胸苷激酶1的研究进展邢晓光【摘要】恶性肿瘤是威胁人类健康最主要的疾病之一,目前主要靠病理学及影像学来诊断,只有患者具有明显占位性病变和临床症状后才能确诊,而此时已非癌变最早期,且已过最佳治疗时期.因此,对恶性肿瘤具有早期诊断作用的血清肿瘤标志物是目前研究的重点.随着现代血清学肿瘤标志物相关研究的迅速发展,一种新的细胞增殖标志物,即胸苷激酶1(TK1)被证实在肿瘤早期发现、疗效评估、预后判断等方面具有重要意义.现从TK1的结构、功能、与DNA合成及肿瘤的关系等方面予以综述.%Malignant tumor is the most important disease threatening human health, current major di-agonsis of which is by pathology and imaging; affirmative diagnosis can only be achieved when patients have obvious space-occupying lesions and clinical symptoms, missing the early stage of canceration, which is the optimal treatment time. Therefore it is the current research focus to find serum tumor marker for early diagnosis of malignant tumor. With the rapid development of the serum tumor marker,thymidine kinase-l( TK1 ),a new cell proliferation marker, has been confirmed to be of great significance in early detection, therapeutic effect evaluation and prognosis of tumors. 此前我们已报道超氧化物歧化酶(superoxidedismutase-1,SOD-1)能明显减轻ET-1引起大鼠心律失常,本实验应用电子自旋共振(ESR)方法经大鼠检测冠状动脉口注射外源性ET-1后对大鼠心肌氧自由基含量的影响,以明确ET-1能否引起大鼠心肌自由基含量的增高。
1、材料与方法1.1实验材料1.1.1动物Sprague-Dawley大鼠30只,雄性,体重(20020)g,由大学医学部实验动物中心提供。
1.1.2仪器E-200电子自旋共振仪(X波段,德国Brucker 公司)。
1.1.3试剂ET-1(日本大阪肽研究所公司);超氧化物歧化酶(SOD-1,南京建成生物公司)。
1.2实验方法1.2.1分组30只雄性SD大鼠,随机分为3组,每组10只。
ET-1组:冠状动脉口注射1.5molL-1的ET-1900pmolkg-1;SOD-1+ET-1组:冠状动脉口注射SOD-10.6mgkg-14min后,再注射1.5molL-1的ET-1900pmolkg-1(各组大鼠ET-1的注射剂量均为0.6mLkg-1);对照组:冠状动脉口注射生理盐水0.6mLkg-1。
1.2.2冠状动脉口注射[6]大鼠用20%尿酯6mLkg-1腹腔注射麻醉,仰卧位固定,气管插管,分离右侧颈总动脉。

目前,国内外对二硫代氨基甲酸盐缓蚀剂的研究较为活跃。
国内外许多学者从不同的角度和方向研究了这种化合物的合成、性能、机理等方面。
但是,目前研究大多数集中在缓蚀效果评价以及相关化合物的合成上,对于机理探究和优化改进研究不足,因此本文对DTC-1缓蚀剂进行了合成和性能研究,并对其缓蚀机理进行了初步探究,为相关领域研究提供一定的借鉴和参考。
本文的内容主要包括三个部分:实验部分、性能测试和机理研究部分。
在实验部分,我们将介绍DTC-1的合成方法和反应机理;在性能测试部分,我们将通过不同的腐蚀性能测试和缓蚀性能测试来评价DTC-1的表现;在机理研究部分,我们将探究DTC-1缓蚀的机理,并寻找适合其应用的改进策略。
最后,我们将综合分析实验结果和机理研究结果,对DTC-1的应用前景和未来研究方向进行展望。
II. 实验部分A. 合成技术路线DTC-1的合成主要是通过二乙酸二甲酯与硫代氨基甲酸双分子进行反应得到。

1 TET1的基本特征TET家族是重新调控已经分化细胞的一种重要功能蛋白,在DNA去甲基过程和干细胞编程、抑制肿瘤细胞增殖等方面起关键作用[4]。
人类TET蛋白家族共有3名成员:TET1,TET2和TET3,其中TET1发现最早,由Tahiliani等[5]在研究1例存在t(10,11)(q22,q23)异位的白血病患者中作为融合蛋白发现的。

