1.简介

验证是药品生产管理规范(GMP)的一个重要组成部分;也正因如此,所以它同时也是产品或工艺的质量保证计划的一个不可或缺的要素。质量保

证的基本原则以生产出符合设计用途的产品为目的,它的主要内容为:

要事先设计质量、安全和效力,并使之在产品中实现。

不能仅仅只考察和检验成品的质量。

生产工艺的每一关键步骤都必须经过验证,非关键步骤也必须得到控制,从而实现在最大程度上生产出符合全部质量和既定标准的产品的目的。

工艺和系统的验证是实现上述目标的基础之一。通过对生产的设计和验证,可使生产出的产品持续符合标准,并建立一定的置信区间。

与验证有关的文件如下:

—标准操作规程(SOP)

—合格标准

—验证主计划(VMP)

—确认方案和确认报告

—验证方案和验证报告。执行验证工作时,需要的资源有:

时间:通常,验证工作必须严格按照时间表的安排开展。

资金:验证工作的执行常常需要专业人员和昂贵的技术支持。

人员:验证工作的开展要求有不同领域的专家的合作(如一支由来自多个领域的专家组成的验证团队,根据待验证的产品和工艺的要求,包括质量保证、工程、生产和其它领域)。

这些指导方针能给药品生产设施的检查员及药品生产商提供关于验证要求的指导。正文部分的内容主要是关于验证和确认,附录中另有其它方面的内容(如清洁、计算机和计算机化系统、设备、公用工程和公用系统、分析方法等)。

2.范围

2.1这里给出的指导方针主要涵盖了关于验证的所有方面,可供GMP检查员

和生产商作为基本指南使用,但不可将本文视作验证要求的专门说明文章;

而应当仅将本文视为一般的指南。在递交关于活性药物成分(API)和药物成品的生产和控制资料时,本文列出的基本原则也可作为参考。特定工艺和产品的验证,如灭菌生产,通常需要考虑更多的方面以及详细的方法,本文的内容不足以满足它们的需要。

2.2验证的类型虽然繁多,但却有许多共同的因素能对之形成影响;因此,

3.术语

以下是指导方针中使用的术语的定义;需注意,它们在其他文献中出现时,可能有不同的含义。

校准

在规定的条件下,用来建立测量用(例如测量重量、温度和pH值)、记录用、控制用仪器或系统、及物料测量仪器测得的值与已知参照标准的相应值之间的关系的粗操作;在该项操作中,还应建立测量结果的合格限。

计算机验证

用以保证计算机化系统可提供正确的分析、控制和记录数据及保证数据处理符合既定标准的文件证明。

试运行

根据用户需求说明书所述及设计人员或研发人员的能力,用来保证设备或仪器的装配、调整和测试符合要求,及保证设备或仪器满足其设计和研发人员的要求的活动。试运行应先于确认和验证执行。

同步验证

针对销售产品的常规生产过程进行的验证。

清洁验证

在考虑了批量、剂量、毒理学和设备大小因素的前提下,用以表明清洁方法去除残余物的能力可满足既定合格标准的文件证明。

设计确认(DQ)

用以表明厂房建筑、支持系统、公用工程、设备和工艺的设计符合GMP 要求的证明文件。

工程设计规范(GEP)

对于项目的整个运作周期,建立了工程方法和技术标准来设计适当且又低本高效的解决方法。

安装确认(IQ)

performance quali?cation (PQ)

Documented veri?cation that the equipment or system operates consistently and gives reproducibility within de? ned speci?cations and parameters for prol onged periods. (In the context of systems, the term ―process validation‖ may also be used.)

为证明设备或系统可长期实现设计性能的文件化确认工作,如正确运转并表现出良好的再现性。(就系统而言,性能确认即―工艺验证‖。)

process validation

Documented evidence which provides a high degree of assurance that a speci?c process will consistently result in a product that meets its predeter-mined speci?cations and quality characteristics.

prospective validation

前验证

Validation carried out during the development stage on the basis of a risk analysis of the production process, which is broken down into individual steps; these are then evaluated on the basis of past experience to determine whether they may lead to critical situations.

