方法:血浆样本经甲醇沉淀蛋白后,以伊马替尼为内标,采用液相色谱-串联质谱(LC-MS/MS)法测定。
以Ultimate AQ-C18为色谱柱,以甲醇(含0.1%甲酸)-水(0.1%甲酸)为流动相进行梯度洗脱,流速为0.4 mL/min,柱温为40 ℃,进样量为5 µL。
离子源为电喷雾离子源,以多反应监测模式进行正离子扫描,用于定量分析的离子对分别为m/z 583.4→138.3(吡咯替尼)、494.5→393.4(内标)。
选择2020年6-11月于青岛大学附属医院接受吡咯替尼治疗的乳腺癌患者30例,测定其用药1周后的吡咯替尼稳态谷濃度。
结果:吡咯替尼检测质量浓度的线性范围为5~300ng/mL(r=0.999 3),定量下限为5 ng/mL;日内、日间RSD均不高于9.30%,相对误差为-6.70%~5.04%;稳定性试验的相对误差为 -1.92%~5.42%;提取方法、基质效应、残留效应均不影响待测物的定量分析。
30例乳腺癌患者体内吡咯替尼的稳态谷浓度为32.6~82.8 ng/mL,平均血药浓度为53.8 ng/mL,存在2.54倍的个体差异。
结论:所建LC-MS/MS法操作简便、灵敏度和准确度较高,可用于乳腺癌患者体内吡咯替尼的血药浓度监测。

METHODS: After precipitated with methanol,the plasma sample was determined by LC-MS/MS using imatinib as internal standard. The determination was performed on Ultimate AQ-C18 column with mobile phase consisted of methanol (containing 0.1% formic acid) and water (containing 0.1% formic acid)(gradient elution) at the flow rate of 0.4 mL/min. The column temperature was 40 ℃, and the sample size was 5 µL. The ion source was electrospray ionization source, and the positive ion scanning was carried out in multiple rea ction mode. The ion pairs for quantitative analysis were m/z 583.4→138.3 (pyrrotinib)and m/z 494.5→393.4 (internal standard), respectively. Thirty breast cancer patients taking pyrrotinib were collected from the Affiliated Hospital of Qingdao University during Jun.-Nov. 2020 to determine their steady-state trough concentrations of pyrrotinib after a week of treatment. RESULTS: The linear range of pyrrotinib were 5-300 ng/mL (r=0.999 3). The lower limit of quantification was 5 ng/mL. RSDs of intra-day and inter-day were not higher than 9.30%, and relative errors (REs) ranged -6.70%-5.04%. REs of stability tests were in the range of -1.92%-5.42%. The extraction method, matrix effect and residual effect did not affect the quantitative analysis of the substance to be tested. The steady-state trough concentrations of pyrrotinib were 32.6-82.8 ng/mL,with an average plasma concentration of 53.8 ng/mL; there was about 2.54 fold individual difference. Abstract: A UPLC-MS/MS method was established to quantitatively determine the content of alliin in animal plasma to study whether alliin and alliin in garlic enteric preparations can react to produce the active ingredient allicin in the in vivo environment. Methods Reversed-phase C18 column (Waters ICQUITY UPLC BEH, 100 × 2.1 mm, 1.7μm), column temperature: 40 ℃, flow rate: 0.15 mL/min, injection volume: 2μl, Mobile phase: 0.1% formic acid (A)-acetonitrile (B), gradient elution; mass spectrometry ionization: ESI+, determination of allicin in rat plasma . Results The results of two parallel experiments of garlic enteric preparation and enzymatic garlic powder showed that in the garlic enteric preparation with allinase, the plasma concentration of alliin in the blood of rats was significantly lower. Conclusion A UPLC-MS/MS method for the quantitative determination of alliin in animal plasma has been established. 基于CFDA SE 荧光标记的细胞增殖检测和[3H]-thymidine 掺入、BrdU 标记获得的检测结果完全一致,但同时可以提供更多的细胞增殖信息。
