[摘要]目的:对巴戟天炮制前后3种蒽醌类化学成分的含量进行比较研究。方法:采用HPLC 同时测定3种蒽醌类成分1,8-二羟基-3-羟甲基蒽醌,1,8-二羟基-3-甲基-5-羟甲基蒽醌,1-甲基蒽醌的含量,色谱柱:Agilent-C 18(4.6mm ˑ150mm ,5

μm );流动相以甲醇-水梯度洗脱(60ʒ40→85ʒ15,25min );流速1.00mL ·min -1

;柱温30ħ;检测波长278nm ;进样量10μL 。

测定巴戟天生品和炮制品中三种蒽醌的含量。结果:3种蒽醌的线性范围分别为1,

8-二羟基-3-羟甲基蒽0.0199 0.1992μg ,

1,8-二羟基-3-甲基-5-羟甲基蒽醌0.0331 0.3312μg ,1-甲基蒽醌0.0197 0.1968μg ,3种蒽醌的平均回收率均在98.50% 100.31%。巴戟天炮制前后蒽醌类成分有所变化。结论:所建立的分析方法可以同时测定巴戟天不同炮制品3种蒽醌类化合物的含量,为解析巴戟天炮制原理提供一定的科学依据。

[Abstract ]Objective :To compare the content of anthraquinones in the different processed products from Morinda officinalis How.Method :1-methylanthraquinone ,1,8-dihydroxy-3-methyl-5-hydroxymethyl-anthraqui-none ,1,8-dihydroxy-3-hydroxymethyl-anthraquinone were determined simultaneously by HPLC.The separation was performed on a Agilent C 18column at 30ħwith MeOH-H 20(60:40→85:15,25min )as a mobile phase.The de-tection wavelength was set at 278nm and the flow rate was 1.00mL ·min -1.Result :Three components were linear within the range of 0.0199-0.1992μg ,0.0331-0.3312μg ,0.0197-0.1968μg ,and the average recoveries were 98.50%-100.31%respectively.Conclusion :The method can be used for evaluating the quality in different processed pieces of M.officinalis .
