方法枸杞子药材经正己烷-无水乙醇-丙酮-甲苯(10 ∶ 6 ∶ 7 ∶ 7,V/V/V/V)超声提取后,采用高效液相色谱法进行测定。
以YMC C30为色谱柱,以甲醇-乙腈-水(81 ∶ 14 ∶ 5,V/V/V)为流动相A、二氯甲烷为流动相B进行梯度洗脱,柱温为20 ℃,流速为1.0 mL/min,检测波长为450 nm,进样量为20 μL。
以玉米黄质为参照物,计算β-胡萝卜素、β-隐黄素棕榈酸酯、玉米黄素双棕榈酸酯的相对校正因子,再以此计算上述成分的含量,并与外标法测定结果进行比较。
结果玉米黄质、β-胡萝卜素、β-隐黄素棕榈酸酯、玉米黄素双棕榈酸酯检测质量浓度的线性范围分别为0.119 4~2.983 8、0.121 7~1.521 6、0.285 9~5.718 8、8.460 5~211.513 3 μg/mL(R2均大于0.999);精密度、重复性、稳定性(16 h)试验的RSD均小于4%;平均加样回收率分别为103.34%、107.37%、105.64%、96.16%(RSD关键词枸杞子;高效液相色谱法;一测多评法;玉米黄质;β-胡萝卜素;β-隐黄素棕榈酸酯;玉米黄素双棕榈酸酯Establishment of quantitative analysis of multi-components by singer marker for content determination of 4 carotenoids in Lycium barbarumCHEN Huan1,2,MA Ling1,SU Xiaojuan3,LI Yanting3,MA Xia1,MA Xueqin3(1. METHODS L. barbarum was extracted with n-hexane-anhydrous ethanol-acetone-toluene (10 ∶6 ∶7 ∶ 7, V/V/V/V) by ultrasonic method. High performance liquid chromatography (HPLC) method was adopted. The determination was performed on YMC C30 column with mobile phase A consisted of methanol-acetonitrile-water (81 ∶ 14 ∶ 5, V/V/V) and mobile phase B consisted of dichloromethane (gradient elution) at the flow rate of 1.0 mL/min. The column temperature was set at 20 ℃. The detection wavelength was set at 450 nm, and sample size was 20 μL. Using zeaxanthin as control, the relative correction factors (RCFs)of β-carotene,β-cryptoxanthin palmitate and zeaxanthin dipalmitate were calculated, and then the content of each component was calculated according to RCFs and compared with the results of external standard method(ESM). RESULTS The linear range of zeaxanthin,β-carotene,β-cryptoxanthin palmitate and zeaxanthin dipalmitate were 0.119 4-2.983 8, 0.121 7-1.521 6, 0.285 9-5.718 8,8.460 5-211.513 3 μg/mL(all R2>0.999). RSDs of precision, repeatability and stability(16 h)tests were all less than 4%. 经响应面优化后,获得的最佳提取工艺为:液固比为33、提取时间为41min、提取温度为48℃,该条件下提取枸杞中的叶黄素,最终获得0.93mg/g的提取量。
关键词:枸杞干果;叶黄素;提取;最佳工艺枸杞之所以具有良好的眼保健功效,与其含有大量的一羟叶黄素与二羟叶黄素的原因分不开。
叶黄素(lutein)是含有多个共轭双键的类胡萝卜素,研究表明,其具有预防动脉硬化、抗诱变、抗肿瘤、治疗白内障的功效[1-3]。
然而,目前对枸杞叶黄素提取工艺的研究基础较为薄弱,这大大限制了枸杞中叶黄素的开发和应用,因此急需开发枸杞中叶黄素的提取方法。
本实验进行叶黄素提取工艺的研究,并通过响应曲面法优选最佳工艺参数,为进一步开发利用这一天然色素提供参考与借鉴。
一、材料与方法仪器和设备。
SP-752型紫外可见分光光度计;HZS-H型水浴恒温振荡器;KQ-100DB型超声波清洗器;YHG-50×55型远红外快速恒温干燥箱。
材料和试剂。
叶黄素标准品,CHROMADAEX公司(进口);分析纯无水乙醇;分析纯石油醚;分析纯丙酮;分析纯正己烷;分析纯乙酸乙酯;色谱级二氯甲烷。
提取方法的选择。
本文分别采用有机溶剂浸提法、超声波辅助有机溶剂提取法、微波辅助有机溶剂提取法提取枸杞叶黄素。
(1)超声波辅助有机溶剂提取法单因素试验:称取0.50g枸杞粉,加入15mL提取剂,分别考察提取溶剂、固液比、超声时间、超声温度及提取次数对提取的影响。
(2)正交组合试验:从单因素中选出影响叶黄素提取量的主要因素,进行组合试验,采用Design-Expert 7.0进行分析,优化叶黄素提取工艺。
二、试验结果与讨论提取方法的选择。
通过试验,溶剂提取法、超声波辅助提取法及微波辅助提取法的吸光值分别为0.231、0.412和0.378,故而得出,超声波辅助法提取枸杞叶黄素效果最佳。