在生产工艺投入使用前的研发阶段开展的验证工作;它通常以风险分析为基础,并被分解为单个步骤来验证。根据过去的经验对前验证进行评估,判断该工艺是否会导致关键情况的出现。

quali?cation

确认

Action of proving and documenting that any premises, systems and equip-ment are properly installed, and/or work correctly and lead to the expected results. Quali?cation is often a part (the initial stage) of validation, but the individual quali?cation steps alone do not constitute process validation.

为证明所有厂房建筑、系统和设备均正确安装且/或正确运行,并能实现预期结果的文件化证明活动。确认常常是验证的(起始阶段的)一个部分,但单独的确认步骤不是验证。

retrospective validation

回顾性验证

Involves the evaluation of past experience of production on the condition that composition, procedures, and equipment remain unchanged.

revalidation

再验证

Repeated validation of an approved process (or a part thereof) to ensure continued compliance with established requirements.

standard operating procedure (SOP)

An authorized written procedure giving instructions for performing opera-tions not necessarily speci?c to a given product or material but of a more general nature (e.g. equipment operation, maintenance and cleaning; vali-dation; cleaning of

premises and environmental control; sampling and in-spection). Certain SOPs may be used to supplement product-speci? c master batch production documentation.

Action of proving and documenting that any process, procedure or method actually and consistently leads to the expected results.

A document describing the activities to be performed in a validation, in-cluding the acceptance criteria for the approval of a manufacturing process — or a part thereof — for routine use.

validation report (VR)

验证报告(VR)

A document in which the records, results and evaluation of a completed validation programme are assembled and summarized. It may also contain proposals for the improvement of processes and/or equipment.

validation master plan (VMP)

验证主计划(VMP)

The VMP is a high-level document that establishes an umbrella validation plan for the entire project and summariz es the manufacturer’s overall phi-losophy and approach, to be used for establishing performance adequacy. It provides information on the manufacturer’s validation work programme and de?nes details of and timescales for the validation work to be performed, including a statement of the responsibilities of those implementing the plan.

to monitoring, to determine compliance with the GMP principles.

worst case

A condition or set of conditions encompassing the upper and lower processing

limits for operating parameters and circumstances, within SOPs, which pose the greatest chance of product or process failure when compared to ideal con-ditions.

Such conditions do not necessarily include product or process failure.

4. Relationship between validation and quali? cation

验证和确认之间的联系

Validation and quali?cation are essentially components of the same concept. The term quali?cation is normally used for equipment, utilities and systems, and validation fo r processes. In this sense, quali?cation is part of validation.

验证和确认系同一概念的基本组成部分,即是说它们是同义词。―确认‖这个词往往用在设备、公用工程和系统以及工艺验证中;在这层意义上,确认是验证的一部分。

5. Validation

5.1 Approaches to validation

验证方法

5.1.1 There are two basic approaches to validation —one based on evi-dence

obtained through testing (prospective and concurrent validation), and one based on the analysis of accumulated (historical) data (retrospective validation).

Whenever possible, prospective validation is preferred. Retro-spective validation is no longer encouraged and is, in any case, not appli-cable to the manufacturing of sterile products.

5.1.2 Both prospective and concurrent validation, may include:

13extensive product testing, which may involve extensive sample testing (with the estimation of con?dence limits for individual results) and the demonstration of intra- and inter-batch homogeneity;

15challenge/worst case tests, which determine the robustness of the process; and

5.2 Scope of validation

5.2.1 There should be an appropriate and suf?cient system including

organizational structure and documentation infras tructure, suf?cient personnel and ?nancial resources to perform validation tasks in a timely manner.

Management and persons responsible for quality assurance should be involved.

