Abstract: Rhinogobio cylindricus is an important commercial fish and spawns drifting eggs in the Yangtze River������ Dam construc ̄ tion and overfishing may seriously threaten the resource������ In this studyꎬ mitochondrial DNA Cyt b gene sequence in R������ cylindricus was used to assess the level of genetic diversity and structure of R������ cylindricus populations from seven stations in the Yangtze Riv ̄ er as Yibinꎬ Luzhouꎬ Hejiangꎬ Jiangjinꎬ Bananꎬ Fuling and Huanggang������ The results showed that 14 variation sites were found from 998 bp size of datasetꎬ and a total of 15 unique haplotypes were defined from 140 individuals������ The average haplotype diversi ̄ ty(Hd)and nucleotide diversity(Pi)were 0������ 625 and 0������ 000 77ꎬ respectivelyꎬ which indicated moderately low genetic diversity for R������ cylindricus population������ The analysis of molecular variation(AMOVA)revealed no significant genetic divergence occurred among sampled stations with Fst = - 0������ 009 98������ The haplotype network analysis showed a star ̄like topology������ Phylogenetic(NJ)tree showed low support values among branches of haplotypeꎬ which suggested that there was little genetic divergence within spe ̄ cies������ Neutrality testsꎬ mismatch distribution and BSP analysis all indicated that R������ cylindricus had experienced population history expansion event and the expansion time was estimated in about 0������ 06 Ma(million years before)������ In summaryꎬ the present genetic diversity of R������ cylindricus population was low and no genetic structure was illuminated������ We further suggest that R������ cylindricus popu ̄ lation can be protected in situꎬ and a long term monitoring item should be carried out������ Key words: Rhinogobio cylindricusꎻ mitochondrial Cyt b sequenceꎻ genetic diversityꎻ middle and upper reach of the Yangtze River

长江中上游圆筒吻鮈群体线粒体 Cyt b 遗传多样性分析

摘要: 为探明大坝建设及过度捕捞等可能会对圆筒吻鮈资源造成的影响ꎬ 采用线粒体 DNA Cyt b 基因序列对长江 中上游宜宾、 泸州、 合江、 江津、 巴南、 涪陵和黄冈 7 个圆筒吻鮈( Rhinogobio cylindricus) 群体进行研究ꎬ 分析其 遗传多样性及遗传结构ꎮ 结果表明: 在获得的 998 bp 序列长度中检测到 14 个变异位点ꎬ 140 尾样本定义 15 个单 倍型ꎬ 平均单倍型多样性( Hd) 和平均核苷酸多样性( Pi) 分别为 0������ 625、 0������ 000 77ꎬ 说明圆筒吻鮈种群遗传多样性 较贫乏ꎮ 分子变异方差分析( AMOVA) 表明圆筒吻鮈地理群体间未出现显著遗传分化ꎮ 单倍型 Network 网络分析 呈星型结构ꎬ 邻接系统发育( NJ) 树显示单倍型没有出现高支持率的分支ꎬ 表明群体内部没有发生遗传分化ꎮ 中 性检验、 错配分布及 BSP 分析均表明圆筒吻鮈发生历史种群扩张事件ꎬ 种群扩张发生在 0������ 06 Ma( 百万年前) ꎮ 综上表明ꎬ 圆筒吻鮈的群体遗传多样性较低ꎬ 群体间遗传分化不显著ꎬ 建议将圆筒吻鮈群体作为一个整体就地 保护ꎬ 并在今后应加强长期监测ꎮ 关键词: 圆筒吻鮈( Rhinogobio cylindricus) ꎻ 线粒体 Cyt bꎻ 遗传多样性ꎻ 长江中上游 中国分类号: S917������ 4 文献标识码: A 文章编号: 1000 ̄6907 ̄(2019)01 ̄0014 ̄06
