RESEARCH APPLICATIONS
These controls can be used to estimate the amount of nonspecific binding of mouse monoclonal antibodies to human cell-surface antigens in peripheral blood and tissue, in particular binding caused by Fc receptors.1
The PerCP-Cy?5.5*, PE-Cy?7, APC-Cy7?, and APC-H7 controls can also be used to estimate the amount of binding mediated by the known interaction of the cyanine dye with CD64 protein.1
DESCRIPTION
Specificity The antibodies react specifically with keyhole limpet hemocyanin (KLH), an antigen not expressed on human cells or human cell lines.
Clones
γ1 (IgG 1), clone X40, and γ2a (IgG 2a ), clone X39, are each derived from hybridization of mouse Sp2/0-Ag 14 myeloma cells with spleen cells from BALB/c mice immunized with KLH.
Composition
γ1, clone X40, is composed of mouse IgG 1 heavy chains and kappa light chains.γ2a , clone X39, is composed of mouse IgG 2a heavy chains and kappa light chains.Product configuration

PROCEDURE Consult the monoclonal antibody data sheet for each test antibody to determine the
amount of control (in μg) to use. Test and control antibody concentrations should be
matched. FITC, PE, PerCP, and PerCP-Cy5.5 reagents are all used at 20 μL/106 human
peripheral blood mononuclear cells (PBMCs). Concentrations of monoclonal controls
vary. PE-Cy7, APC, APC-Cy7, APC-H7, AmCyan, and V450 reagents are formulated
at 5 μL per test.
CAUTION Some nonspecific staining might be observed when using Mouse IgG1 PE-Cy7
at titers of 0.25 μg per test (100 μL whole blood).
Do not use Mouse IgG1 PE or Mouse IgG2a PE control for intracellular staining or for
platelet assays. Use Mouse IgG1 PE Platelet Control (Catalog Number 340760) or
Mouse IgG2a PE Intracellular Control (Catalog Number 340756).
Some APC-Cy7 (and to a lesser extent PE-Cy7 and APC-H7) conjugates show changes
in their emission spectra with prolonged exposure to formaldehyde.

HANDLING AND STORAGE Store vials at 2° to 8°C. Conjugated forms should not be frozen and should be protected from exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.
WARNING Reagents contain sodium azide. Sodium azide is harmful if swallowed. Keep away from
food, drink, and animal feedingstuff. Wear suitable protective clothing. If swallowed,
seek medical advice immediately and show this container or label. Contact with acids
liberates very toxic gas. Azide compounds should be flushed with large volumes of
water during disposal to avoid deposits in lead or copper plumbing where explosive
conditions can develop.
CHARACTERIZATION To ensure consistently high-quality reagents, each lot of monoclonal antibody is tested
for conformance with characteristics of a standard reagent. Representative flow
cytometric data is included in this data sheet.
