多肽无疑是最热门的功效原料之一。

蛋白质是生命活动的主要承担者。

多个、多种氨基酸缩合成多肽链，单根多肽链或多根多肽链连接后，继而空间折叠形成蛋白质。

多肽是蛋白质的片段，具有生理调节功能和营养功能，对于人体的生长发育、营养代谢、免疫调节、内分泌平衡等生理机能都有调节作用，与人体的健康、衰老、疾病、治疗、康复密切相关。

多肽应用广泛。

作为药物被应用，目前世界上有80多个多肽药物上市，全球有150多个品种处于临床研究阶段，400～600个治疗多肽在进行临床前研究；作为美容功效被应用，全世界有数百种美容肽；作为食物调味被应用，著名的代糖阿斯巴甜，就是典型的二肽衍生物；被应用于营养，如我国批准的新资源食品中就有玉米低聚肽粉及小麦低聚肽；被应用于表面活性剂，有日本Kaneka 生产的Surfactin。

多肽获取方式主要有化学合成法、发酵法、提取法及降解法。

提取法产量极小，实验室做研究可以，但几乎无法工业化；降解法比较粗放，无论酸碱法还是酶法，蛋白质降解很难得到单一分子结构的多肽，因为肠道可以直接吸收部分小肽，降解法用于生产保健品原料可行，成本较低，产量也可以很大。

目前也有部分化妆品原料是此类，比如水解大豆蛋白，但这类混合物成分复杂。

发酵法的优势是利用细胞工厂，获得纯度高的目标多肽，不存在消旋问题，规模放大容易，还可以获得合成困难的多肽。

发酵法需要先构建高效表达的工程菌株，这是困难且花费大量时间和成本的。

但发酵法优点突出，绿色可持续，用合成生物学来生产多肽，是未来的发展方向。

目前发酵法获得的短肽如谷胱甘肽，获得的长肽如乳酸链球菌素、ε-聚赖氨酸盐酸盐、溶菌酶、r-聚谷氨酸等大宗产品，还有如干扰素类、白介素类、生长因子类、重组胶原蛋白、超氧化歧化酶SOD 等珍贵原料，这些都是用化学合成法难以获得的成分，尤其如超氧化歧化酶SOD,是生物体细胞内广泛存在且必需的抗氧化成分，功效全面。

重复上述肽键形成反应，使肽链从C端向N端生长，直至达到所需要的肽链长度。

最后脱去保护基X，用HF水解肽链和固相载体之间的酯键，就得到了合成好的肽。

固相合成的优点主要表现在最初的反应物和产物都是连接在固相载体上，因此可以在一个反应容器中进行所有的反应，便于自动化操作，加入过量的反应物可以获得高产率的产物，同时产物很容易分离。

化学合成多肽现在可以在程序控制的自动化多肽合成仪上进行。

Merrifield成功地合成出了舒缓激肽（9肽）和具有124个氨基酸残基的核糖核酸酶。
