、0.077~0.151 mg/g ,2种方法所测结果的差异百分比不大于4.00%。
结论 建立了基于多种内参物同时测定金莲清热颗粒中6种成分含量的一测多评法,该方法与外标法所得结果无明显差异,可用于金莲清热颗粒的质量控制。
中国饲料2016年第17期DOI:10.15906/11-2975/s.20161711高效液相色谱紫外检测法测定饲料中N-氨甲酰谷氨酸的含量李裕(湖南省兽药饲料监察所,湖南长沙410006)N-氨甲酰谷氨酸是N-乙酰谷氨酸的稳定结构类似物,能激活动物体内氨甲酰磷酸合成酶-Ⅰ和二氢吡咯-5-羧酸合成酶,从而促进精氨酸的生成。
精氨酸在调控动物机体营养代谢和免疫反应方面有重要作用,可提高多胎动物窝产仔数和初生重、促进幼龄动物生长和发育(Wu等,2004)。
N-氨甲酰谷氨酸的成本仅为精氨酸的1/10(冯涛等,2012)。
目前含有N-氨甲酰谷氨酸的饲料产品主要有饲料添加剂和配合饲料两大类。
目前报道的检测饲料中N-氨甲酰谷氨酸的方法有高效液相色谱电喷雾串联质谱法(梁婧等,2015;唐圣芸等,2014)。
相比常见的高效液相色谱紫外检测法,串联质谱法对色谱条件、样品净化容忍度较高,而且灵敏度高、定性更准确,常用于复杂基质中极痕量物质的检测。
但串联质谱仪器昂贵,运行和维护成本也较高,大部分饲料企业尚未配备。
而且对于含量较高样品的检测,由于串联质谱上机的稀释倍数大、离子源的离子化效率及稳定性等因素,定量可能不如高效液相色谱紫外检测法准确。
因此,本实验旨在建立一种适用于大多数饲料企业实验室使用,能准确测定饲料中N-氨甲酰谷氨酸含量的方法。
1材料与方法1.1仪器Waters2695高效液相色谱仪配2489型紫外检测器、2998型二极管阵列检测器(美国Waters公司);Delta320型pH计(梅特勒-托利多公司);OASIS MAX固相萃取柱(150mg,6cc,美国Waters公司)。
1.2试剂N-氨甲酰谷氨酸(纯度≥98%,Sigma 公司);甲醇(色谱纯,德国Merck公司);磷酸(优级纯,国药集团化学试剂有限公司)。
N-氨甲酰谷氨酸标准储备液:准确称取N-氨甲酰谷氨酸对照品102mg置于100mL容量瓶中,用水溶解,定容至刻度,摇匀。

基于HITEMP数据库的分子吸收光谱高精度快速建模方法钱宝健,蔡静*,常海涛,高一凡(航空工业北京长城计量测试技术研究所,北京 100095)摘要:为解决高温环境下分子吸收光谱精确计算的时间复杂性,满足宽光谱测量领域对理论吸收光谱计算的需求,本研究利用Python语言以逐线计算为基础,结合线型函数的简化、线翼截止准则和谱线数据库的优化,建立了基于高温分子吸收参数数据库(High⁃Temperature molecular spectroscopic absorption parameters data⁃base,HITEMP)的分子吸收光谱精确快速计算模型。
以Hartmann⁃Tran线型函数作为吸收光谱标准线型编写部分相关二次速度依赖硬碰撞函数(partially⁃Correlated quadratic⁃Speed⁃Dependent Hard⁃Collision Profile,pCqSDHC),结合复概率函数(Complex Probability Function,CPF)简化模型实现了线型函数的精确快速计算,相较于理论计算模型计算速度提高了20倍。
按照光谱计算残差在10-5量级确定了固定波数截断结合谱线半宽等倍数截断的线翼截止准则。
以阈值线强度10-25 cm-1/(mol∙cm-2)为标准筛选了每100 K温度梯度时的光谱数据,整合得到优化数据库。
在6 500 ~ 8 000 cm-1范围内对水分子的吸收光谱进行计算,并与“”分子气体集成光谱建模网站仿真结果对比,逐线模型的计算误差在10-7量级,优化模型的计算误差在10-5量级,计算速度平均提升25倍。
该模型满足吸收光谱测量中对于理论吸收光谱的高效准确计算,为复杂环境中基于宽调谐、超连续激光吸收光谱的测量研究提供了理论模型基础。

The line wing truncation criteria were determined based on the spectral calculation residual at the level of 10-5 and involved the truncation of fixed wavenumbers combined with equal multiple truncations of spectral line half widths. Spectral data for each temperature gradient of 100 K were selected using a threshold line intensity of 10-25 cm-1/(mol∙cm-2) and integrated to create an opti⁃mized database. The absorption spectra of water molecules were calculated within the range of 6 500 ~ 8 000 cm-1 and compared with the simulation results from "", a molecular gas integrated spectral modeling website. The calculation error of the line⁃by⁃line model was at the level of 10-7, while the optimized model achieved a calculation error at the level of 10-5, with an average speed improvement of 25 times.
