α-甲基-辅酶A-消旋酶 (P504s) 2001年Jiang等[12]首次将P504s抗体应用于前列腺癌的诊断。P504s即α-甲基-辅酶A-消旋酶((alpha-methylacyl-CoA racemase)，其基因定位于染色体5P13，它所编码的蛋白由382个氨基酸组成，在支链脂肪酸的β-氧化和衍生过程中起作用。P504s在前列腺癌中高表达，而在正常中前列腺组织中则呈阴性或灶性弱阳性。Jiang等在所检测的137例前列腺癌中阳性率高达100%，并提出P504s是前列腺癌高度敏感性和特异性的标记物。
第二节 染色方法
本科标抗体法 酶标抗体技术是通过共价键将酶连结在抗体上，制成酶标抗体，再借酶对底物的特异催化作用，生成有色的不溶性产物或具有一定电子密度的颗粒，于光镜或电镜下进行细胞表面及细胞内各种抗原成分的定位。
酶的种类及特点 从理论上讲，用细胞化学方法能显示的酶，均可用于标记抗体，进行ICC染色，但实际上在ICC中所能用的酶并不多。现将常用的几种酶列于表4-1，供选用时参考。
Cumming（1980）报告，不固定的组织切片ICC染色时，环鸟苷酸含量丢失80%以上。固定的目的是使构成组织细胞成分的蛋白等物质不溶于水和有机溶剂，并迅速使组织细胞中各种酶降解、失活，防止组织自溶和抗原弥散，保持组织细胞的完整性和所要检测物质的抗原性。因此迅速充分固定是ICC染色成功的关键一步。用于ICC的固定剂种类较多，选择时应根据所要检测物质的抗原性质和切片方法以及所用抗体特征等做最佳筛选。制片及固定方法见第一章 ，下面介绍两种近年报道的新方法。
