Regulationofcellwallbiosynthesis

Plantcellwallsdifferintheiramountandcompositionamong

variouscelltypesandevenindifferentmicrodomainsofthewall

ofagivencell.Plantsmusthaveevolvedregulatory

mechanismscontrollingbiosynthesis,targetedsecretion,and

assemblyofwallcomponentstoachievetheheterogeneityin

cellwalls.Anumberoffactors,includinghormones,the

cytoskeleton,glycosylphosphatidylinositol-anchoredproteins,

phosphoinositides,andsugarnucleotidesupply,havebeen

implicatedintheregulationofcellwallbiosynthesisor

deposition.Inthepasttwoyears,therehavebeenimportant

discoveriesintranscriptionalregulationofsecondarywall

biosynthesis.SeveraltranscriptionfactorsintheNACandMYB

familieshavebeenshowntobethekeyswitchesforactivation

ofsecondarywallbiosynthesis.Thesestudiessuggesta

transcriptionalnetworkcomprisedofahierarchyof

transcriptionfactorsisinvolvedinregulatingsecondarywall

biosynthesis.Furtherinvestigationandintegrationofthe

regulatoryplayersparticipatinginthemakingofcellwallswill

certainlyleadtoourunderstandingofhowwallamountsand

compositionarecontrolledinagivencelltype.Thismay

eventuallyallowcustomdesignofplantcellwallsonthebasis

ofourneeds.

Introduction

Plantcellsareencasedinrigidwallscomposedofcellu-

lose,hemicellulose,pectin,proteins,and/orlignin,which

varyinamountdependingoncelltypes.Thewalls

functionastheplant‘exoskeleton’anddefinethecell

shape,andultimatelyorganandwholeplantmorphology.

Theynotonlyregulatecellgrowthandprovidestructural

andmechanicalsupporttoplants,butalsoactasaphysical

barriertobioticandabioticstresses.Plantcellwallsare

importantinhumanlife,providingmajordietaryfibers

andessentialrawmaterialsfortextile,lumber,pulping,

andpotentiallyforbiofuels.Therefore,understandinghowplantsmakecellwallsisimperativeforbasicplant

biologyandwillhavefar-reachingimpactsonbiotechno-

logicalapplications.

Inthelastdecade,tremendousprogresshasbeenmade

towardidentificationandfunctionalcharacterizationof

genesinvolvedinthebiosynthesisofcellwallpolysac-

charides,includingcellulose,glucomannan,xyloglucan,

xylan,andpectin[1󰀂,2󰀂,3,4󰀂,5󰀂,6󰀂,7󰀂],aswellasgenes

responsibleforthebiosynthesisofsugarnucleotide

donorsinvolvedinpolysaccharidebiosynthesis[8].Mol-

ecularandbiochemicaldissectionofgenesparticipating

inthebiosynthesisofindividualcellwallcomponentsis

thefirstcriticalsteptowardunderstandingcellwallbio-

synthesis.Althoughplantcellwallsaremadeofcommon

components,theiramountandcompositiondifferamong

variouscelltypesandevenindifferentmicrodomainsof

thewallofagivencell[9].Thewallscanbemodifiedto

meetthespecialneedsofcertaincelltypes,suchaswater-

conductingtrachearyelements,guardcells,andendoder-

mis.Inaddition,theamountandcompositionofwalls

maychangeduringcellgrowth[10]andinresponseto

bioticandabioticstresses[11].Therefore,plantsmust

haveevolvedmechanismstoturnondifferentbiosyn-

theticpathwaysofwallcomponentsinvariouscelltypes,

synthesizetherightamountofwallcomponents,and

assemblethemintherightplacetosuitawallfitto

functionsofspecificcelltypes.

Tounderstandhowplantsmakedifferentwalltypes,itis

necessarytoknowthebiosyntheticgenesresponsiblefor

theirsynthesisandalsotheregulationofcellwallbio-

synthesisandassembly.Anumberoffactors,suchas

hormones[12–15],thecytoskeleton[16󰀂],glycosylpho-

sphatidylinositol-anchoredproteins[17–19],phosphoino-

sitides[20–22],sugarnucleotidesupply[23],and

coordinationofwallbiosynthesis[24,25],havebeen

implicatedintheregulationofcellwallbiosynthesisor

deposition.Otherfactorsmightalsobeinvolvedinthe

targetedsecretionofwallmaterials,therebyregulating

walldynamicsandheterogeneity[26].Owingtospace

limitation,thisreviewonlyfocusesontherecent

advancesinthetranscriptionalregulationofcellwall

biosynthesis.

Heterogeneityofcellwalls

Beforethediscussionofregulationofcellwallbiosyn-

thesis,itisimportanttobrieflyreviewthediversityofcell

wallsinplants.Althoughcellwallsaregenerallycom-

posedofcellulose,hemicellulose,pectin,proteins,and/or

lignin,theamountandcompositionofindividualwall

componentsandtheshapesofwallsvaryconsiderably
