DOI:10.16305/j.1007-1334.2016.01.024补肾益气方联合有氧运动对衰老大鼠骨骼肌线粒体生物合成相关信号分子表达的影响徐划萍1,金国琴2,韩志芬21.上海中医药大学体育部(上海201203);2.上海中医药大学基础医学院(上海201203)【摘要】目的观察补肾益气方及有氧运动对D-半乳糖所致衰老大鼠骨骼肌线粒体生物合成相关信号分子丝裂原活化蛋白激酶(p38MAPK)、钙调蛋白激酶Ⅱ(CaMKⅡ)、过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)、心肌细胞增强因子2C (MEF2C)及细胞色素氧化酶(COXⅣ)表达的影响。
方法将40只雄性SD大鼠随机分为5组,分别为正常组、模型组、中药组、运动+中药组、美雄酮组。
除正常组外,其余各组每天颈背部皮下注射10%的D-半乳糖(100μg/g)溶液,制作“拟衰老”大鼠模型。
模型大鼠分别经补肾益气方药、有氧跑台运动、美雄酮干预10周后,采用ELISA法测定大鼠血清β-半乳糖苷酶(β-Galase)含量,采用甲基百里香酚蓝法(MTB)检测大鼠腓肠肌线粒体钙含量,采用Western blotting法检测大鼠腓肠肌线粒体生物合成信号通路分子p38MAPK、CaMKⅡ、PGC-1α、MEF2C及COXⅣ蛋白表达变化。
结果与正常组比较,模型组大鼠衰老生物学标志物β-Galase、线粒体钙含量显著增高,CaMKⅡ、MEF2C、PCG-1α蛋白表达明显下降,而P38MAPK蛋白表达显著升高(P<0.05,P<0.01)。
与模型组比较,中药组可显著降低血清β-GALase蛋白含量、线粒体钙含量(P<0.01);运动+中药组可显著降低线粒体钙含量,上调CaMKⅡ、PGC-1α、MEF2C及COXⅣ蛋白表达;美雄酮组显著降低线粒体钙含量,上调PCG-1α、COXIV蛋白表达(P<0.05,P<0.01)。
结论衰老大鼠骨骼肌功能减退可能与线粒体生物合成相关信号转导通路分子CaMKⅡ、p38MAPK、PCG-1α、MEF2C及能量代谢相关酶蛋白基因(COXIV)表达异常变化有关;补肾益气方联合有氧运动可一定程度改善上述指标的异常变化,延缓骨骼肌衰老。
【关键词】补肾益气方;有氧运动;衰老大鼠;骨骼肌;线粒体生物合成The influence of Bushen Yiqi Decoction and aerobic exercise on the expression of mitochondria biosynthesis related signaling molecules in the aging rats’skeletal muscleXU Hua-ping1,JIN Guo-qin2,HAN Zhi-fen21.Sports Department,Shanghai University of Traditional Chinese Medicine,Shanghai201203;2.College of Basic Medicine,Shanghai University of Traditional Chinese Medicine,Shanghai201203Abstract:Objective To observe the influence of Bushen Yiqi Decoction and aerobic exercise on the expression of mitochondria biosynthesis related signaling molecules p38MAPK,CaMKⅡ,PGC-1α,MEF2C and COXⅣin D-galactose-induced aging rats’skeletal muscle.Methods40male SD rats were randomly divided into five groups:normal group,model group,traditional Chinese medicine group,exercise and traditional Chinese medicine group and methandienone group.The rats were received a subcutaneous injection of10%D-galactose solution(100mg·kg-1,once a day)for10weeks to make the aging model except normal group rats.Bushen Yiqi Decoction,aerobic exercise and methandienone were used to the corresponding experimental groups to intervene for10weeks.