方法:以提取A549肺癌细胞和BEAS-2B正常人肺上皮细胞上清液制备条件培养基,培养的hBMSCs分别为A549组和BEAS-2B组;2组均添50 nmol/L趋化因子-8(CXCL-8),分别为A549 C组和BEAS-2B C组,若同时添加100 nmol/L triciribine (Akt抑制剂)分别为A549 CT组和BEAS-2B CT组;A549组加入100 nmol/L triciribine为A549 T组。
正常条件下培养的hBMSCs为对照组。
ELISA检测细胞上清液中CXCL-8含量和hBMSCs裂解液Akt、P-Akt蛋白的表达。
MTT实验、流式细胞仪和transwell细胞小室实验分别检测各组hBMSCs吸光度值(A490值)、细胞凋亡率和细胞迁移数目。
结果:与BEAS-2B上清液相比,A549上清液中CXCL-8含量明显升高。
各组hBMSCs的A490值、细胞凋亡率、细胞迁移数目、Akt和P-Akt蛋白表达等均具有显著差异,尤以A549 C组hBMSCs最明显。
与A549 C 组相比,A549 CT组hBMSCs的A490值和细胞迁移数目、Akt和P-Akt蛋白表达均降低,细胞凋亡率升高较显著。
结论:A549细胞表达的CXCL-8能激活Akt通路,促进hBMSCs增殖和运动。

Methods: The supernatants of A549 cells and BEAS-2B cells (the normal human bronchial epithelial cell line)were extracted. hBMSCs cultured with the supernatants were called A549 group and BEAS-2B group respectively; 50 nmol/L chemokine-8 (CXCL-8)was added to both groups to form A549 C group and BEAS-2B C group; 100 nmol/L triciribine (Akt inhibitor) was added at the same time to form A549 CT group and BEAS-2B CT group; 100 nmol/L triciribine was added to A549 group to form A549 T group. hBMSCs incubated in the conventional condition were served as the control group. The CXCL-8 in the cell culture supernatants and the expression of Akt and P-Akt in hBMSCs lysates were tested by ELISA. MTT assay, flow cytometry and transwell assay were used to detect the absorbance (A490), apoptosis rate and the number of migrating cells in each group respectively. Results: Compared with the BEAS-2B supernatant, the CXCL-8 in the A549 supernatant was significantly increased. There were significant differences in A490, apoptosis rate, the number of migrating cells, and Akt and P-Akt expressions of hBMSCs in each group, especially in A549 C group. Compared with the A549 C group, the A490 and the number of migrating cells, Akt and P-Akt expression of hBMSCs in the A549 CT group were reduced, but the apoptosis rate was increased significantly. 方法 将人神经胶质瘤细胞系U 87随机分为对照组,低、中、高质量浓度SM 组(50、100、200 μg/mL ),蛋白激酶B (又称Akt )激活剂组(SC 79 20 μmol/L ),高质量浓度SM 联合Akt 激活剂组(SM 200 μg/mL+SC 79 20 μmol/L )。
经药物(对照组除外)处理后,检测各组细胞的光密度(OD )值、克隆形成率、凋亡率、增殖/凋亡相关蛋白[增殖细胞核抗原(PCNA )、B 细胞淋巴瘤2(Bcl-2)、Bcl-2相关X 蛋白(Bax )、胱天蛋白酶3(caspase-3)]的表达情况和Akt/促分裂原活化的蛋白激酶(MAPK )信号通路相关蛋白[Akt 、p 38 MAPK 、胞外信号调节激酶1/2(ERK 1/2)]的磷酸化水平。
于右侧腋窝内皮下注射U 87细胞建立异种移植肿瘤裸鼠模型并将其分为对照组,低、中、高剂量SM 组(25、50、100 mg/kg ),Akt 激活剂组(SC 79 40 mg/kg )、高剂量SM 联合Akt 激活剂组(SM 100 mg/kg+SC 79 40 mg/kg ),每组5只。
经药物(裸鼠对照组除外)干预后,称定其瘤体质量并计算瘤体体积。
结果 与对照组比较,SM 各质量浓度组细胞的OD 值,克隆形成率,PCNA 、Bcl-2蛋白的表达水平,Akt 、p 38 MAPK 、ERK 1/2蛋白的磷酸化水平,以及裸鼠SM 各剂量组裸鼠体内的瘤体质量及体积均显著降低/减小,细胞凋亡率和Bax 、caspase-3蛋白的表达水平均显著升高,且均有剂量依赖性(P <0.05);Akt 激活剂组上述指标的变化趋势与之相反(P <0.05),且Akt 激活剂可明显减弱高质量浓度/高剂量SM 对神经胶质瘤的体内外抑制作用(P <0.05)。