使用CFDA SE 检测可以提供整个细胞群中有多少比例的细胞分裂了1次、2次或更多次数,同时如果和其它荧光探针联用,可以获取不同分裂次数细胞的其它相关信息。
CFDA-SE 的分子式为C 29H 19NO 11,分子量为557.47,CAS number 为150347-59-4。
CFDA SE 可以通透细胞膜,进入细胞后可以被细胞内的酯酶(esterase)催化分解成CFSE ,CFSE 可以偶发性地(spontaneously)并不可逆地和细胞内蛋白的Lysine 残基或其它氨基发生结合反应,并标记这些蛋白。
在加入荧光探针CFDA SE 后大约24小时,即可充分标记细胞。
被CFDA SE 标记的非分裂细胞的荧光非常稳定,稳定标记的时间可达数个月。
CFDA SE 标记细胞的荧光非常均一,比以前使用的其它细胞示踪荧光探针例如PKH26的荧光更加均一,并且分裂后的子代细胞的荧光分配也更均匀。
由于CFDA SE 标记细胞的荧光非常均匀和稳定,每分裂一次子代细胞的荧光会减弱一半,这样通过流式细胞仪检测就可以检测出没有分裂的细胞,分裂一次的细胞(1/2的荧光强度),分离两次的细胞(1/4的荧光强度),分裂三次的细胞(1/8的荧光强度)以及类似的其它分裂次数的细胞。
采用CFDA SE 通过流式细胞仪检测获得的检测结果参考右图。
每一个峰代表一种分裂次数的细胞,从右至左的峰通常依次为分裂0次、1次、2次、3次等次数的细胞。
高尔基体概述高尔基体(Golgi apparatus)是由许多扁平的囊泡构成的以分泌为主要功能的细胞器。
又称高尔基器或高尔基复合体;在高等植物细胞中称分散高尔基体。
最早发现于1855年,1898年由意大利人卡米洛•高尔基(Camillo Golgi,1844-1926)在光学显微镜下研究银盐浸染的猫头鹰神经细胞内观察到了清晰的结构,因此定名为高尔基体。
因为这种细胞器的折射率与细胞质基质很相近,所以在活细胞中不易看到。
高尔基体从发现至今已有100多年的历史,其中一半以上的时间是进行关于高尔基体的形态甚至是它是否真实存在的争论。

方法:采用Agilent Poroshell 120 EC-C18色谱柱;流动相为纯水(0.1%甲酸):甲醇(V/V)=60:40;稀释剂为乙腈(0.1%甲酸):纯水(V/V)=50:10;柱温为40℃;进样体积为5µl;流速为0.4ml/min;采用正离子模式进行扫描。
对甲苯磺酸甲酯测定浓度在0.76ng/ml~15.27ng/ml范围内,线性关系良好;对甲苯磺酸乙酯测定浓度在0.75ng/ml~15.01ng/ml范围内,线性关系良好。
对甲苯磺酸甲酯的定量限为0.0038ng;对甲苯磺酸乙酯的定量限为0.0038ng。
杂质回收率在限度浓度80%、100%和160%三个浓度水平均在90~110%之间,该方法准确度良好。
该方法适用于西他沙星原料中对甲苯磺酸甲酯和对甲苯磺酸乙酯的检测。
西他沙星(sitafloxacin)是日本第一制药有限公司继左氧氟沙星后开发出的一种强力广谱新氟喹诺酮类抗菌剂,该药对革兰氏阳性球菌,革兰氏阴性菌以及厌氧菌的抗菌活性是左氧氟沙星的4~32倍,同时对肺炎球菌DNA 促旋酶和拓扑同功酶有双重抑制作用。
临床表现有极广的抗菌谱,特别是对呼吸道的病菌有极强的抗菌活性。
因西他沙星的一个起始物料为对甲苯磺酸盐,在后续反应中对甲苯磺酸若有残留,可能会与溶剂甲醇、乙醇反应生成具有基因毒性的杂质—对甲苯磺酸甲酯和对甲苯磺酸乙酯,故采用LC-MS法对产品中的对甲苯磺酸甲酯/乙酯进行控制。
1、实验部分1.1仪器与试药Agilent 1200液相色谱仪(美国安捷伦公司);Agilent 6460三重串联四极杆质谱仪(美国安捷伦公司);XP205型电子天平(瑞士梅特勒托利多公司)。
实验研究依达拉奉右莰醇通过铁死亡-脂质过氧化通路对脑出血大鼠神经保护的作用机制毛权西,李作孝△摘要:目的探讨依达拉奉右莰醇对脑出血大鼠的神经保护作用及血肿周围脑组织脂质过氧化的影响。
方法将128只SD大鼠随机分为假手术组、脑出血组、依达拉奉组和依达拉奉右莰醇组,每组32只。
除假手术组外,其余组大鼠构建急性脑出血模型,依达拉奉组、依达拉奉右莰醇组于造模后分别腹腔注射依达拉奉6mg/kg、依达拉奉右莰醇7.5mg/kg,每12h注射1次,假手术组和脑出血组腹腔注射等量生理盐水。
术后1d、3d、7d和14d按Garcia评分标准进行神经功能评分,HE染色观察血肿周围脑组织病理变化,化学荧光法检测血肿周围脑组织活性氧(ROS)含量,微量酶标法检测血肿周围脑组织还原型谷胱甘肽(GSH)含量,蛋白免疫印迹法检测血肿周围脑组织谷胱甘肽过氧化物酶4(GPX4)、长链脂酰辅酶A合成酶4(ACSL4)和磷脂胆碱酰基转移酶3(LPCAT3)表达。
结果与假手术组比较,脑出血组大鼠神经功能评分降低,血肿周围脑组织出现大量炎性细胞浸润及神经细胞变性,ROS含量、ACSL4和LPCAT3蛋白表达水平升高,GSH含量、GPX4蛋白表达水平降低(P<0.05);与脑出血组比较,依达拉奉组和依达拉奉右莰醇组大鼠神经功能评分升高,血肿周围脑组织病理损伤明显减轻,ROS含量、ACSL4和LPCAT3蛋白表达水平降低,GSH含量、GPX4蛋白表达水平增加(P<0.05);依达拉奉右莰醇组干预效果优于依达拉奉组(P<0.05);除假手术组外,其余各组均在术后3d时变化最明显,术后7d、14d逐渐恢复(P<0.05)。
结论依达拉奉右莰醇可能通过调节脑出血大鼠神经细胞铁死亡相关蛋白的表达,减少脑组织脂质过氧化,抑制神经细胞铁死亡,从而发挥脑保护作用。
