酶是广泛分布于生命有机体的组织细胞内具有催化活性的特殊蛋白质，参与体内几乎所有的细胞功能活动进程。

有些酶类还是不同亚细胞器的标志酶，如酸性磷酸酶是溶酶体的标志酶，过氧化氢酶是过氧化物酶体的标志酶，葡萄糖-6-磷酸酶是内质网的标志酶，氨肽酶是微粒体的标志酶，琥珀酸脱氢酶和细胞色素氧化酶是线粒体的标志酶[1–2]。

目前，由国际生物化学酶学委员会(Enzyme Commission, EC)认定并注册登记编号的酶约有2000多种。

按照酶对底物的特异性、催化性和来源，酶的命名可分为6大类：氧化还原酶(Oxidoreductase, EC 1)、转移酶(Transferase, EC 2)、水解酶(Hydrolase, EC 3)、裂解酶(Lyase ，EC 4)、异构酶(Isomerase, EC 5)和合成酶(Synthetase, EC 6), 这些酶中已知能被现有的组织化学和细胞化学技术原位检测显示的酶仅有200多种[3]。

在保持活组织和细胞结构完整性的条件下，利用组织化学、细胞化学、免疫学和显微检测等技术研究酶的即位定位，是了解酶在组织、细胞和亚细胞中的分布、活性动态与定量及酶功能等的重要途径。

对植物体中酶定位的组织化学和细胞化学方法的概念、原理与研究进展进行了综述，并根据国际酶化学分类编号顺序，分别介绍了25种酶的组织化学染色定位所用的反应介质和染色方法及46种酶的细胞化学定位方法的参考文献。

α-亚麻酸的提取和分离方法作者：柏薇薇来源：《食品界》2018年第04期自 1978年Dyerberg发表的爱斯基摩人的饮食与心血管疾病的调查报告以来，国外人们就对ω- 3系列脂肪酸产生了兴趣，对ω- 3系不饱和脂肪酸（α-亚麻酸、EPA、DHA等）的代谢和生理作用研究日益深入，对其研究迅猛发展。

80年代初期就有多篇论文发表。

特别是它们在降血脂、降血压、抗血栓、抗动脉粥样硬化、乃至提高机体免疫力和抗癌等方面的药用价值得到充分肯定。

α-亚麻酸的传统提取方法为冷榨法和溶剂法。

目前常见的方法有：脂肪酸金属盐法、低温结晶法、柱层析色谱法、银离子络合法、尿素包合法、分子真空蒸馏法和超临界流体萃取法、脂肪酶催化水解法等。

硝酸银柱层析法是将硅胶加入浓硝酸银溶液中处理，用来分离碳原子数相等而其中C- C双键数目不等的一系列化合物，如不饱和醇、酸等。

其主要机理是由于不不饱的C- C键能与由于吸附在硅胶上的银离子形成络合物，而饱和的C- C键则不与硝酸银络合。

双键数较多，结合Ag+就越多。

因此在硝酸银薄层上，化合物可由于饱和程度不同而获得分离。

硝酸银柱层析法的优点：对设备要求不是很高，此法分离效果好，可获得较高纯度的α-亚麻酸制品，适宜实验室操作，α-亚麻酸不易发生氧化、聚合等现象。

其缺点：硝酸银价格贵，成本高，制备量少，需要大量溶剂且容易造成污染。

难于进行大规模生产，操作较为繁杂。

目前用此方法进行提取，纯度可达97%。

脂肪酸金属盐法主要是利用饱合度不同的脂肪酸金属盐在溶剂中的溶解度的差异来进行分离，可将长链、多不饱和脂肪酸与短链脂肪酸和饱和及低不饱和脂肪酸分离，达到提纯α-亚麻酸的目的。

应用较多的是锂盐和钠盐。

低温结晶分离法是利用低温下不同脂肪酸在有机溶剂中溶解度和熔点差异来分离纯化，通常脂肪酸在有机溶剂中的溶解度随碳链长度的增加而减小，随不饱和度的增加而增加，随着温度降低这种溶解度的差异表现得更为显著。

缺点是需要回收大量溶剂。