PCR产物鉴定:1)正常肾上腺髓质和嗜铬细胞瘤组织均可见ADM(410bp)RAMP2(283bp)、CRLR(497bp)的PCR产物(图2—2)。
2)ADM、RAMP2、CRLR的PCR产物提纯后经自动测序仪双向测序,与GenBank登记的序列同源性达95%,排除PCR反应错配、测序错误及种属内同源性差异,可认为扩增片段为人ADM、RAMP2、CRLR的cDNA(图2—3、2—4、2-5)。

米诺环素抑制促炎因子生成保护多巴胺能神经元汪锡金;颜志强;谷胜利;叶民;刘振国【摘要】Objective To investigate the effect of minocycline on inflammation-induced dopaminergic neurodegeneration. Methods SD rat model of inflammation-induced dopaminergic neurodegeneration was established in mesencephalic neuron/ glia cultures by lipopolysaccharides. The culture system was treated by minocycline before or after the addition of lipopolysaucharides. The number of dopaminergic neurons was determined by immunocytochemistry and cell counting. Cell bioactivity was detected by liquid scintillation uptake assay. Besides, the changes of key proinflammatory factors such as tumor necrosis factor-α (TNF-α) (by ELISA), interleukin-1β (IL-1β) ( by ELISA), nitric oxide ( NO) ( by Griess method) and superoxide (by cytochrome C reduction method) were examined in the cultures. Results Minocycline (2-10 (μmol/L) pretreatment dose-dependently attenuated the reduction in dopamine uptake and dopaminergic cell number in mesencephalic cultures treated with lipopolysaccharides ( 1 - 100 ng/mL). Minocycline (10 μmol/L) posttreatment also protected dopaminergic neurons from lipopolysaccharides (10 ng/mL) -induced neurodegeneration inneuron/glia cultures. Minocycline pretreatment and posttreatment significantly inhibited the production of TNF-a, IL-1β. NO and superoxide in neuron/glia cultures. AbstractUptopresent,alargenumberofreportsunveiledexacerbatingeffectsofbothlong-andshort-termadministrationofmorphine,asapotentanalgesicagent,onopium-addictedindividualsandaplethoraofcellkinetics,althoughcontradictoryeffectofmorphineondifferentcellshavebeenintroduceduntilyet.Toaddressthepotentmodulatoryeffectofmorphineonneuralmultipotentpre-cursorswithemphasisonendogenoussex-relatedneuros-teroidsbiosynthesis,weprimedtheratneuralstemcellsisolatedfromembryonicrattelencephalontovariouscon-centrationsofmorphineincluding10,20,50and100lMaloneorincombinationwithnaloxone(100lM)overperiodof72h.FlowcytometricKi-67expressionandAnnexin-V/PIbasednecrosisandapoptosisofexposedcellswereevaluated.Thetotalcontentofdihydrotestos-teroneandestradiolincellsupernatantwasmeasuredbyELISA.Accordingonobtaineddata,bothconcentration-andtime-dependentdecrementofcellviabilitywereorchestratedthoroughdown-regulationofki-67andsimultaneousup-regulationofAnnexin-V.Ontheotherhand,theadditionofnaloxone(100lM),asMuopiatereceptorantagonist,couldbluntthemorphine-inducedadverseeffects.Italsowellestablishedthattime-course
exposureofratneuralstemcellswithmorphinepotentlycouldacceleratetheendogenousdihydrotestosteroneandestradiolbiosynthesis.Interestingly,naloxonecouldcon-sequentlyattenuatetheenhancedneurosteroidogenesistime-dependently.Itseemsthatourresultsdiscoverabiochemicallinkagebetweenanacceleratedsynthesisofsex-relatedsteroidsandratneuralstemcellsviability.