β-Galase of blood serum were tested by ELISA;Calcium content of gastrocnemius mitochondria were tested by Methyl thyme phenol blue(MTB);The expression of mitochondrial biosynthesis related signal transduction molecules(p38MAPK,CaMKⅡ,PGC-1α,MEF2C and COXⅣ)were tested by Western blot respectively.Results Compared with normal group,model group rats’aging biology markersβ-Galase,calcium content in mitochondria increased significantly.The protein expression(protein)of model rats’CaMKⅡ,MEF2C,PCG-1alpha decreased obviously,and the protein expression of P38MAPK increased significantly.The Bushen Yiqi Decoction combined with aerobic exercise could obviously decrease calcium content,and increase the protein expression of CaMKⅡ,PGC-1α,MEF2C and COXⅣof the model aging rats.The methandienone could obviously increase the protein expression of PCG-1αand COXIV of the model aging rats,decrease calcium content obviously.Conclusion The decline of aging model rats’skeletal muscle function may related to expression changes of mitochondria biosynthesis related signal transduction pathways molecular CaMKⅡ,p38MAPK,PCG-1alpha,MEF2C and energy metabolism related genes(COXIV).The Bushen Yiqi Decoction combined with aerobic exercise can delay age-related degradation of the skeletal muscle in some degree through improving the abnormal changes of the above index.Keywords:Bushen Yiqi Decoction;aerobic exercise;aging rats;skeletal muscle;mitochondria biosynthesis[基金项目]国家自然科学基金项目(81473583);上海市教委资助项目(2012JW29,085ZY1204)[作者简介]徐划萍,女,博士,讲师,主要从事有氧运动及中药抗衰老方面研究工作[通讯作者]金国琴,教授,博士生导师。

具备一定的骨骼肌质量和力量水平是人体进行各种活动的前提,然而随着年龄的增长,骨骼肌逐渐出现质量改变、力量下降等衰退变化,严重影响老年人的生活质量。
骨骼肌细胞线粒体数量和功能是骨骼肌的重要动力基础,亦是骨骼肌衰老的重要影响因素之一。
线粒体在衰老引起的骨骼肌质量和强度下降中发挥着关键作用,其生物合成的高低影响着骨骼肌能量代谢的效率。
老年骨骼肌线粒体生物合成相关信号分子表达异常,可使线粒体功能受损、能量代谢障碍,从而加速衰老。
中医学认为,补肾填精、健脾益气是防治骨骼肌衰老的基本法则。
已有研究证明,补肾益气类中药可不同程度地改善衰老骨骼肌线粒体的结构及功能[1-5]。
“流水不腐,户枢不蠹,动也。
”长期坚持运动,能使人的衰老明显延迟,但有氧运动延缓骨骼肌衰老的具体作用机制仍有待探明。
本研究主要观察补肾益气方(由制何首乌、肉苁蓉、黄芪和党参四味中药组成)及有氧运动对D-半乳糖所致衰老大鼠骨骼肌线粒体生物合成相关信号分子表达的影响,以期从能量代谢角度揭示中药延缓骨骼肌衰老的部分作用机制。
1材料与方法1.1材料1.1.1动物雄性清洁级SD大鼠40只,6周龄,购自上海中医药大学动物实验中心,体质量(217ʃ19)g。
动物合格证号:SCXK(沪)2007-0005。
1.1.2药物与试剂补肾益气方(由黄芪15g、党参15g、制何首乌15g、肉苁蓉15g组成),上海康桥中药饮片有限公司生产。
常规煎制,浓缩,储存于玻璃瓶,-20ħ保存。
β-半乳糖苷酶ELISA检测试剂盒,上海硕嘉生物科技有限公司;组织线粒体分离试剂盒,碧云天生物技术研究所;钙测试试剂盒,南京建成生物工程研究所;兔抗大鼠β-tubulin单克隆抗体、兔抗大鼠COXⅣ单克隆抗体、兔抗大鼠P38MAPK单克隆抗体、兔抗大鼠CAMKⅡ单克隆抗体,EPITMICS公司;山羊抗大鼠PGC-1α多克隆抗体、兔抗大鼠MEF2C多克隆抗体,Abcam(Hong Kong)公司;HRP标记兔抗山羊IgG二抗、HRP标记山羊抗兔IgG二抗,上海冠泰生物科技有限公司(BIOTNT);DEPC、PVDF膜、RIPA裂解液、DBA 显色试剂盒,上海硕嘉生物科技有限公司;预染蛋白MARKER、BCA蛋白定量试剂盒,Thermo scientific公司;超纯水为实验室自制。