为了及时执行验证工作,应当建立一个适当且能满足需要的系统,包括组织机构和文件管理的下层组织结构、充足的人力和财政资源,同时还应指定质量保证的管理和责任人。

5.2.2 Personnel with appropriate quali?cations and experience should be

responsible for performing validation. They should represent different departments depending on the validation work to be performed.

执行验证工作的人员应当具备相应的资格和经验,并对验证负责。根据待执行的验证工作的需要,决定由哪些部门的人员参与验证工作的执行。

5.2.3 There should be proper preparation and planning before validation is

performed. There should be a speci?c programme for validation activities.

执行验证之前,应有充分的准备和计划。应针对具体的验证活动,制订专门的计划。

5.2.4 Validation should be performed in a structured way according to the

documented procedures and protocols.

应根据成文的程序和方案,以设计好的方式执行验证工作。

5.2.5 Validation should be performed:

验证应:

— for new premises, equipment, utilities and systems, and processes and procedures;

在新的厂房建筑、设备、公用工程和系统、工艺和方法上执行;

— at periodic intervals; and

定期执行;

— when major changes have been made.

在发生了重大变更后执行。

(Periodic revalidation or periodic requali?cation may be substituted, where appropriate, with periodic evaluation of data and information to establish whether

requali?cation or revalidation is required.)

5.2.6 Validation should be performed in accordance with written protocols. A written report on the outcome of the validation should be produced.

5.2.7 Validation should be done over a period of time, e.g. at least three consecutive batches (full production scale) should be validated, to demon-strate consistency. Worst case situations should be considered.

验证项目的执行时间的设定应能证明验证对象的持续性。如验证应当至少在三个连续批上开展(生产规模)。验证时,要考虑最差状况。

5.2.8 There should be a clear distinction between in-process controls and validation. In-process tests are performed during the manufacture of each batch according to speci?cations and methods devised during the develop-ment phase. Their objective is to monitor the process continuously.

过程控制和验证之间应有明显的区别。为监测工艺的连续性,应根据针对研发阶段设计的技术标准和方法,开展每批产品生产的过程测试。

5.2.9 When a new manufacturing formula or method is adopted, steps should be taken to demonstrate its suitability for routine pro cessing. The de?ned process, using the materials and equipment speci? ed, should be shown to result in the consistent yield of a product of the required quality.

采用新的生产配方或方法时,应证明它们对常规工艺的适应性。验证时,应使用规定的物料和设备,运行既定的工艺,持续得到的产品质量应符合要求。

5.2.10 Manufacturers should identify what validation work is needed to prove that critical aspects of their operations are appropriately controlled. Signi?cant changes to the facilities or the equipment, and processes that may affect the quality of the product should be validated. A risk assessment approach should be used to determine the scope and extent of validation required.

生产商需明白为了证明他们的操作的关键问题得到了合理的控制,应采取哪些验证工作。比如,对产品质量可能产生影响的厂房设施、设备和工艺的重大变更是需要验证的。为确定需要开展的验证的范围和验证的程度,可使用风险评估的方法。

6. Quali?cation

6.1 Quali?cation shoul d be completed before process validation is per-formed. The process of quali?cation should be a logical, systematic process and should start from the design phase of the premises, equipment, utilities and equipment.

确认应当先于工艺验证完成。确认工作应当是一个有条理的、系统的过程,并应从厂房建筑、设备、公用工程和公用设备的设计阶段着手开始。

6.2 Depending on the function and operation of the equipment, utility or system, only installation quali?cation (IQ) and operational quali?cation (OQ) may be

required, as the correct operation of the equipment, utility or system could be considered to be a suf?cient indicator of its performance (refer to Section 11 for IQ, OQ and performance quali? cation (PQ)). (The equipment, utility and system should then be maintained, monitored and calibrated according to a regular schedule.)

6.3 Major equipment and critical utilities and systems, however, require IQ, OQ

and PQ.

