茶多糖对小鼠肠道健康及免疫调节功能的影响李海珊,刘丽乔,聂少平*(南昌大学食品科学与技术国家重点实验室,江西南昌 330047)摘 要:本实验以从江西婺源绿茶中提取的茶多糖(tea polysaccharides,TPS)为研究材料,研究不同剂量TPS对正常小鼠肠道健康及免疫调节功能的影响。
实验小鼠随机分为正常组和低、中、高剂量TPS干预组。
TPS干预21 d 后,收集小鼠腹腔巨噬细胞,进行中性红吞噬实验检测腹腔巨噬细胞的吞噬能力,解剖获取小鼠脾脏、胸腺后称其质量并计算脾指数和胸腺指数。
气相色谱法检测小鼠结肠内容物中短链脂肪酸(short chain fatty acids,SCFAs)的含量;靛酚蓝比色法测定结肠内容物中氨含量;pH计检测结肠内容物pH值;恒质量法检测结肠内容物含水量。
结果显示:高、中、低剂量TPS干预组的小鼠免疫器官指数均显著增大,腹腔巨噬细胞吞噬能力呈剂量依赖性增强,结肠内容物的乙酸,丙酸和正丁酸的含量升高,而异丁酸、正戊酸、异戊酸的含量无明显变化;结肠内容物中氨含量下降,结肠内容物pH值下降,含水量升高。
实验结果提示TPS具免疫调节功能及促进小鼠肠道健康的作用。
关键词:茶多糖;免疫调节;肠道健康Effects of Green Tea Polysaccharides on Intestinal Health and Immune Regulation in MiceLI Haishan, LIU Liqiao, NIE Shaoping*(State Key Laboratory of Food Science and Technology, Nanchang University, Nanchang 330047, China) Abstract: The objective of this work was to evaluate the effect of tea polysaccharides (TPS, from Wuyuan green tea) on intestinal health and immune regulation in mice. Healthy C57/BL6 mice were randomly divided into four groups, and they were administrated by gavage with different concentrations of tea polysaccharides. After adaptive feeding for one week, the animals in these groups were given high, mediumand low doses of TPS and normal saline as blank control for 21 days, respectively. At the end of the experimental period, the mice were sacrificed, and spleen and thymus were harvested and weighted to calculate immune organ indexes. Peritoneal macrophages were used to examine phagocytosis activity by using neutral red phagocytosis assay. Short chain fatty acids (SCFAs) in colonic contents were analyzed by gas chromatography (GC) and ammonia by the indophenol blue colorimetric method. Meanwhile, the pH and water content of colonic contents were also measured. Results showed that immune organ indexes were increased significantly after TPS administration. The phagocytosis of peritoneal macrophages was improved in a dose-dependent manner. Some SCFAs (acetic acid, propionic acid and butyric acid) were increased while no difference in isobutyrate, pentanoic acid and delphinic acid was detected. 普洱熟茶茶多糖对ApoE—/—小鼠动脉粥样硬化的作用研究目的:探讨普洱熟茶提取物茶多糖对ApoE-/-小鼠高脂血症和动脉粥样硬化的作用。
方法:24只ApoE-/-小鼠随机分为ApoE-/-对照组、茶多糖组、洛伐他汀组,另设8只相同遗传背景的C57BL/6J 小鼠为ApoE+/+对照组。
4组分别给予蒸馏水、茶多糖、洛伐他汀、蒸馏水12周,实验结束后检测血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)的含量;ELISA检测血清白介素-6(IL-6)及脂联素(APN)的含量;光镜观测主动脉病理形态学变化。
结果:与ApoE-/-对照组相比,茶多糖组体重明显减轻(P<0.05);TG、TC和LDL-C明显降低,HDL-C明显升高(P<0.05);血清IL-6水平降低,APN水平升高(P<0.05);光镜下主动脉损伤病变程度减轻。
结论:普洱熟茶茶多糖有调节ApoE-/-小鼠血脂、保护血管内皮抗动脉粥样硬化的作用,作用机制可能与其抗炎作用有关。
