人核基质蛋白-22ELISA检测试剂盒使用说明书提供商:上海乔羽生物科技有限公司本试剂仅供研究使用标本:尿液试验原理:NMP-22试剂盒是固相夹心法酶联免疫吸附实验(ELISA).已知NMP-22浓度的标准品、未知浓度的样品加入微孔酶标板内进行检测。
先将NMP-22和生物素标记的抗体同时温育。
人核基质蛋白-22ELISA检测试剂盒洗涤后,加入亲和素标记过的HRP。
再经过温育和洗涤,去除未结合的酶结合物,然后加入底物A、B,和酶结合物同时作用。
产生颜色。
颜色的深浅和样品中NMP-22的浓度呈比例关系。
自备材料蒸馏水。
加样器:5ul、10ul、50ul、100ul、200、500ul、1000ul。
振荡器及磁力搅拌器等。
安全性避免直接接触终止液和底物A、B。
一旦接触到这些液体,请尽快用水冲洗。
实验中不要吃喝、抽烟或使用化妆品。

试剂盒使用说明和包被实验示例:一、包被液的配制:coating buffer 包被缓冲液(EL-CPX01):该试剂为10X 浓缩液,使用前请根据实验用量,用蒸馏水稀释至1X,即配即用,例:10ml coating buffer + 90ml蒸馏水;Coating buffer 用于抗体的包被;可替代以PBS为主的包被液体;请用1* coating buffer 和适量一抗配成包被液,抗体浓度请参考抗体的说明。
二、包被实验示例:在ELISA酶标板(EL-CPX02)中每孔加入100ul包被液(包被液若有剩余可多包被一点,以条为单位),在板上贴膜(封板,plate sealer,),室温(控制室温在25℃,板放置处旁放置温度计控温)温育过夜(下午4点到第二天早上9点,不宜缩短时间,会降低包板效率);结束温育(也请参考抗体说明书的包被条件进行相应调整)。
多余的配好的包被液不能保存。
三、洗板封闭:1)将洗液20×Wash Buffer(EL-00402)(可以用PBS +Tween 20代替)配制成1×Wash Buffer(实验前请合理计算用量,配制成足够的量待用),用1×Wash Buffer洗板4次(注:包被组分在加稳定剂前不能剧烈震荡或完全干燥,所以洗板过程需快速、温和,不要拼命拍板);2)洗板后立即加入200ul每孔的酶标板稳定剂(EL-CPX03),37℃温育1小时;3)弃掉或吸取酶标板稳定剂(注:拍干,千万不要洗板)。
封闭好的酶标板可以有以下几种处理方式。
方式一:直接进行检测。
方式二:冻干(需有冻干机,长期保存)1)冻干成品:把酶标板放入冰箱,-20℃冰箱冷冻过夜。
2)调试真空冻干机,立即将冷冻过夜的酶标板放入真空冻干机冻干24小时;3)取出冻干的酶标板,立即将酶标板放入一个装有干燥剂的铝箔袋内(EL-CPI05-20),贴上标签,放4℃冰箱长期保存。
(冻干同时可进行标准品冻干,标准品冻干需要购买标准品冻干液)。

摘要和说明1, 2, 3在人血清中,血清蛋白主要包括白蛋白(占总蛋白的50-60%),其余部分包括α1-、α2-、β- 和γ-球蛋白。
总蛋白的浓度对维持血液和组织之间水的正常平衡与交换是很重要的。
血清蛋白浓度较低,可能是由于吸收不良、合成受损引起的,或是由于出血或过度分解代谢导致蛋白损失而引起的,如肾病和肝病中观察到的情况,这种情况可能导致低蛋白血症。
在高免疫球蛋白血症(如多发性骨髓瘤和感染)或脱水情况下,可能发生高蛋白血症。
方法的原理本分析法采用双缩脲反应测定血清蛋白,其中碱性溶液中的二价铜离子与化合物(含 2 个或多个、与碳原子结合的酰胺基或肽基)反应,形成一种有色络合物4。
碱性 pH血清蛋白质类 + Cu++———— 紫色络合物紫色络合物以分光光度法在 550nm 处测定,以 700nm 作为空白波长。
在较宽的线性范围内,该络合物的密度与蛋白质浓度成正比5。
试剂五水合硫酸铜0.3g/dL氢氧化钠0.8g/dL碘化钾、酒石酸钾钠和叠氮化钠作为防腐剂。
注意事项1.当处理任何实验室试剂时,应遵守良好实验室安全规范(CLSI, GP17-A2)。
2.氢氧化钠具有腐蚀性,能引起烧伤。
不要用嘴吸取试剂。
如果吞服,应立即就医处理。
避免眼部和皮肤接触。
如发生眼部接触,立即用大量水清洗眼睛,并就医处理。
如果发生皮肤接触,用水清洗皮肤至少 15 分钟。
3.本试剂含叠氮化钠 <0.1%;叠氮化钠可与铅和铜水管反应,形成高爆炸性金属叠氮化物。

INTENDED USEThe 1st International Standard for HPV Type 16 (HPV-16) DNA Nucleic Acid Amplification Techniques consists of a freeze-dried preparation of recombinant plasmid containing full-length HPV-16 DNA cloned via its unique BamH1 site (Quint et al., 2006). The standard has been formulated in a background of purified human genomic DNA, lyophilized in 0.5 ml aliquots and stored at -20 °C. The material was calibrated in an international collaborative study involving 19 laboratories (Wilkinson et al., 2008). The International Standard contains material that is proprietory to third parties and should be used for the sole purpose of calibrating in-house or working standards for the amplification and detection of HPV-16 DNA. The International Standard should not be used for any other purpose and should be discarded after use. 2. CAUTIONThis preparation is not for administration to humans .This material contains DNA derived from C33A cells. As with all materials of biological origin, this preparation should be regarded as potentially hazardous to health. It should be used and discarded according to your own laboratory's safety procedures. Such safety procedures should include the wearing of protective gloves and avoiding the generation of aerosols. Care should be exercised in opening ampoules or vials, to avoid cuts.3.