7. Calibration and veri?cation

校准和核查

7.1 Calibration and veri?cation of equipment, instruments and other devices, as

applicable, used in production and quality control, should be performed at regular intervals.

用于生产和质量控制的设备、仪器和其它器械应定期进行校准和核查。

7.2 Personnel who carry out calibration and preventive maintenance should have

appropriate quali?cations and training.

执行校准和预防维护的人员应当具备相应的资格且得到过适当的培训。

7.3 A calibration programme should be available and should provide infor-mation

such as calibration standards and limits, responsible persons, calibra-tion intervals, records and actions to be taken when problems are identi? ed.

应制订好校准计划,准备好资料,如校准标准和限度、负责人、校准间隔、记录和发生问题后所采取的措施。

7.4 There should be traceability to standards (e.g. national, regional or

international standards) used in the calibration.

应当有可追溯的校准标准(如国家标准、地区标准或国际标准等)。

7.5 Calibrated equipment, instruments and other devices should be la-belled,

coded or otherwise identi?ed to indicate the status of calibration and the date on which recalibration is due.

7.6 When the equipment, instruments and other devices have not been used for a

certain period of time, their function and calibration status should be veri?ed and shown to be satisfactory before use.

17Validation master plan

The validation master plan (VMP) should re?ect the key elements of the validation programme. It should be concise and clear and contain at least the

following:

9. Quali?cation and validation pr otocols

9.1 There should be quali?cation and validation protocols describing the

quali?cation and validation study to be performed.

应有说明待执行的确认和验证的确认和验证方案。

9.2 As a minimum the protocols should include the following signi? cant

background information:

9.3 There should be a description of the way in which the results will be analysed.

9.4 The protocol should be approved prior to use. Any changes to a pro-tocol

should be approved prior to implementation of the change.

在投入使用之前,这些方案应得到批准。如变更了方案,那么,在将这些变更投入使用之前,这些方案的变更也需要获得批准。

10. Quali?cation and validation reports

确认和验证报告

10.1 There should be written reports on the quali?cation and validation performed.

应有已执行的确认和验证活动的书面报告。

10.2 Reports should re?ect the protocols followed and include at least the title

and objective of the study; reference to the protocol; details of material, equipment, programmes and cycles used; procedures and test methods.

报告的内容应表明方案已执行,且至少要说明研究的标题和对象、方案的参考资料、使用到的物料、设备、计划和时间的明细、程序及实验方法。

10.3 The results should be evaluated, analysed and compared against the

pre-determined acceptance criteria. The results should meet the acceptance criteria;

deviations and out-of-limit results should be investigated. If these deviations are accepted, this should be justi?ed. Where necessary further studies should be performed.

要对结果进行评估和分析,并用既定的合格标准与之比较。结果应当符合合格标准,如发现有偏差和超出接受限度的结果,应对之展开调查。如果这些偏差尚在接受范围内,需给出相应证明。需要时,要展开进一步的研究。

10.4 The departments responsible for the quali?catio n and validation work should

approve the completed report.

完成的报告应得到确认和验证工作的负责部门的批准。

10.5 The conclusion of the report should state whether or not the outcome of the

quali?cation and/or validation was considered successful.

报告中给出的结论应阐述了该确认和/或验证工作的结果是否成功。

10.6 The quality assurance department should approve the report after the ?nal

review. The criteria for approval should be in accordance with the company’s quality assurance system.

10.7 Any deviations found during the validation process should be acted upon

and documented as such. Corrective actions may be required.

34Quali?cation stages

11.1 There are four stages of quali?cation:

11.2 All SOPs for operation, maintenance and calibration should be prepared

Design quali? cation

11.4 Design quali?cation should provide documented evidence that the design

spec i?cations were met.

Installation quali? cation

11.5 Installation quali?cation should provide documented evidence that the

installation was complete and satisfactory.

11.6 The purchase speci?c ations, drawings, manuals, spare parts lists and vendor

details should be veri?ed during installation quali? cation.