本研究旨在探究在提取枸杞皮渣中类胡萝卜素的最佳工艺,并研究其稳定性,为其应用于食品和药品工业提供科学依据。
通过酶解、溶剂提取和净化析出等步骤,成功提取到了枸杞皮渣中的类胡萝卜素。
同时,通过稳定性研究验证了其适用性。
关键词:枸杞皮渣;类胡萝卜素;提取;稳定性引言类胡萝卜素是一类色素化合物,广泛存在于植物组织中,具有抗氧化、抗肿瘤、抗衰老、保护视力等多种生物活性。
枸杞皮渣是枸杞加工过程中剩余的废弃物,含有丰富的类胡萝卜素,但其利用率却相对较低。
因此,本研究旨在提取枸杞皮渣中的类胡萝卜素,并研究其稳定性,为其在食品和药品工业中的应用提供科学基础。
材料与方法1. 枸杞皮渣的制备:将新鲜枸杞果实收获后,进行果肉与种子的分离,然后将果皮晒干至含水量低于10%,并研磨成粉末。
2. 酶解提取类胡萝卜素:将10g枸杞皮渣粉末加入50mL 0.6% NaOH溶液中,于60°C水浴中酶解2小时,反应结束后通过离心分离上清液。
3. 溶剂提取:将上清液与1:1的醇和石油醚混合,振荡抽提20分钟,再通过离心分离,得到石油醚相。
4. 净化析出:将石油醚相置于旋转蒸发器中,待溶剂大部分挥发后,加入正己烷研磨均匀,形成含有类胡萝卜素的油状物质。
5. 稳定性研究:将提取出的类胡萝卜素溶于不同溶剂(如甲醇、乙醇、二甲基亚砜等),在不同温度和时间下进行测定。
结果与讨论经过酶解和溶剂提取的提取液中成功检测到了类胡萝卜素的吸收峰,且其浓度与提取液中的类胡萝卜素含量呈正相关关系。
通过净化析出,获得了高纯度的类胡萝卜素油状物质。
稳定性研究结果显示,在室温下保存72小时内,类胡萝卜素的含量基本保持稳定,但随着时间和温度的增加,其含量逐渐下降。
结论本研究成功提取了枸杞皮渣中的类胡萝卜素,并通过稳定性研究验证了其适用性。

为了提高枸杞叶中多酚物质的提取效率及探究其生物活性功能,以枸杞叶为试验原料,基于超声波辅助酶法探究乙醇体积分数、料液比、超声时间和纤维素酶添加量等4个单因素对枸杞叶多酚提取效率的影响,通过响应面分析优化枸杞叶多酚的提取工艺,并探究枸杞叶多酚提取物对DPPH 和ABTS 自由基的清除能力。
结果表明,4个单因素对枸杞叶多酚提取效率均具有一定影响,枸杞叶多酚提取效率均呈先升高后降低的趋势;经过拟合优化取得最优的提取条件为:乙醇体积分数68%、料液比1∶89(g/mL )、超声时间39 min 、纤维素酶添加量2 mg/g ,在此条件下,枸杞叶提取液中多酚含量为5.09 mg/g ,与预测值(5.21 mg/g )的相对误差仅为2.3%。
抗氧化试验表明,枸杞叶多酚提取物对DPPH 和ABTS 自由基的清除率随着溶液多酚质量浓度的增加而升高,半数抑制浓度IC 50值分别为27.28、190.00 μg/mL ,表明具有较好的抗氧化能力。
综上,超声辅助酶法可提高枸杞叶多酚的提取效率,所提取的枸杞叶中多酚具有明显的体外抗氧化活性。

In order to improve the extraction efficiency and explore biological activity of polyphenols from Lycium barbarum leaves, in this study, taking Lycium barbarum leaves as the test material, the effects of four single factors including ethanol volume fraction, solid -liquid ratio, ultrasonic time, and the amount of cellulase added on the extraction efficiency of polyphenols from Lycium barbarum leaves were investigated using the ultrasonic -assisted enzymatic extraction. The response surface analysis was used to optimize the extraction technology of polyphenols from Lycium barbarum leaves and the scavenging ability of polyphenol extracts from Lycium barbarum leaves on DPPH and ABTS free radicals was studied. The results showed that the four single factors had the same trend that increased first and then decreased on the extraction efficiency of polyphenols from Lycium barbarum leaves. After design optimization, the optimal extraction conditions were defined with: 68% of ethanol volume fraction, 1∶89(g/mL) of solid -liquid ratio, 39 min of ultrasonic time, and 2 mg/g of cellulase, while the content of polyphenols from Lycium barbarum leaves obtained under this condition was 5.09 mg/g, which had a relative error with the predicted value(5.21 mg/g) was 2.3%. The results of antioxidant activities showed that the removal rate of the polyphenols from Lycium barbarum leaves on DPPH and ABTS increased with increasing mass concentration of polyphenols. The IC 50 values of half inhibitory concentration were 27.28 μg/mL and 190 μg/mL, respectively, which indicated that Lycium barbarum leaves had excellent antioxidant capacity. The method of ultra-sound-assisted extraction with a series of advantages such as energy saving, time-saving and high efficiency, has received substantial attention. In current work, combined with the related literature domestic and foreign, the mass transfer mechanism of ultrasound-assisted extraction of carotenoids was explained. The applications and developments of ultrasound-assisted extraction technology in extracting carotenoids were investigated in present paper as well.
