1)常规制备DNA盐析法1.用淋巴细胞分离液从抗凝血中分离出白细胞。
2.每3ml细胞悬液加10ml红细胞裂解液溶解红细胞两次,再加2ml白细胞裂解液溶解白细胞。
3.加50μl 10%SDS和70μl蛋白酶K消化蛋白质,37℃过夜或55℃3h。
4.加0.25体积饱和醋酸钠溶液,剧烈摇动15s。
5.2000r/min离心15min,收集上清。
6.加入2.5倍预冷无水乙醇沉淀DNA,-20℃过夜。
7.吸取DNA团块用70%乙醇洗3次,每次10 000r/min,10min。
再溶于适量TE缓冲液中,4℃保存。
酚抽提法1.采集EDTA抗凝血0.5ml,加双蒸水2.5ml,溶解红细胞。
2.离心10 000r/min,5min,弃上清。
3.加0.5ml生理盐水,混匀,离心10 000r/min,5min,弃上清。
4.加500μl TES,混匀,再加20%SDS 25μl,蛋白酶K 4μl置55℃水浴4h或37℃水浴过夜。
5.加等体积饱和酚,混匀,离心10 000r/min,5min,吸取上层水相,再加等体积饱和酚抽提一次。
6.加等体积氯仿:异戊醇再抽提一次。
7.吸取上层液,加1/10体积醋酸钠,2.5倍体积无水乙醇,混匀,-20℃过夜,沉淀DNA。
8.吸出DNA团块,用预冷70%乙醇洗1次,弃尽液体,真空抽干DNA,加适量TE溶解。
4℃保存。
2)限制性内切酶消化在无菌EP管中依次加入双蒸去离子水7μl,10×缓冲液2μl,限制性内切酶1μl,DNA 10μl。
反应体系含1×缓冲液、DNA 5μg、酶10U。
稍离心以混匀,37℃水浴1~1.5h。
酶解结束后向反应管中加1/10体积的终止液终止反应,冷却至4℃,准备电泳。
3)琼脂糖凝胶电泳1.配制1%琼脂糖凝胶板:取0.4g琼脂糖加40ml TBE,微波炉熔化,加少量双蒸水补充蒸发的水分,将其冷却至60℃,倒入凝胶槽内,充分凝固后拔出样品梳。
2.从制胶槽中卸下凝胶板,放入电泳槽,加入1×TBE,使其液面略高于琼脂糖板。

According to the differences of the key loci of the three alleles, the PCR-RFLP method was used to amplify the DNA fragments and perform specific enzyme digestion. The genotype could be preliminarily determined based on the enzyme diges-tion results, and then the enzyme digestion results are verified by sequencing method.