采购标准、图纸、使用手册、零部件清单和供应商的详细资料都应在安装确认阶段得到确认和核实。

11.7 Control and measuring devices should be calibrated.

应校准控制和测量仪器。

Operational quali? cati on

运行确认

11.8 Operational quali?cation should provide documented evidence that utilities,

systems or equipment and all its components operate in accor-dance with operational speci? cations.

运行确认的活动应可提供公用工程、系统或设备及其所有部件的运行均符合运行技术标准的文件化证明。

11.9 Tests should be designed to demonstrate satisfactory operation over the normal operating range as well as at the limits of its operating condi-tions (including worst case conditions).

设计此阶段进行的试验时,应确保在正常运行范围及运行条件的上下限值(包括最差状况)的情况下运行时,确认对象都能实现满意的运行结果。

11.10 Operation controls, alarms, switches, displays and other operational components should be tested.

11.11 Measurements made in accordance with a statistical approach should be fully described.

应充分说明测量活动与统计方法一致。

Performance quali?cation

性能确认

11.12 Performance quali?cation should provide documented evidence that utilities, systems or equipment and all its components can consistently per-form in accordance with the speci?cations under routine use.

性能确认活动应能给出文件证明,说明在常规使用时,公用工程、系统或设备及其所有部件的性能均符合标准,且具备持续性。

11.13 Test results should be collected over a suitable period of time to prove consistency.

为证明结果的持续性,应以适当的时间间隔采集试验结果。

Requali?cation

再确认

11.14 Requali?cation should be done in accordance with a de? ned schedule. The frequency of requali?cat ion may be determined on the basis of factors such as the analysis of results relating to calibration, veri?cation and maintenance.

应按照事先制订的时间表执行再确认工作。可根据校准相关结果的分析、核实确认和维护等因素确定执行再确认的频率。

11.15 There should be periodic requali?cation, as well as requali? ca tion after changes (such as changes to utilities, systems, equipment; maintenance work; and movement). (See also point 5.2.5 above and section 12 below.)

11.16 Requali?cation sh ould be considered as part of the change control procedure.

11.17 Processes and procedures should be revalidated to ensure that they remain

capable of achieving the intended results.

11.18 There should be periodic revalidation, as well as revalidation after changes.

药品微生物检验替代方法验证指导原则 本指导原则是为所采用的试验方法能否替代药典规定的方法用于药品微生物的检验提供指导。 随着微生物学的迅速发展,制药领域不断引入了一些新的微生物检验技术,大体可分为三类:(1)基于微生物生长信息的检验技术,如生物发光技术、电化学技术、比浊法等;(2)直接测定被测介质中活微生物的检验技术,如固相细胞技术法、流式细胞计数法等;(3)基于微生物细胞所含有特定组成成分的分析技术,如脂肪酸测定技术、核酸扩增技术、基因指纹分析技术等。这些方法与传统检查方法比较,或简便快速,或具有实时或近实时监控的潜力,使生产早期采取纠正措施及监控和指导优良生产成为可能,同时新技术的使用也促进了生产成本降低及检验水平的提高。 在控制药品微生物质量中,微生物实验室出于各种原因如成本、生产量、快速简便及提高药品质量等需要而采用非药典规定的检验方法(即替代方法)时,应进行替代方法的验证,确认其应用效果优于或等同于药典的方法。 微生物检验的类型及验证参数 药品微生物检验方法主要分两种类型:定性试验和定量试验。定性试验就是测定样品中是否存在活的微生物,如无菌检查及控制菌检査。定量试验就是测定样品中存在的微生物数量,如菌落计数试验。 由于生物试验的特殊性,如微生物检验方法中的抽样误差、稀释误差、操作误差、培养误差和计数误差都会对检验结果造成影响,因此,药品质量标准分析方法验证指导原则(附录XIX A)不完全适宜于微生物替代方法的验证。药品微生物检验替代方法的验证参数见表1。 表1 不同微生物检验类型验证参数 注: 尽管替代方法的验证参数与药品质量标准分析方法验证参数有相似之处,但是其具体的内容是依据微生物检验特点而设立的。替代方法验证的实验结果需进行统计分析,当替代方法属于定性检验时,一般采用非参数的统计技术;当替代方法属于定量检验时,需要采用参数统计技术。 进行微生物替代方法的验证时,若替代方法只是针对药典方法中的某一环节进行技术修改,此时,需要验证的对象仅是该项替代技术而不是整个检验方法。如无菌试验若改为使用含培养基的过滤器,然后通过适宜的技术确认活的微生物存在,那么,验证时仅需验证所用的微生物回收系统而不是整个无菌试验方法。 替代方法验证的一般要求 在开展替代方法对样品检验的适用性验证前,有必要对替代方法有一个全面的了解。首先,所选用的替代方法应具备必要的方法适用性证据,表明在不含样品的情况下,替代方法

