HPLC测定罗布白麻叶中的芦丁和槲皮素

【摘要】目的建立测定罗布白麻叶中芦丁和槲皮素含量的方法。
方法采用HPLC 法,色谱柱为ZorbaxSB-C18(250mm×4.6mm,5μm),流动相为甲醇-0.04%磷酸(40:60),流速为1.0ml·min-1,检测波长为360nm,柱温为35℃。
结果芦丁和槲皮素的线性范围分别为0.01~0.20μg和0.007~0.099μg;r分别为0.9999和0.9998;平均回收率分别为102.1%(RSD=2.0%,n=6)和99.5%(RSD=1.4%,n=6)。
结论所建方法快速、简便、准确、重复性好,可作为罗布白麻叶药材质量控制的有效方法。

不同产地不同采收期中仙鹤草中槲皮素成分含量的分析比较研究目的:应用HPLC法对不同产地不同采收期的仙鹤草药材中槲皮素含量进行分析比较。
方法:采用HPLC法,色谱柱:Thermo BDS Hypersil-C18柱(Part No:28105-254630;4.6 mm × 250 mm,5 μm ),流动相为乙腈-甲醇-0.2%磷酸,检测波长为368nm,柱温30℃,流速1.0ml/min。
结果:槲皮素在0.108~2.160μg (R2=0.9998)范围内呈良好线性关系;平均加样回收率为98.82% (RSD=0.81);不同采收期的仙鹤草药材中槲皮素的含量差别较大,其中八月份采收的样品中槲皮素含量最高;不同产地的仙鹤草药材中槲皮素含量有明显差别,其中以浙江湖州产含量最高。
结论:仙鹤草药材采收时间的不同,其中的槲皮素成分会发生很大变化,八月份为仙鹤草药材的最佳采收期;不同产地仙鹤草中槲皮素含量差别很大,说明仙鹤草有效成分槲皮素受地域的影响很大;该方法精密度高、重复性好、稳定性好,能够准确测定仙鹤草中槲皮素成分的含量,结果表明不同产地的仙鹤草差别较大,为仙鹤草的质量控制提供依据。

Method:HPLC conditions included Thermo BDS Hypersil-C18 column (Part No:28105-254630;4.6 mm ×250 mm,5 μm)and the mobile phase was a mixture of acetonitrile,methanol and 0.2% phosphoric acid for gradient elution. The wavelength was 368nm and the column temperature was at 30℃. The flow rate was 1.0 ml/min. Results:Quercetin had good linear relationship in the range of 0.108~2.160μg (R2=0.9998),its average recoveries was 98.82% (RSD=0.81%);Contents of quercetin in agrimoniae from different areas and different harvest time were of much difference,while agrimoniae from Huzhou of Zhejiang and harvesting at August had higher content. Conclusion:Along with the change of harvesting time and places of produce,the contents of quercetin in agrimoniae were much different. The best harvest time was August and the better place for planting agrimoniae was Huzhou,Zhejiang Province. This method was steady with high precision and good repeatability. 方法:采用Waters CORTECS T3型色谱柱(100mmx2.8mm, 1.7pu),乙腈-28%磷酸为流动相进行梯度洗脱,流速为0.8mL-min-1,进样量为1p L,柱温为35t,检测波长为360nm,建立16批罗布麻叶UPLC特征图谱,并进行相似度、聚类分析和主成分分析;同法测定药材中2种黄酮类成分(金丝桃苷、异槲皮苷)的含量。
结果:建立的6批罗布麻叶药材特征图谱共标定16个共有峰,相似度为0.287-0.999;聚类分析结合主成分分析结果表明,不同产地药材整体质量不太稳定;对金丝桃苷和异槲皮苷进行了含量测定,结果1批药材中2个成分的总含量差异不大,以安徽省阜阳市和河南省驻马店市产罗布麻叶药材黄酮类成分含量较高。
结论:建立的UPLC特征图谱结合含量测定及相似度评价、聚类分析和主成分分析评价方法,能全面地用于罗布麻叶药材的质量评价。

方法 选择毛细管区带电泳分离模式,以45mmol ・L -1硼砂缓冲液(pH 912)为电泳介质,未涂渍标准熔融石英毛细管(6415c m ×75μm,有效长度56c m )为分离通道,分离电压为25k V,检测波长为206n m,毛细管温度为25℃,压力进样为5000Pa,5s 。
结果 4种黄酮的峰面积与浓度的线性关系良好(r >019986);加样回收率为95134%~102165%。
结论 该方法简单、准确,重现性较好,可用于罗布麻叶药材内在质量的评价和控制。

HPLC 法测定维吾尔药大花罗布麻叶中异槲皮苷的含量穆建;佟瑞敏;苏来曼·哈力克【摘要】Objective To establish a method for the determination of isoquercitrin in leaves of Pocvnum hendersonii (Hook.f.)Woodson by HPLC.Methods HPLC method was adopted for a quantitative analysis of isoquercitrin.It was carried out on C18 column,with the mobile phase of methnal-acetonitrile-0.3% phosphoric acid water (11∶ 10∶ 79),the flow rate of 1.00 ml/min,the column temperature of 35℃ and the wavelength for detection of 354 nm.Results There were good linear relationships between the peak area and concentration at the range of 0.020 2~0.202 mg/mL for isoquercitrin (r =0.999 9).The average recovery rate of isoquercitrin was 104.45% (n =6),with RSD of 0.45%.The average content of ten different samples was 0.37%.Conclusion HPLC method to determine the isoquercitrin in leaves of Pocvnum hendersonii (Hook.f.)Woodson was proved to be simple,accurate and reproducible,which can provide scientific basis for quality control of leaves of Pocvnum hendersonii(Hook.f.)Woodson.%目的:建立高效液相色谱法测定大花罗布麻叶中异槲皮苷含量的方法。

罗布麻不同药用部位有效成分的含量考察

【摘要】目的:研究罗布麻不同药用部位有效成分的含量与药用价值。
方法:通过薄层色谱法、紫外分光光度法对罗布麻不同药用部位有效成分的含量进行研究。
结果:罗布麻各药用部位紫外吸收在204±2nm处有最大吸收;薄层色谱分析除罗布麻茎(2)、罗布麻根(3)外均在Rf值0.64、0.52处有翠绿色斑点;高效液相含量测定结果:罗布麻叶中槲皮素含量均>中国药典规定的0.60%,罗布麻花、茎、根、全草中槲皮素的含量均