而频率合成器则为混频器提供本振信号源。
直接数字频率合成技术(DDS)是根据相位概念直接合成所需的频率,DDS具有频率转换快的特点,而且频率在一定的范围内可以适当地扩展,能够输出宽带本振信号,同时在一定程度上抑制杂散。
该设计是音频处理接收机模拟前端模块的本振源。
2 直接数字频率合成(DDS)的工作原理和结构直接数字频率合成技术可提供快速的信号建立时间,纯净的信号频谱[1]。
DDS 技术是通过DAC把数字量的信号转换为模拟量的信号,从而合成所需的频率。
DDS的结构如图1所示。
图1 DDS的框图结构在DDS工作时,在参考时钟的触发之下,相位累加器与频率控制字(Frequency Control Words,FCW)K在每一个时钟周期内相加一次,相加之后的相位值在每个周期内以二进制码的形式去寻址正弦查询表ROM,将相应的相位信息转化到相应的数字幅度值,ROM输出的数字量经过DAC之后转化为模拟量,如图2后半部分所示的阶梯波,再经过一个低通滤波器滤掉其他分量得到一个平滑的正弦波本振信号[2]。

They are fully screened and fully submersible when mated with the proper bushing or plug and they meet the requirements of CENELEC HD 629.1 S2. The bolted tee connectors can be mounted vertically, horizontally or any angle in between with proper cable support. It is in good condition when accepted by the carrier for shipment. Upon receipt of a bolted tee connector, inspect it thoroughly for damage and loss of parts incurred during shipment. If damage or loss is discovered, file a claim with the carrier immediately.Handling and storageBe careful during handling and storage of the bolted tee connector to minimize the possibility of damage. Train the cable to the desired finished position and cutthe cable. (See Figure 3.)ote:N If the cable is visibly bent, straighten it beforeinstalling the tee. A bent cable will result in aloose interface and reduce the dielectric withstand of the interface, especially for large cable sizes. Recommended method for straightening the cable: Heat up the cable to 90 °C by using a heat tape. Keep the tape on for approximately 120 minutes and then remove the heat tape. Keep the cable straight while cooling. It may be helpful to tape a splint to the cable while it is cooling down.Crimp LugChamferMechanical LugChamferFigure 3 . Remove the cable jacket for a distance of 315 mm (forcrimp lug only) or 320 mm (for mechanical lug only) from the end of the cable.3. Pull back drain wires. Use vinyl tape to secure wirestemporarily.4. Remove 265 mm (for crimp lug only) or 270 mm (formechanical lug only) of insulation screen. Chamfer the sharp end at a 45° angle.5. Remove “L ” mm of cable insulation.ote:N Determine the value of “L ” from Table 1.6. Apply three layers of vinyl tape of the end of theconductor.7. Remove vinyl tape from the screen wires. Wrap twohalf-lapped layers of sealing tape over cable sheath, then pull down the screen wires. Apply 3-4 layers of semi-conductive tape over theadapter edge and cable screen wires. Then apply 2 layers of sealing tape over the semi-conductive tape and caulk screen wires. Wire brush conductor (aluminum only). Wipe offaluminum dust. Do not get dust on cable adapter.2. Immediately insert the conductor into the crimpingbarrel as far as it will go. Ensure that the flat of the lug spade is parallel to the face of the bushing.3. Crimp the lug starting at the mark on the barrel of thespade end of the lug as shown in Figure 7. Rotate the crimping tool 90 degrees for each successive crimp.4. Remove any excess inhibitor that may have come outof the crimp lug. File off any sharp projections. With the appropriate Allen hex key (per Table 2), screwin bolt No. 1 until it breaks off smoothly. Proceed with bolt No. 2 in a similar way. If the lug has more than two (2) bolts, continue with No. 3 (See Figure 8 for tightening sequence).5. Clean the outer surface of the cable adapter and theinterior of the cable entrance of the tee connector with a lint-free cloth. Apply a thin layer of lubricant to both surfaces.2. Push the tee connector body over the cable adapter asfar as it will go. Ensure that the hole in the top of the crimp connector is visible through the interface end ofthe tee.Figure 9 .