药物注射剂研发技术指导意见 (征求意见稿) 第一章总论 第一节为落实《关于深化审评审批制度改革鼓励药品医疗器械创新的意见》,严格药物注射剂审评审批有关要求,我中心制定《药物注射剂研发的技术指导意见》(以下简称意见)。 第二节本意见适用于药品上市许可持有人药物研发,也适用于药品审评中心技术审评。本意见适用于化学药品注射剂(包括多组分生化药注射剂)和生物制品注射剂,不适用于中药注射剂。 第二章药物注射剂总体考虑 第三节剂型选择基本原则。注射剂可分为溶液型、注射用无菌粉末、注射用浓溶液以及乳剂、混悬剂、注射用油溶液、注射用微球、胶束、纳米粒、脂质体等特殊类型载药系统的注射剂。药物注射剂剂型的研发,以满足临床治疗需求为前提,并综合药物的理化性质、生物学特性等因素,做出科学、合理的选择。

方法学验证指导原则

一、准确度 准确度系指采用该方法测定的结果与真实值或参考值接近的程度,一般用回收率(%)表示。准确度应在规定的范围内测定。 1.化学药含量测定方法的准确度 原料药采用对照品进行测定,或用本法所得结果与已知准确度的另一个方法测定的结果进行比较。制剂可在处方量空白辅料中,加入已知量被测物对照品进行测定。如不能得到制剂辅料的全部组分,可向待测制剂中加人已知量的被测物对照品进行测定,或用所建立方法的测定结果与已知准确度的另一种方法测定结果进行比较。准确度也可由所测定的精密度、线性和专属性推算出来。 2.化学药杂质定量测定的准确度 可向原料药或制剂处方量空白辅料中加人已知量杂质进行测定。如不能得到杂质或降解产物对照品,可用所建立方法测定的结果与另一成熟的方法进行比较,如药典标准方法或经过验证的方法。在不能测得杂质或降解产物的校正因子或不能测得对主成分的相对校正因子的情况下,可用不加校正因子的主成分自身对照法计算杂质含量。应明确表明单个杂质和杂质总量相当于主成分的重量比(%) 或面积比(% )。 3.中药化学成分测定方法的准确度 可用对照品进行加样回收率测定,即向已知被测成分含量的供试品中再精密加人一定量的被测成分对照品,依法测定。用实测值与供试品中含有量之差,除以加入对照品量计算回收率。在加样回收试验中须注意对照品的加人量与供试品中被测成分含有量之和必须在标准曲线线性范围之内;加入对照品的量要适当,过小则引起较大的相对误差,过大则干扰成分相对减少,真实性差。 回收率:%= (C - A ) /S X 100% 式中:A为供试品所含被测成分量;B 为加入对照品量; C 为实测值。 4.校正因子的准确度 对色谱方法而言,绝对(或定量)校正因子是指单位面积的色谱峰代表的待测物质的量。待测定物质与所选定的参照物质的绝对校正因子之比,即为相对校正因子。相对校正因子计算法常应用于化学药有关物质的测定、中药材及其复方制剂中多指标成分的测定。校正因子的表示方法很多,本指导原则中的校正因