IntroductionPain,anunpleasantfeelingsense,isinducedbydifferentsurroundingstimuli[1].Whentreatmentbyroutinenon-opioidanalgesicandphysicalmethodsisnotresponding,opioidtreatmentisconsideredanalternativestrategywhileitsmedicationhadlonghistoricalbackground[2,3].Generally,opioidsapplicationhasbeendocumentedforsedatingofseverepains[4].Inspiteofrobustanalgesicproperties,awiderangeofsideeffectsinindividualswithshort-orlong-termaddictionhavealsobeendocumented[5].Forexample,constipation,nausea,urinaryretention,cognitiveimpairment,cardiovascularsysteminvolvementandetc.areexemplifiedinpeoplewithopiates[6–8].Inadditiontoclinicaladverseeffects,anarrayofbehavioraldisordersandtheinhibitoryeffectofmorphineontissue,cellularandevenmolecularlevelswasalsodetermined[9].Forinstance,neuronsize,outgrowthpattern,neuritearrangementand—inparticular—neurogenesisareagitatedbystructuralchangesofneuronsandrelatedsynapses[10].Aspreviouslyhighlighted,fourclassicaldifferentopioid,G-proteincoupledreceptors,namelyMu(l),Kappa(j),delta(d)andopioidreceptorlike-1(ORL1),havebeen

NeurochemResDOI10.1007/s11064-016-1847-7showntoplaykeyroleinpathophysiologyofopiates[11].Ofnote,anactivationstatusofaforementionedreceptorsbyopiateagonistsresultsinGaandGbcsubunitswhichinturncontributetodiminishcyclicadenosinemonophos-phateproductionandeventuallymodulationofcalciumandpotassiumionchannels[12].Basedontakendata,cellularhyperpolarizationandtonicneuralactivityinhibitionwerefollowedbyover-activationofmorphinecounterreceptors[12].Adultmammalianneuralstemcells(NSCs),anundif-ferentiatednervousprogenitorswithproliferationandself-renewalability,resideinspecificareasofcentralnervoussystem(CNS)consistingofsubventricularzone(SVZ)oflateralventricalandhippocampaldentategyrusregions[13–15].Agreatbodyofnovelexperimentsdecipheredtheeffectiveimpactofvariousfactorsandhormones,notice-ablytestosterone,estrogen,prolactin,corticosteroids/adre-nalstressstroidsdehydroepiandrosterone,pregnenolonederivativessuchaspregnenolone-sulfate,allopregnenoloneandprogesteroneoncellkineticsofNSCsmitosisandtargetedproliferation[16].Someauthorspreviouslyacclaimedanactivepartici-pationoftestosteroneand17b-estradiol(E2),assex-relatedandrogens,ininductionofneuriteandhippocampalgrowth,development,andneurogenesiscapabilityofNSCs[17–19].Abiologicallyactiveformoftestosteronemetabolite,dihydrotestosterone(DHT)isproducedby5a-reductaseenzymeactivity[20].However,observationinthemulti-bodysettingofexperimentsrevealedalargediscrepanciesrelatingtoDHTactiononstructureandfunctionofnervoussystem[21–23].Inhippocampalneu-rons,testosteroneanditsderivative,dihydrotestosterone,playaroleviaandrogenreceptorwhichevokesintracel-lularmitogen-activatedproteinkinase/extracellularsignal-regulatedkinase(MAPK/ERK)signalingpathway.DHTalsocouldactivaterelevantdownstreamofMAPK/ERK,cyclicAMPresponseelementbindingproteinSREBandpeculiarlyproteinkinaseCsignaling[18].Inaddition,E2couldimpedeexcitotoxicityandoxidativeinsultsinflictedbymanyexcessiveneurotransmittersandharmfulstimuli[24].Itseemsthatfunctionalandphysiopathologicalimplicationsregardingtosurroundingnicheshavebeenilluminatedpostmorphineadministration[24].Goodmanetal.conceivedmorphine,indosedependentmanner,couldinitiateneuropathysimultaneouslybydiminishingthetotallevelofintracellulartestosteronecontent,sensi-tizingcellstoglucosedeprivation[24].Theaimofcurrentexperimentistoscrutinizetheadverseeffectofmorphinesulfatewith/withoutnaloxoneonratNSCsDHTandE2synthesisproperties.