标签:ApoE-/-小鼠;动脉粥样硬化;茶多糖;白介素-6;脂联素Abstract:Objective To explore the effect of tea polysaccharides(TPS)from fermented Puer tea on atherosclerosis (AS)in ApoE-/- mice.Methods ApoE-/- mice were divided into 3 groups (n=8):ApoE-/- control group,TPS group and lovastatin group. The mice of C57BL/ 6J were used as the ApoE+/+ control group (n=8). The 4 groups of mice were administered with distilled water,TPS or lovastatin for 12 weeks. Triglyceride (TG),total cholesterol(TC),high density lipoprotein cholesterol (HDL-C)and low density lipoprotein cholesterol (LDL-C)levels were assayed. The concertrations of IL-6 and APN in serum were measured by ELISA. Pathomorphology changes of aortas were observated. Results The comparison between TPS group and ApoE-/- control group revealed that:Bodyweights were reduced obviously in TPS group(P<0.05). Serum levels of TG,TC and LDL-C were decreased significantly in TPS group(P<0.05).The contents of IL-6 were lowered markedly,and the level of APN increased obviously in TPS group(P<0.05). The aortas pathological changes were relieved obviously in TPS group. Conclusions TPS from fermented Puer tea can regulate blood lipid levels and bodyweights of ApoE-/- mice.TPS can protect vascular endothelial cells to inhibit the development of atherosclerosis,and this mechanism may be related with anti-inflammation.Keywords:ApoE-/- Mice;Atherosclerosis;Tea Polysaccharides;IL-6;APN動脉粥样硬化(Atherosclerosis,AS)是心血管疾病的主要病理基础,是与血脂代谢异常、内皮受损、平滑肌细胞增殖和血栓形成等有关的慢性炎症性疾病[1-2]。
新藏茶新藏茶是一种高温深度发酵茶,发酵温度在70-80摄氏度之间,俗称乌茶。
一、概述新藏茶是根据现代茶叶科学对茶多糖和茶色素的研究成果,对传统宫廷藏茶工艺进行优化,采用1芽2、3、4、5叶为原料,将制茶重点放到茶多糖、茶色素(茶褐素、茶红素、茶黄色)和茶类芳香物质的转化上,经过鲜嫩叶拼配、杀青、揉捻、烘干、多次高温渥堆发酵、二次烘干、筛分、二次拼配、复试秘制等特殊工序,制成具有“纯和迷甜、圆润厚滑”口感特征;汤色中茶褐素色、茶红素色、茶黄素色明亮清晰,品饮时茶多糖微甜持久;新藏茶香味混合丰富,芳香醇、芳香酶作用明显,具有较高的营养功能价值。
长期饮用对人体血液系统、消化系统和免疫力系统健康有很好帮助。
新藏茶是相对于老藏茶制茶工艺而言的,与老藏茶存在较大区别,具体在阅读本文第五节《老藏茶与新藏茶的区别》。
二、发展简史【藏茶起源】藏茶出现在唐朝,其前身是雅安高温紧压发酵茶,藏茶因文成公主入藏而起。
公元641年,文成公主16岁时入藏与松赞干布和亲,带去了全国各地的贡茶,经过长期饮用实践,发现只有雅安高温发酵茶最适应于生活在青藏高原上的藏族人,到文成公主50岁去世时,整个藏区已经养成煮饮雅安高温发酵茶的习惯,逐渐成为了生活在青藏高原地区藏族人的生命茶,到了“宁可三日无米,不能一日无茶,一日无茶则滞,三日无米则病”的地步,至今已经持续了1373年,从未改变。
千百年来,由于西藏的气候土壤不适合茶叶的生长,西藏不产茶,藏茶也不是西藏生产的茶,而是雅安生产的高温发酵茶。
紧压发酵茶始于唐朝,当时茶风盛行,上至帝王将相,下至普通百姓,无不品茶论道,因此就带来了全国制茶水平的空前繁荣,龙团凤饼就是其代表茶品,还有不同形式的紧压砖茶和条茶。
唐朝紧压发酵茶工艺各异,发酵温度不尽相同,大体可分为低温发酵、中温发酵和高温发酵。
黑茶属于低温发酵,温度在30°~40°之间;红茶发酵温度在50°~60°之间;雅安发酵茶属于高温发酵茶,发酵温度在70°~80°之间。
茶多糖对链脲菌素诱导高血糖小鼠血糖血脂的影响张黎杨艳【摘要】目的研究茶多糖对链脲菌素(STZ )诱导高血糖小鼠血糖血脂的影响,初步探讨茶多糖降血糖的作用机制。
方法采用链尿菌素诱导高血糖小鼠模型,给药至第14d 、28d 测定其空腹血糖;给药至28d ,测定血清胆固醇与甘油三酯,取肝脏,测定肝糖原含量。
结果茶多糖能显著性降低给药第14d 、28d 空腹血糖及血清总胆固醇、甘油三酯含量,提高肝糖原含量。
结论茶多糖对链脲菌素诱导的高血糖小鼠具有一定的降血糖降血脂的作用,其可能的作用机制是增加肝糖原含量有一定的关系。

结果表明:腹腔注射50m g kg ・bw 茶多糖有明显降低血糖的作用(P <0101);对血清胆固醇和血清甘油三酯影响不大,但有提高高密度脂蛋白胆固醇的效果;每天连续口服25m g kg ・bw 茶多糖,有增强免疫功能的作用。
急性毒性试验表明,24h 内腹腔注射总量110g kg ・bw ,未发现小鼠对茶多糖有中毒反应。