1. 范围
准确测定生物基质(如全血、血清、血浆、尿)中的药物浓度,对于药物和 制剂研发非常重要。这些数据可被用于支持药品的安全性和有效性,或根据毒动 学、药动学和生物等效性试验的结果做出关键性决定。因此,必须完整地验证和 记录应用的生物分析方法,以获得可靠的结果。
本指导原则提供生物分析方法验证的要求,也涉及非临床或临床试验样品实 际分析的基本要求,以及何时可以使用部分验证或交叉验证,来替代完整验证。
生物样品定量分析方法验证和试验样品分析应符合本指导原则的技术要求。 应该在相应的生物样品分析中遵守 GLP 原则或 GCP 原则。
2. 生物分析方法验证
2.1 分析方法的完整验证
分析方法验证的主要目的是,证明特定方法对于测定在某种生物基质中分析 物浓度的可靠性。此外,方法验证应采用与试验样品相同的抗凝剂。一般应对每 个物种和每种基质进行完整验证。当难于获得相同的基质时,可以采用适当基质 替代,但要说明理由。
一个生物分析方法的主要特征包括:选择性、定量下限、响应函数和校正范 围(标准曲线性能)、准确度、精密度、基质效应、分析物在生物基质以及溶液 中储存和处理全过程中的稳定性。
有时可能需要测定多个分析物。这可能涉及两种不同的药物,也可能涉及一 个母体药物及其代谢物,或一个药物的对映体或异构体。在这些情况下,验证和 分析的原则适用于所有涉及的分析物。
对照标准物质 在方法验证中,含有分析物对照标准物质的溶液将被加入到空白生物基质 中。此外,色谱方法通常使用适当的内标。 应该从可追溯的来源获得对照标准物质。应该科学论证对照标准物质的适用 性。分析证书应该确认对照标准物质的纯度,并提供储存条件、失效日期和批号。 对于内标,只要能证明其适用性即可,例如显示该物质本身或其相关的任何杂质 不产生干扰。 当在生物分析方法中使用质谱检测时,推荐尽可能使用稳定同位素标记的内 标。它们必须具有足够高的同位素纯度,并且不发生同位素交换反应,以避免结 果的偏差。

1概述 无菌药品是指法定药品标准中列有无菌检查项目的制剂和原料药,一般包括注射剂、无菌原料药及滴眼剂等。从严格意义上讲,无菌药品应完全不含有任何活的微生物,但由于目前检验手段的局限性,绝对无菌的概念不能适用于对整批产品的无菌性评价,因此目前所使用的“无菌”概念,是概率意义上的“无菌”。一批药品的无菌特性只能通过该批药品中活微生物存在的概率低至某个可接受的水平,即无菌保证水平(Sterility Assurance Level, SAL)来表征。而这种概率意义上的无菌保证取决于合理且经过验证的灭菌工艺过程、良好的无菌保证体系以及生产过程中严格的GMP管理。 无菌药品通常的灭菌方式可分为:1)湿热灭菌;2)干热灭菌;3)辐射灭菌;4)气体灭菌;5)除菌过滤。按工艺的不同分为最终灭菌工艺(sterilizing process)和无菌生产工艺(aseptic processing)。其中最终灭菌工艺系指将完成最终密封的产品进行适当灭菌的工艺,由此生产的无菌制剂称为最终灭菌无菌药品,湿热灭菌和辐射灭菌均属于此范畴。无菌生产工艺系指在无菌环境条件下,通过无菌操作来生产无菌药品的方法,除菌过滤和无菌生产均属于无菌生产工艺。部分或全部工序采用无菌生产工艺的药品称为非最终灭菌无菌药品。基于无菌药品灭菌/除菌生产工艺的现状,本指导原则主要对在注射剂与无菌原料药的生产中比较常用的湿热灭菌与无菌生产工艺进行讨论。本指导原则中的湿热灭菌工艺验证主要包括灭菌条件的筛选和研究,湿热灭菌的物理确认,生物指示剂确认等内容;无菌生产工艺验证主要包括无菌分装、除菌过滤、培养基模拟灌装、过滤系统的验证等验证内容。 最终灭菌工艺和无菌生产工艺实现产品无菌的方法有本质上的差异,从而决定了由这两类工艺生产的产品应该达到的最低无菌保证水平的巨大差异。最终灭菌无菌产品的无菌保证水平为残存微生物污染概率≤10-6,非最终灭菌无菌产品的无菌保证水平至少应达到95%置信限下的污染概率<0.1%。由此可见,非最终灭菌无菌产品存在微生物污染的概率远远高于最终灭菌无菌产品,为尽量减少非最终灭菌无菌产品污染微生物的概率,鼓励企业在生产中采用隔离舱等先进技术设备。 基于质量源于设计的药品研发与质量控制的理念,为保证无菌药品的无菌保证水平符合要求,研发者在产品的研发过程中应根据药品的特性选择合适的灭

