临床查体均呈阴性。
本院超声检查:左肾中部一低回声光团,大小约34 mm×26 m m,向外凸,后方回声衰减,考虑肾癌。
CT平扫和增强扫描:左肾中部肾实质内见等及稍高密度实性肿块,向肾轮廓外及肾盂突出,最大层面约27mm×39mm,病变内未见坏死及囊变,未见明确脂肪密度,平扫CT值约27~39 HU(图1),增强扫描皮质期肿块强化程度明显低于周围相对正常的肾皮质,其内可见斑片状轻度强化,CT值约45~55 HU(图2);髓质期肿块持续强化,呈斑片状明显强化灶,CT 值约60~75 HU(图3)。
影像诊断:左肾少血供肿块,考虑恶性肿瘤。
行左肾部分切除术,术中见肿瘤位于左肾中上极,偏腹侧,外生性,距肿瘤边界0.5 cm 处完整切除肿瘤。
大体观:肿瘤包膜完整,切面呈鱼肉样。
镜下观:肿瘤由梭形细胞构成,大部分细胞异型性不明显,少许细胞伴轻-中度异型(图4),细胞密集与疏松区交替分布丰富;肿物与肾组织界限尚清,其内可见较多血管,局部黏液样变。
免疫组化:肿瘤细胞 BCL-2 (+),CD34 (+),SMA 局部(+),EMA (±),HMB45(-),S-100(-),CD117(-),DOG-1(-),Desmin(-),Calponin(-),CD68(-),AE1/AE3(-),Ki-67 指数约 1%~3%。

大肠杆菌DNA复制相关蛋白PriC的初步研究孔敏敏;李艳华;董宇辉;王文雅;袁其朋【期刊名称】《中医学报》【年(卷),期】2011(26)6【摘要】目的:对大肠杆菌DNA复制相关蛋白PriC进行研究。
方法:构建重组表达载体pET21a-PriC及pET21a-PriC1-101,并进一步诱导表达纯化。
利用Pulldown手段将pET21a-PriC-notag与pET28at-plus-PriB分别转化到大肠杆菌BL21(DE3)中,诱导表达及共纯化后检测PriC与PriB的相互作用情况。
结果:PriC经Ni柱亲和层析纯化获得的蛋白稳定性较差,易沉淀。
根据二级结构和蛋白稳定性预测及限制性蛋白酶水解结果,构建了PriC的截短突变蛋白PriC1-101,经Ni亲和层析和superdex75纯化,获得了稳定性较好的蛋白,经Western blot鉴定为目的蛋白。
PriC1-101的圆二色谱分析发现其具有较多的α-helix和无规卷曲,这为更好的了解其结构奠定了重要的基础。
通过Pulldown手段发现PriC与PriB 存在相互作用,并且PriC的稳定性有所提高。
结论:PriC蛋白的C-端氨基酸序列是PriC蛋白与PriB蛋白相互作用的主要区域,并对PriC蛋白的稳定性有较大影响。