一、范围
由于蒸汽-湿热灭菌本身具备无残留,不污染环境,不破坏产品表面,并容易控制和重现等 优点,被广泛应用于最终灭菌药品(注射剂)的除菌过程中。
本指南为有关人员提供最终灭菌药品(注射剂)的蒸汽灭菌柜的验证指南,以及蒸汽灭菌工 艺及验证的一些操作方法的指南。
本指南依据《药品生产质量管理规范》(1998 年修订)的相关准则,但本指南叙述的通用原 则和方法不是法定的。本指南的着重于最终灭菌药品(注射剂)的蒸汽-湿热灭菌工艺的验 证,但有些通用原则和方法对于冻干机的湿热灭菌、某些设备的在线蒸汽灭菌等可能也具备 参考价值。
二、目的
蒸汽-湿热灭菌验证的目的,就是通过一系列验证试验提供足够的数据和文件依据,从而找 到最有效最合理的灭菌参数,并把已经验证过的饱和蒸汽灭菌设备和灭菌工艺参数应用到药 品生产的除菌过程中去,以证明用于药品生产过程中的每一台饱和蒸汽灭菌设备都能起到灭 菌的效果,并且对不同灭菌物品的灭菌过程和灭菌效果具有可靠性和重现性,即验证结果必 须证明生产中所采用的灭菌过程对经过灭菌的物品能够保证残存微生物污染的概率或可能 性低于百万分之一。
蒸汽-湿热灭菌周期的设计和开发与蒸汽灭菌柜的性能以及被灭菌产品的适用性有关。蒸汽湿热灭菌介质包含以下几种:饱和蒸汽,空气-蒸汽混合气体,过热水等等。其中:饱和蒸 汽的加热速度最快,但是对于大型的软包装产品,过热水浸泡灭菌的方法效率更高,然而在 过热水灭菌法中,热量的转移很大程度上依赖于容器中介质的强制运动。
饱和蒸汽是与液体状态的水保持平衡时的水蒸汽,因此饱和蒸汽只能存在于水汽的分界线 上,即温度与压力之间的关系是固定的。灭菌效果是通过蒸汽,蒸汽-空气混合物,过热水 等介质与灭菌物品的热传递或产生冷凝水的水合作用来实现的。
蒸汽-空气混合物与受压的水或蒸汽相比,单位体积所包含的热容量较低,但是,蒸汽-空气 混合物作为灭菌戒指具有能够适当调整蒸汽-空气比例达到不同结果的优点。
选择一种适合的蒸汽灭菌方式,能在满足产品本身性能的情况下取得满意的灭菌效果,但是 任何一种灭菌方法,都必须在实际应用前予以验证。
